Summary 1. The splitting ofl-Leucyl-β-naphthylamide-HCl by cytolyzed white blood cells is measured. The enzyme content of normal polymorphonuclear cells, cells of chronic myeloic leukemia and macrophages from the peritoneal cavity of guinea pigs is 3 times higher than that of cells from chronic lymphatic leukemia. 2. In the assay system employed the reaction runs lineary for 30 min. The pH optimum of the enzyme is 7.1, the substrate optimum 5×10−4 M, the Michaelis constant 5×10−5 M. There is no activation by MnCl2 (5×10−5 M, pH 7.1), the reaction is even inhibited in the alkaline range and at higher concentration. 3. The enzyme differs from leucine aminopeptidase.

Zusammenfassung 1. Die Spaltung vonl-Leucyl-β-naphthylamid-HCl durch Cytolysate von weißen Blutzellen wird biochemisch gemessen. Normale Granulocyten, Zellen von chronischen Myelosen und Meerschweinchen-Makrophagen weisen einen etwa dreifach höheren Enzymgehalt als Lymphocyten von chronischen Lymphadenosen auf. 2. Das pH-Optimum beträgt 7,1, Mangan-Ionen (5×10−5 Mol/l, pH 7,1) bewirken keine Aktivierung, im alkalischen Bereich und in höherer Konzentration wird eine Hemmung beobachtet. Unter den gegebenen Versuchsbedingungen folgt die LNA-Spaltung einer Reaktion nullter Ordnung. Das Substrat-Optimum liegt bei 5×10−4 Mol/l. Die Michaelis-Konstante beträgt 5×10−5 Mol/l. 3. Das untersuchte Enzym weist dieselben Eigenschaften auf, wie sie für LNA spaltende Enzyme anderer Gewebe beschrieben wurden, unterscheidet sich jedoch von der Leucinaminopeptidase.
