    Notizen: Nanoemulsions have recently become increasingly important as potential vehicles for the controlled delivery of cosmetics and for the optimized dispersion of active ingredients in particular skin layers. The preparation of conventional nanoemulsions requires mainly high-pressure homogenization, which is unproductive and requires high energy due to its lower efficiency, limiting their practical applications. In order to solve these problems novel nanoemulsions were studied using a model system of pseudo-ternary water/emulsifier/paraffin oil. Nanoemulsions were prepared by stirring a mixture of the tocopherol-containing block co-polymer emulsifier PPG-20 Tocophereth-50, paraffin oil, and distilled water at the Θ-point using weight fractions of the dispersed phase (φ) of 0.31 to 0.82 and an emulsifier content of 1.0 to 9 wt.%. The emulsifying property of PPG-20 Tocophereth-50 in nanoemulsions was compared with that of the conventional emulsifiers Tocophereth-43, a mixture of polysorbate 60 and sesquioleate (3/1), and phospholipids. Also the emulsifying property of PPG-20 Tocophereth-50 in the more hydrophilic oils caprylic/capric triglyceride and octyldodecanol was compared with that in paraffin oil. The stability and morphology of the resulting nanoemulsions were studied by visual inspection, optical microscopy, particle size analysis, and cryo-scanning electron microscopy. In the nanoemulsion systems containing caprylic/capric triglyceride and octyldodecanol, respectively, as an oil phase PPG-20 Tocophereth-50 showed emulsification properties similar to those in paraffin oil. The conventional emulsifiers Tocophereth-43, a mixture of polysorbate 60 and sesquioleate (3/1), and phospholipids did not give nanoemulsions with high-speed stirring. The block co-polymer nonionic emulsifier PPG-20 Tocophereth-50 was found to produce stable nanoemulsions of mean droplet diameters ranging from 204 to 499 nm. The emulsification method of high-speed stirring at the Θ-point using PPG-20 Tocophereth-50 was found to be very effective for the preparation of stable nanoemulsions useful for applications in skincare cosmetics, cosmeceuticals, and drugs.

    Notizen: 1 To investigate anti-inflammatory activity of organic germanium, we measured the effect of germanium-concentrated yeast on arachidonic acid release, prostaglandin E2 (PGE2) production, histamine release, and intracellular H2O2 or hydroperoxide generation in RBL 2H3 cells, and carrageenan-induced paw oedema in rats. 2 Germanium-concentrated yeast dose-dependently inhibited carrageenan-induced paw oedema, suggesting that germanium-concentrated yeast has anti-inflammatory activity in acute inflammation. 3 Germanium-concentrated yeast significantly inhibited melittin-induced arachidonic acid release and PGE2 production in RBL 2H3 cells. 4 Germanium-concentrated yeast did not affect melittin-induced histamine release and silica-induced intracellular H2O2 or hydroperoxide generation in RBL 2H3 cells. 5 These results suggest that anti-inflammatory activity of germanium-concentrated yeast appears partly to be related to the inhibition of arachidonic acid release and PGE2 production in RBL 2H3 cells.

    Notizen: A 3 × 3 factorial experiment was conducted to determine proper levels of dietary protein, lipid and dextrin for juvenile flounder. Nine experimental diets were formulated to contain three protein levels (410, 460 and 510 g kg−1) and three lipid levels (60, 130 and 190 g kg−1) with corresponding dextrin levels (250, 150 and 50 g kg−1). Triplicate groups of fish (8.9 ± 0.4 g) were hand-fed the diets to apparent satiation for 7 weeks in flow-through system. Specific growth rate was the highest in fish fed the 510 g kg−1 protein diet with 60 g kg−1 lipid, and was not significantly different from that of fish fed 460 g kg−1 protein diet with 60 g kg−1 lipid. Feed efficiency ratio tended to increase as dietary protein level increased. The feed efficiency ratio of fish fed the 510 g kg−1 protein diets with 60–190 g kg−1 lipid levels was not significantly different from that of fish fed 460 g kg−1 protein diet with 60 g kg−1 lipid. Daily feed intake tended to decrease with increasing dietary lipid level at each protein level. Daily protein intake increased with increasing dietary protein level at 60 g kg−1 lipid level. Hepatosomatic index and visceralsomatic index increased with increasing dietary lipid level at each protein level. The lipid contents of liver, viscera and whole body, and concentrations of plasma total cholesterol and triglyceride increased with increasing dietary lipid levels; however, no significant difference was observed in the contents of dorsal muscle lipid. The results of this study suggest that the diet containing 460–510 g kg−1 protein with low lipid level (60 g kg−1) is optimal for growth and efficient feed utilization of juvenile flounder.

    Notizen: Expression of calretinin in retina has been ascribed to multiple biological and functional aspects in the visual system. In this study, we examined the distribution patterns of calretinin immunoreactivity in gerbil and rat retina. In the gerbil, calretinin immunoreactivity was present in bipolar and amacrine cells of the inner nuclear layer and in neurones of the ganglion cell layer. In the rat, amacrine and ganglion cells showed calretinin immunoreactivity, but bipolar cells did not contain calretinin immunoreactivity. In both species, calretinin immunoreactivity was absent in cones, cone bipolars, and horizontal cells. In conclusion, gerbil as well as rat has a rod-dominant retina. The differences in calretinin expression between rat and gerbil require further investigations under various functional and developmental conditions.

    Notizen: Protein tyrosine kinases (PTK) are proteins that may play importants roles in survival, proliferation and progression to metastasis in several tumors, including melanoma. There is little information on the expression pattern of the phosphorylated (i.e. activated) forms of such kinases in melanoma and their impact on overall survival (OS). We sought to determine the expression, by immunohistochemistry, of phosphorylated MAP kinase (pMAPK), Akt (pAkt) and C-kit (pCkit) in primary and metastatic melanoma, and their correlation with OS. Fifteen patients with Stage III and Stage IV (metastatic melanoma) disease were randomly selected from the UT MD Anderson database. Expression was rated by two investigators on intensity (negative, weak, moderate, and strong) and proportion of reactive tumor cells (0 = 〈5%, 1 = 5–25%, 2 = 〉25–75%, 3 = 〉75%). Eleven of the 15 tumors (primary and/or metatasis) exhibited pMAPK expression (73%). Corresponding percentages were 100% for pAKT and 〈50%(7/15) for pCkit. OS ranged from less than one year to 9 years. However, there was no correlation between OS and expression of any of these markers.
