Zusammenfassung Es wird eine Mikromethode zur quantitativen Bestimmung von Diazepam (Valium) in den verschiedenen Schichten von Kälberaugenlinsen mit Hilfe der Hochdruck-Flüssigkeits-Chromatographie beschrieben. Die Methode ist gekennzeichnet durch einfache Probenvorbereitung (Extrelutsäule) und durch gute analytische Zuverlässigkeitskriterien. Die Praktikabilität des Analysenverfahrens wird durch die Verwendung eines automatisierten Gerätes und der Reversed-Phase-Technik (Füllmaterial mit C8-Gruppierungen) gewährleistet. Als Detektor dient ein Spektralphotometer. Die Präzision beträgt 3,0%, die Wiederauffindung 85,5%. An 80 Augenlinsen wurde festgestellt, daß der Gehalt an Diazepam pro Linsenepithel zwischen 0,3 und 2,9%, pro Linsenrinde zwischen 13,6 und 18,5% und pro Linsenkern zwischen 2,6 und 5,7% der eingesetzten Diazepammenge liegt.

Summary A simple and reliable micromethod is described for the quantitative determination of Diazepam (Valium) in different compartments of calf-lens using high-pressure liquid chromatography. The method is characterized by a simple sample preparation (Extrelut columns) and by a high analytical reliability. The practicability of this analytical method is guaranteed by the application of an automatically working highpressure liquid chromatograph and by the reversedphase technique on material containing C8-groupings. A spectrophotometer is employed as detector. Precision is 3.0%, the recovery rate 85.5%. An investigation of 80 lenses showed that the epithelium contained 0.3–2.9%, the cortex 13.6–18.5 and the core 2.6–5.7% of the amount of Diazepam applied.
