Summary By cell-electrophoresis the presence of neuraminic acids in the membranes of Hela cells seems most likely; chemically they are present. The addition of neuraminidase to the culture medium reduces the electrophoretic motility of the cells. This reduction is inversely proportional to the logarithm of the concentration of the neuraminidase. In spite of this reduction the viability of the cell cultures is not impaired. A daily change in the medium of neuraminidase-treated Hela cells by a neuraminidase-free medium leads to the restoration of the original mobility of the cells after three days. A change every third day requires six to seven days for this. In like manner, the electrophoretic mobility of malignant liver ascites cells is greatly reduced by an intraperitoneal (i.p.) injection of neuraminidase without decreasing the viability and transplantability of the cells. The i.p. injected neuraminidase may be detected in the ascites fluid 24 hours after the injection. The rate of successful transplantation in rats is not altered by the simultaneous i.p. injection of neuraminidase. Furthermore, in Ehrlich ascites-mice no increase in the longevity of the animals may be expected by the i.p. injection of neuraminidase. Hence, neuraminidase affects neither the in vivo nor the in vitro viability of the tumor cells studied.

Zusammenfassung Die Anwesenheit von Neuraminsäuren wurde in Membranen von Hela-Zellen zellektrophoretisch sehr wahrscheinlich gemacht und chemisch bewiesen. Die Zugabe von Neuraminidase zu dem Kulturmedium vermindert die elektrophoretische Beweglichkeit der Zellen. Die Verminderung ist dem Logarithmus der Neuraminidasekonzentration invers proportional. Trotz dieser Verminderung ist die Vitalität der Zellkulturen nicht beeinträchtigt. Der tägliche Mediumwechsel bei Neuraminidase-behandelten Hela-Zellen mit einem Neuraminidasefreien Kulturmedium führt nach 3 Tagen zur Wiederherstellung der Ausgangsbeweglichkeit der Zellen. Ein dreitägiger Wechsel benötigt hierfür 6–7 Tage. Bei malignen Leberasciteszellen wird die elektrophoretische Beweglichkeit durch eine i.p. Injektion von Nueraminidase ebenfalls stark vermindert, ohne daß die Vitalität und Transplantabilität der Zellen herabgesetzt ist. Die i.p. injizierte Neuraminidase kann in der Ascitesflüssigkeit noch 24 Std nach der Injektion nachgewiesen werden. Die Angehrate bei Ratten wird durch die gleichzeitige i.p. Injektion von Neuraminidase nicht verändert. Auch bei Ehrlich-Ascitesmäusen kann durch i.p. Injektion von Neuraminidase keine Lebensverlängerung der Tiere erwartet werden. Demnach beeinflußt Neuraminidase weder in vivo noch in vitro die Vitalität der untersuchten Tumorzellen.
