The rate of enzymatic hydrolysis of o-carboxy-phenyl-phosphate (I) has been compared with that of other substrates commonly used for phosphatase determinations. An acid phosphatase from wheat germ hydrolysed I faster than β-glycero-phosphate, phenyl-phosphate and phenolphtalein-diphosphate. Two alkaline phosphatases decomposed I somewhat more slowly than β-glycero-phosphate, but still over 10 times faster than phenolphtalein-diphosphate. Even in cases where another substrate is hydrolysed faster, the use of I is recommended because of the high accuracy and simplicity of the spectro-photometric determination described in an earlier communication.
