    Notes: A high-torque rheometer was used to facilitate the polycondensation of 4-[5-amino-6-hydroxybenzoxazol-2-yl]benzoic acid (ABA) with trimesic acid and 1,3,5,7-tetrakis(4-carboxylatophenyl)adamantane to yield two- and three-dimensional benzobisoxazole polymers, respectively. Although the resultant polymer dopes exhibited improved homogeneity compared to polymer dopes previously prepared in glassware, improved polymer solution viscosities were not achieved. Fibers spun from the two- and three-dimensional polymers did not show a significant increase in compressive strength compared to fibers of the linear or one-dimensional benzobisoxazole polymer derived from the homopolymerization of ABA. Morphological studies of the polymer fibers and films by wide-angle X-ray scattering and scanning electron microscopy strongly indicated more lateral disorder and a more isotropic character for the three-dimensional structures compared to the one-dimensional structures. © 1997 John Wiley & Sons, Inc.     Notes: Inosine-5′-monophosphate dehydrogenase (IMPDH) from the protozoan parasite Tritrichomonas foetus has been expressed in E. coli and crystallized. Crystals were grown to 0.1 mm in each dimension in 18 to 72 h using ammonium sulfate and low-molecular-weight polyethylene glycols. The crystals belong to the cubic space group P432 with unit cell edge = 157.25 Å. The enzyme is a homotetramer with each monomer having a molecular weight of 55,534 Da. There is one monomer per asymmetric unit, based on a volume/mass ratio of 2.7 Å3/Da and self-rotation analysis. The crystals are adequately stable to allow a complete data set to be collected from a single crystal. Complete native data sets have been collected to 2.3 Å resolution at 4°C using synchrotron radiation. High-quality complete data extending to 3.0 Å resolution have been collected from crystals of four putative derivatives, and the data appear to be isomorphous with that of the native crystals in each case. Efforts to solve the derivatives for use in MIR phasing are underway. © 1995 Wiley-Liss, Inc.

    Notes: Polycondensation in polyphosphoric acid of 2,5-diamino-1,4-benzene dithiol dihydrochloride with bicyclo[2.2.2]octane-1,4-dicarboxylic acid, as well as the corresponding dimethyl ester or diacid chloride, led to rigid-rod benzobisthiazole polymers. Colorless and soluble polymers with intrinsic viscosities as high as 30.6 dL/g (methanesulfonic acid, 30°C) were obtained. The ultraviolet-visible spectrum of a polymer film cast from methanesulfonic acid under reduced pressure displayed no absorptions in the visible range (400-900 °m). The polymer was thermooxidatively stable up to 420°C in air as determined by thermogravimetric analysis. Fibers spun from a lyotropic polyphosphoric acid solution exhibited a tensile strength of 300-450 Ksi, a modulus of 26 Msi, and a compressive strength of 53 Ksi. Wide-angle X-ray scattering patterns of polymer fibers indicated a 3-dimensional crystal structure rather than a nematic liquid crystal structure. © 1994 John Wiley & Sons, Inc.

    Notes: A stereospecific high-performance liquid chromatographic (HPLC) method was developed for the quantitation of the enantiomers of venlafaxine, an antidepressant, in dog, rat, and human plasma. The procedure involves derivatization of venlafaxine with the chiral reagent, (+)-S-naproxen chloride, and a postderivatization procedure. The method was linear in the range of 50 to 5,000 ng of each enantiomer per ml of plasma. No interference by endogenous substances or known metabolites of venlafaxine occurred. Studies to characterize the disposition of the enantiomers of venlafaxine were conducted in dog, rat, and human, following oral administration of venlafaxine. The Cmax, area under the curve (AUC) and (S)/(R) concentration ratios of the (R)- and (S)-enantiomers were compared. In rats, the mean plasma ratio of (S)-venlafaxine to that of (R)-venlafaxine over 0.5 to 6.0 h varied from 2.97 to 8.50 with a mean value of 5.51 ± 2.45. The Cmax, AUC0 - ∞, and t1/2 values of the (R)- and (S)-enantiomers in dogs were not significantly different from one another (P〉0.1). The mean ratios [(S)/(R)] of enantiomers of venlafaxine in human over a 2 to 6 h interval ranged from 1.33 to 1.35 with an overall ratio of 1.34 ± 0.26 (n = 12). These ratios of the enantiomers [(S)/(R)] were not statistically different from unity (P〉0.1) indicating that the disposition of venlafaxine enantiomers in humans is not stereoselective and is more similar to that in dogs than that in rats.

    Notes: In the present paper, the structure and conductivity for the complex of sulfonated poly(phenylene oxide) lithium (SPPOLi) and poly(ethylene oxide) (PEG) were studied. Glass transition temperature change determined by differential scanning calorimeter analysis desmonstrated that the two components had some compatibility. X-ray diffraction showed that PEG could decrease the regularity of SPPOLi to some extent. The compatibility and PEG's effect on the regularity may be due to the interaction between the lithium ions of SPPOLi and the oxygen atoms of PEG. Under polarization by electric field, the bands between lithium ions and sulfonation groups relaxed. Meanwhile, the complexation of oxygen atoms could enhance the dissociation of the polymeric lithium salts. Then lithium ions were transported in the process of alternate complexing and decomplexing. The action between lithium ions and oxygen atoms could explain the improvement on the conductivity of SPPOLi.
