    Notes: A method is described for the quantification of tulobuterol in human plasma, based upon selective extraction and high resolution capillary gas chromatography-mass spectrometry. The mass spectrometric determination is based on selected ion monitoring of two fragment ions derived from the N,O-heptafluorobutyryl derivatives of tulobuterol and its internal standard, deschlorotulobuterol. The plasma work-up procedure consists of extraction of 1.0 ml alkalinised plasma with dichloromethane, back-extraction into an acidified aqueous phase, followed by a final extraction with dichloromethane after realkalinisation. The method gave interference-free and linear results. The between-assay variability is 4.7% CV at the 3.0 μg l-1 plasma concentration level. The assay permitted quantitative measurements down to 170 ng tulobuterol per litre of plasma and hence provided sufficient sensitivity for quantification of plasma levels after therapeutic doses.

    Notes: The capillary gas chromatographic (CGC) analysis of the der-matological drug trans-acitretin (NeotigasonR) and its cis metabolite is described. Separation of the methyl ester derivatives can be achieved on a 90% biscyanopropylsiloxane phase. The importance of using cold on-column injection and short, thin film capillary columns is discussed. For patients treated with the prodrug of acitretin, etretinate (TigasonR), i.e. the ethyl ester of Neotigason, three compounds have to be separated. Selectivity tuning is required for successful CGC separation. An alternative can be found in the selectivity of ion monitoring mass spectroscopy. Analysis of plasma samples involves liquid-liquid extraction, a derivatization step, and HPLC purification.

    Notes: Fluorouracil in human plasma at a concentration as low as 2 ng ml-1 can be assayed with the use of gas-liquid chromatography and a multiple ion detector. Fluorouracil is isolated from plasma by anion exchange extraction. The extract is butylated and the derivative obtained extracted into a mixture of cyclohexane + dichloromethane (95:5). An aliquot is submitted to combined gas-liquid chromatography mass spectrometry. The use of chlorouracil or oxygen-18 labelled flurouracil as internal standard is discussed. Linearity is proven from 100 μg to 5 ng of fluorouracil per ml plasma. The coefficients of variation are 7.2 and 8.4% for within-run and between-run precision, respectively, at the 50 ng ml-1 level. The biological applicability of our procedure is demonstrated by analysing plasma samples obtained at different time intervals from three patients given 1 g of fluorouracil once intravenously and once orally.

    Notes: The formation of a new derivative of aldosterone with heptafluorobutyric anhydride for gas chromatography/mass spectrometry (GC/MS) is presented. The highest and also the most prominent ion of this derivative is observed at m/z 734. The new derivative is, under the described conditions, reproducibly formed, is stable and has good gas chromatographic properties. These characteristics make the new derivative extremely suitable for selective, sensitive, precise and accurate analysis of aldosterone in serum by GC/MS in association with isotope dilution. This is proven by the agreement between the measurement results obtained by two laboratories for samples of three batches of lyophilized control serum. The sample pretreatment procedures used in each laboratory are described.

    Notes: A new method, based upon a selective extraction and gas chromatographic/mass spectrometric analysis, was developed to monitor the effect of combined haemoperfusion-haemodialysis treatment in a case of propericiazine poisoning. The method relies on the selected ion monitoring of the acetate derivatives of propericiazine and its internal standard fluphenazine, after their extraction from 1 ml of alkalinized plasma with n-hexane: isopropanol (8:2, v/v), back-extraction into an acidified water phase, realkalinization and extraction with n-hexane:isopropanol, derivatization with acetic anhydride and gas chromatography on a short (12 m) OV-101 fused silica capillary column. The described procedure is specific and provides between-assay variability of 4.8% CV at 5 μg I-1 plasma concentration. The method enables quantification down to 1 μg I-1 and hence demonstrates sufficient sensitivity to permit pharmacokinetic or drug monitoring studies.

    Notes: Identification of hydrazine monoamine oxidase inhibitor drugs such as isoniazid, iproniazid, nialamide, isocarboxazid and iproclozide is made by electron impact mass spectrometry using the direct insertion technique. The molecular ion itself, although of low relative abundance, is found in the mass spectra of all compounds studied. Relative intensities of the major fragments and data on metastable ions useful in the identification of these compounds are reported. With the aid of synthesized structurally related products, deuterium labelling of exchangeable hydrazidic and hydrazinic protons and by the use of hexadeuterated isopropylic analogues, detailed information about fragmentation patterns is obtained. Splitting processes are chiefly governed by the nature of the aromatic substituent at the hydrazidic end and the alkyl sidechain located at the second hydrazinic nitrogen. The major fragmentations occurring are loss of small neutral molecules, double rearrangement, cleavage of the N—N bond, amide bond rupture, β-cleavage to the hydrazinic nitrogen atom and McLafferty rearrangements.

    Notes: A new gas chromatographic/mass spectrometric method in combination with isotope dilution for the determination of thyroxine in serum is described. Special attention was paid to the methylation step of thyroxine, which was investigated using methanolic HCl, dimethylformamide/dimethylacetal and diazomethane, the latter giving the best results in terms of reproducible isotope ratios. For internal standardization, (13C6)-thyroxine was dissolved in fraction V human albumin solution (70 g l-1). The internal standard-in-albumin solution was mixed with known amounts of thyroxine standard, dissolved in 0.05 M Na2HPO4 buffer at pH 11.6, to give isotope ratios of 0.75, 1.00 and 1.25. The same internal standard solution was also used for isotope dilution of the unknown serum samples. The volume of serum was adapted to give a 1:1 isotope ratio. Sample pretreatment consisted of protein precipitation and a two-step liquid/liquid extraction procedure. After methylation of unlabelled and labelled thyroxine with diazomethane and perfluoroacylation with pentafluoropropionic anhydride and heptafluorobutyric anhydride, respectively, mass spectrometric monitoring was done at m/z 951/957 and 1001/1007. Quantitative determination of thyroxine in five serum samples in duplicate, during three consecutive days, showed a mean overall imprecision of 1.0% and a deviation of +0.4% from the target value as determined by a definitive method.
