    Notizen: Abstract The neurophysiological interactions between the high pressure neurological syndrome (HPNS) and a newβ carboline, abecarnil, were studied in the nonhuman primatePapio anubis. Abecarnil is a partial agonist at the benzodiazepine site on the GABA/benzodiazepine receptor. Six animals were exposed on two occasions to pressures of 91 ATA in an environment of helium and oxygen. One exposure was pretreated with a total dose of abecarnil 1.0 mg/kg, the other with an equivalent volume of vehicle. Treatment with abecarnil prevented the severe signs of HPNS occurring between 51 and 91 ATA. Onset pressures of the various signs were unaffected. Some signs, e.g. myoclonus, became more frequent when abecarnil was used. A residual protective effect of abecarnil was present 4 weeks after the dose was given, active at pressures less than 71 ATA. Changes with pressure in the EEG were recorded primarily from the frontal cortex, but were also present in the parietal and occipital areas of the left cortex. Amplitude and frequency spectra were calculated and changes with pressure in the four conventional wavebands, plus two others, analysed. The most striking change was the prevention by abecarnil of the pressure-induced 100% increase in alpha wave amplitude in the frontal region. It is concluded that modulation of GABA transmission is important in controlling the expression of HPNS.

    Notizen: Three different sieve size fractions of ergotcontaining biodegradable microcapsules were examined both in vitro and in vivo. The sieve sizes and average particle diameter, (μm), were: less than 45-75 (X̄ = 30); 75-106 (X̄ = 79), 106-177 (X̄ = 130). These microcapsules contained ca. 9% drug and were produced from 50:50 poly(DL-lactide-co-glycolide). The objective was to determine the effect of particle size on in vivo and in vitro degradation rates. The microcapsules were injected into rat gastrocnemius muscle and excised and examined at various time points up to 70 days. Initially a minimal tissue response was noted which was characterized by a sharply localized acute inflammatory reaction. Following this, connective tissue and foreign body giant cells engulfed the microcapsules at 20-30 days. Only vestiges of the microcapsules were found surrounded by minimal connective tissue and foreign body giant cells after 60-70 days. The tissue reaction was a minimal, sharply localized foreign body giant cell and connective tissue process for all three size groups of microcapsules. The largest microcapsules (X̄ = 130 μm) exhibited a slightly greater tendency to undergo in vivo and in vitro degradation relative to the other groups. However, it can be concluded that over the microcapsule size ranges examined minimal differences in the degradation properties of the polymeric matrices and consequently those of the microcapsules were noted.

    Notizen: The biodegradation of the copolymer 50:50 poly(DL-lactide-CO-glycolide)-lypressin microcapsules was studied by light and electron microscopic methods and 14C release. Intramuscular injection sites of microcapsules in rats were studied by dissecting and conventional light microscopy as well as scanning (SEM) and transmission electron microscopy. A minimal localized acute myositis was seen initially at the injection sites. By Day 4, a few small foreign body giant cells were present participating in the minimal foreign body response. Later the inflammatory cells decreased and the individual microcapsules were walled off by immature fibrous connective tissue and large syncytial foreign body giant cells. By Day 35, definitive changes in some microcapsules, consisting of a granular and slightly eroded appearance of the internal matrix, were seen by SEM. By Day 42, the outer rims of the microcapsules were extensively eroded. At Day 56, the inflammatory and connective tissue reactions were almost completely resolved and biodegradation continued so that only remnant pieces of the microcapsules were present at Day 63. The morphologic picture correlated well with loss of 14C radioactivity, which could no longer be detected at the injection sites on Day 56. Phagocytosis did not seem to be an important factor in the biodegradation.

    Notizen: The polymerization of N-ethyl-2-vinylcarbazole and N-ethyl-3-vinylcarbazole by an anionic mechanism has been demonstrated. Polymerization reactions were monitored by ultraviolet/visible spectroscopy and λmax and ε values for the propagating carbanions determined. The 2-vinyl monomer exhibits all the features of a standard “living” polymer; the carbanion is stable at ambient temperatures and high molecular weight, M̄n ≃ 106, narrow distribution polymers and block copolymers with styrene have been prepared. The carbanion from the 3-vinyl monomer is much less stable and a clean polymerization can only be conducted at temperatures below -60°C. A comparison of the anionic polymerization characteristics of the N-, 2-, and 3-vinyl carbazole monomer series is presented.

    Notizen: Highly purified samples of N-ethyl-3-vinylcarbazole are readily polymerized in CH2Cl2 by conventional initiators of cationic polymerization, including boron trifluoride etherate and tropylium hexachloroantimonate. Reaction rates measured calorimetrically yield an estimate for the free-cation propagation rate coefficient (kp+ = 2 x 10+4 liter/mole-sec) at 0°C, which is some 20 times smaller than that for the closely related monomer N-vinylcarbazole. Distinguishing aspects of the cationic polymerization of N-ethyl-3-vinylcarbazole are the very high molecular weights obtained and the linear dependence of M̄n of the monomer/catalyst mole ratio, indicating that transfer and termination are comparatively unimportant. Polymerizations initiated by tropylium hexachloroantimonate exhibit a characteristic absorption band at 468 nm, tentatively assigned to the propagating cation, which undergoes rapid changes after all monomer has been consumed. The stability of the species responsible for the absorption band at 468 nm appears to be least in conditions where ion pairs are important.
