    Notizen: Summary In situ hybridization of globin mRNA was performed in four human bone marrows using a 3H-cDNA derived from rat globin messenger. Erythroblasts were classified according to nuclear area as well as to morphological appearance after Giemsa staining. Grains over individual erythroblasts and reticulocytes were counted by incident light microphotometry. Reticulocytes from phenyl hydrazine-treated rats served as internal standards in order to allow a comparison of different experiments. The preliminary results from one normal bone marrow, one of stress erythropoiesis and two of aplastic anemia revealed the major content of globin mRNA in the stages of basophilic erythroblasts or proerythroblasts. In all cases there was a tendency of decreasing globin mRNA content towards orthochromatic erythroblasts, and even more so, towards reticulocytes. The results show some differences between normal and diseased erythropoiesis which are under study on a larger scale now in order to ascertain their significance.

    Notizen: Summary Auer-rod positive acute myelocytic leukemia (AML) blasts from a 33-year-old patient were cultured in diffusion chambers (DC) in order to test their differentiation potential. The cells were labeled with anti-human granulocyte antiserum known to be negative for eosinophils, and evaluated using the unlabeled peroxidase-antiperoxidase (PAP) method. Parallel to a decline in the number of leukemic blasts there was an increase of up to 86% in the number of granulocytic cells belonging to the eosinophilic series. Auer-rod bodies were found in the eosinophilic cells even after 20 days in culture. Staining with anti-granulocyte antiserum failed to demonstrate positive cells at any time during DC culture. Based on the negative reaction with the anti-granulocytic antibodies already at an early stage of development evidence is provided for the existence of a progenitor cell exclusively committed to the eosinophilic pathway.

    Notizen: Summary An analysis of erythroblast cell kinetics utilizing quantitative 14C-autoradiography has been performed in five cases of acute Plasmodium falciparum malaria prior to and, in four patients, 3 or 6 days after the onset of antimalarial therapy. Associated with no or only moderate anemia were changes of erythroblast morphology, a considerable shift in the frequency of red and white blood cell precursors in the bone marrow, and a reduced rate of erythroblast proliferation. There was a marked loss of polychromatic erythroblasts, which was smaller but still detectable during the therapeutic phase. The results provide some quantitative data on the extent of “parenchymal damage” of bone marrow and stress the impact of ineffective erythropoiesis and reduced rate of erythropoietic proliferation on the emergence of anemia in Plasmodium falciparum malaria.
