    Notizen: Pancreatic islets of the Syrian golden hamster were maintained in culture for extended periods of time. Toxicity of streptozotocin in these cultures was evaluated by measurement of insulin secretion. Exposure of islets to 1 or 2 mM streptozotocin immediately following isolation resulted in a permanent and dose-related inhibition of insulin secretion. This was accompanied by islet disruption as observed by phase-contrast microscopy. Culture of islets for 24 hours before streptozotocin exposure afforded protection from toxicity. For example, exposure of freshly isolated islets to 2 mM streptozotocin resulted in complete destruction of beta cells, whereas islets similarly exposed after a 24 hr culture period continued to secrete insulin for many months. Islets maintained in culture for one week before exposure to 0.1–0.5 mM streptozotocin, however, became more sensitive than freshly isolated islets. Repeated weekly exposure of cultured islets to a “non-toxic” concentration (0.1 mM) resulted in sustained suppression of insulin secretion after 11 weeks.

    Notizen: The solubilization of fish protein concentrate (FPC) by Bacillus subtilis protease (Monzyme) has been investigated. Conditions have been defined which eliminate the problems of microbial contamination and salt accumulation. A Kinetic treatment revealed that enzyme is adsorbed to the surface of the substrate, exposed to aqueous phase. The overall kinetics were described by a sequence of two first-order processes - an initial, fast reaction in which loosely bound polypeptide chains are cleaved from an insoluble protein particle, and a second, slower reaction in which more compacted core protein is digested.

    Notizen: We have demonstrated the use of bacterial chromatophores for the continuous photosynthetic regeneration of ATP from ADP in an ultrafiltration reactor. Biphasic kinetics of the degradation of chromatophore activity are described. Using chromatophores in combination with the enzyme adenylate kinase, we have also demonstrated continuous regeneration of ATP from AMP.

    Notizen: The immobilization of Rhodospirillum rubrum chromatophores was successfully performed by entrapping them in polyacrylamide. Their photophosphorylating activity was about 40% of native chromatophores. The temperature and pH optima for immobilized chromatophores were similar to the native photosynthetic apparatus and kinetic parameters showed that the rate of photophosphorylation in polyacrylamide particles was diffusion controlled. Light penetration of the gel particles was not a limiting parameter. Immobilization considerably increased the stability of the chromatophores towards denaturation.
