    Notizen: Abstract Over the last few years we have developed mass spectrometry-based approaches for selective identification of a variety of posttranslational modifications, and for sequencing the modified peptides. These methods do not involve radiolabeling or derivatization. Instead, modification-specific fragment ions are produced by collision-induced dissociation (CID) during analysis of peptides by ESMS. The formation and detection of these marker ions on-the-fly during the LC-ESMS analysis of a protein digest is a powerful technique for identifying posttranslationally modified peptides. Using the marker ion strategy in an orthogonal fashion, a precursor ion scan can detect peptides which give rise to a diagnostic fragment ion, even in an unfractionated protein digest. Once the modified peptide has been located, the appropriate precursor ion can be sequenced by tandem MS. The utility and interplay of this approach to mapping PTM is illustrated with examples that involve protein glycosylation and phosphorylation.

    Notizen: A method is described for rapid purification of synthetic oligodeoxynucleotides to remove sodium counter ions prior to electrospray ionization mass spectrometry. The oligomers, following purification by gel electrophoresis, are precipitated from ammonium acetate to replace sodium ions by ammonium ions, and dissolved in water. Negative-ion electrospray spectra are presented for oligomers with up to 48 residues in which the most intense peaks correspond to the [M—nH]n- ion. The spectrum of 77-mer (Mr24039) shows the predominant peak as due to retention of only a single sodium ion. Changes in the spectra are reported for different instrument orifice voltages and different solution pH. The method should permit rapid, accurate analysis of most typical, synthetic oligodeoxynucleotides.

    Notizen: A generally applicable strategy has been developed for the primary sequence determination of peptides and proteins containing γ-carboxyglutamic acid. The intact peptide or protein is either dissolved in, or allowed to come into vapor phase contact with, 0.05 M DCl and then heated in vacuo at 110°C for several hours. Under these conditions γ-carboxyglutamic acid quantitatively decarboxylates and incorporates two atoms of deuterium per molecule, resulting in the formation of γ-dideuteroglutamyl residues. Following enzymatic or acidic degradation of the protein the peptide mixture generated is converted (without further isolation of individual peptides) to the N-trifluoroacetylated, O-trimethylsilylated polyamino alcohols and subsequently analyzed by gas chromatography mass spectrometry. Peptide fragments in which γ-carboxyglutamic acid was present show sequence ions in their mass spectra corresponding to those of glutamic acid, but shifted upwards by 2 amu. This approach has been used to identify the positions of Gla in a tryptic peptide isolated from blood coagulation factor IX, and is currently being employed in the sequence determination of the Gla-containing bone protein, osteocalcin.

    Notizen: The derivatization chemistry for conversion of mixtures of oligopeptides to the corresponding N-α, (ω)-trifluoroethyl-O-trimethylsilyl polyamino alcohols, the derivatives of choice for the sequencing of polypeptides by gas chromatographic mass spectrometry, has been optimized. The improvements have minimized undesirable side reactions and resulted in a five- to tenfold increase in sensitivity over previous methods employing either lithium aluminum deuteride or hexadeuterodiborane as the reducing agent. For derivatives of certain very polar peptides increases in yield have exceeded 100-fold. The procedure has been evaluated with mixtures of synthetic oligopeptides and used in the course of the determination of the amino acid sequence of the membrane protein bacteriorhodopsin, as well as other proteins.

    Notizen: Fast atom bombardment (FAB) ionization and two coupled analyzers (BE-EB) have been combined with neutral gas collision (C) to enhance structual information in the mass spectra of oligosaccharides. (B and E are abbreviations for magnetic and electric sectors respectively.) FAB ionization and the first analysers (BE) have provided parent ions free from biological and liquid matrix contaminants. Structural detail of these products were observed after collision and daughter ion analysis in a second coupled analyser (EB). Starting from complex mixtures, this instrumental approach, BE-C-EB, has provided specific oligomeric sequence information which was not observed in the normal FAB mass spectra. Collision spectra obtained from isomeric linear and branched oligosaccharides show unique fragments that can be directly related to structure.

    Notizen: A comparison of continuous-flow fast atom bombardment (FAB) mass spectrometry and nebulization-assisted electrospray for analysis of proteins and glycoproteins by high-performance liquid chromatography (HPLC)/mass spectrometry is presented. The evaluation was made using enzymatic digests of recombinant soluble CD4 glycoprotein and recombinant hepatitis B surface antigen and a mixture of HPLC retention standard peptides. These samples were analyzed by reversed-phase HPLC on a microbore (1 × 100 mm C18) gradient HPLC system with 10% or 20% of the eluent containing ∼20-100 pmol of the sample directed into the continuous-flow FAB mass spectrometric or electrospray mass spectrometric source, respectively. The techniques were evaluated on the criteria of chromatographic integrity, ease of data analysis, protein sequence coverage, discrimination effects, ability to detect glycopeptides, simplicity of operation, and sensitivity. Both techniques produce useful peptide molecular weight data from comparable amounts of sample injected. However, the nebulization-assisted electrospray system is capable of yielding higher peptide mapping coverages with the least sample consumed in toto due in part to the wider mass ranges resulting from the multicharging effect and to the ability to detect glycopeptides. Under the experimental conditions employed here no fragmentation was observed in the electrospray mass spectra. In contrast, significant, sequence informative fragmentation was occasionally observed for peptides in the continuous-flow FAB mass spectral data.

    Notizen: This report describes the use of direct chemical ionization mass spectrometry with ammonia as the reagent gas (NH3-DCI) for structure analysis of underivatized, permethylated and permethylated and reduced glycosphingolipids. In contrast to ionization by electron impact the NH3-DCI mass spectra exhibit intense molecular and carbohydrate sequence-related ions using microgram amounts of sample. Underivatized glycosphingolipids with up to two sugar residues yield abundant protonated and ammonia-cationized molecular ions and structurally significant fragments. Permethylation in conjunction with NH3-DCI can be used to obtain molecular weight as well as oligosaccharide sequence and branching information on neutral, acidic and complex-type glycosphingolipids with up to five sugar residues. Reduction of the permethylated derivatives gives rise to several new, structurally significant fragments in the corresponding NH3-DCI mass spectra which enable fatty acid and base compositions to be determined. Isotopically labeled reagent gases have been used to confirm the assignment of fragment structures and to demonstrate that the ions observed are unique to the NH3-DCI mass spectra.
