Surface-enhanced resonance Raman (SERR) spectra of azurin, a copper-containing protein, were measured on a silver surface with a double grating. Significant differences in intensity and band wavenumber were obtained between SERR and solution resonance Raman spectra in the region 250-300 cm-1 and near 400 cm-1. Preliminary interpretation of these differences suggests a likely orientation of the azurin protein on the silver surface in which the N(His44)—N(His115)—S(Met119) plane is perpendicular to the metal surface and the imidazole group of His115 is in close proximity to the metal.
