    Notes: The kinetics of the gluconic acid fermentation have been studied at pH 5.8 Pseudomonas ovalis B-1486 was used as the fermenting organism. D-Gluconolactone was found as an intermediate in the fermentation and accumulated at times in large amounts. When this occurred, hydrolysis of the lactone was found to be the rate limiting reaction. This can be, under certain conditions, a poor fermentation in which to study oxygen transfer limiting conditions. Further, it is impossible to relate batch and continuous kinetic data for this fermentation unless the lactone hydrolysis reaction is taken into account.

    Notes: This paper is concerned with the study of an enzymatic system in a repeated batch process where the enzyme is subject to deactivation. The particular system studied was the enzymatic hydrolysis of Penicillin G to 6-aminopenicillanic acid. Utilizing standard optimization techniques, pH and temperature control policies were determined that would maximize the product yield.

    Notes: A new fluorescent bioreactor monitoring probe - multiple excitation fluorometric system (MEFS) - has been developed. This probe was compared to the commercially available BioChem Technology FluroMeasure system (NADH probe). In this task the fluorescence behavior of three model fermentation systems, ethanol fermentation by Candida utilis, phenol fermentation by Pseudomonas putida, and glucose fermentation by Saccharomyces cerevisiae, were examined. The results indicated that the fluorescence intensity and behavior of various cellular fluorophors vary significantly between the different fermentation systems. Monitoring a fermentation process using only NAD(P)H fluorescence provided limited information. The NAD(P)H fluorescence was found not to be the best fluorescence signal for monitoring cell concentrations. The best way of monitoring a bioreactor by fluorometry may be to monitor several fluorophors in the whole culture broth simultaneously and to relate these fluorescence signals to various biological parameters.

    Notes: Continuous flow-type reactors have been used to study the kinetics of biological systems for quite some time. For continuous media sterilization, tubular flow reactors are particularly useful being simple in character and easy to control. However, one aspect quite often neglected in sterilization calculations is the residence time distribution of the reactor system. Serious errors in estimating the degree of bacterial destruction can be encountered if the residence time distribution is neglected; especially when a high degree of destruction is desired. This paper reports a study made to characterize and use the residence time distribution of a tubular reactor in the interpretation of high-temperature, short exposure time data for inactivation of Bacillus stearothermophilus spores. Mathematical models accounting for the residence time distribution of the tubular reactor have been proposed and employed to obtain high-temperature death-rate data.

    Notes: The products of cellulose hydrolysis, glucose and cellobiose, caused noncompetitive inhibition patterns to be exhibited when Thermomonospora sp. YX cellulase hydrolyzed dyed cellulose. The glucose binding constant, β1, was 0.00683 ± 0.00031 L/g and the cellobiose binding constant, β2, was 0.095 ± 0.0058 L/g. Thus, cellobiose was about 14 times more inhibitory than glucose.
