    Notes: The hepatic lobules of the family Suidae are unusually large, completely surrounded by fibrous tissue, and supplied by relatively large branches of the hepatic artery. The interlobular septa carry small branches of the artery, vein and bile ducts of the adjacent portal tracts and may be regarded as attenuated extensions of the portal tracts. The hepatic lobules of Tayassuidae and Hippopotamidae lack these distinctive features.

    Notes: The number and spatial relations of muscle spindles in the extensor indicis have been determined. Approximately forty spindles lie adjacent to the motor end plate zone dispersed rather equally through this area of the muscle with a modest increase in density among the distally originating fibers. A study of the entire innervation pattern of this muscle has also been completed.These findings have greatly facilitated the isolation of spindles for electron microscopic study and attempts to perform in vivo recording from the sensory nerves of muscle spindles in this laboratory. It is now possible, with the aid of suitable magnification and stimulating and recording devices, to more rapidly find muscle spindles relative to the gross innervation and the easily located motor end plate zone.

    Notes: Various physiological and anatomical studies have suggested that the ciliary epithelium may be under the influence of adrenergic nerve fibers. This study was undertaken in an attempt to characterize the innervation of the ciliary epithelium. Both light and electron microscopy revealed that the ciliary epithelium is only sparsely innervated. In addition, the ciliary epithelium overlies a highly vascular stroma, and the distance between the large vascular channels and the ciliary epithelium is small. These features of the stroma indicate it is structurally suited for a transport function, a fact which supports earlier reports that the elaborate membrane infoldings of the ciliary epithelium are indicative of a transport function. The discrepancy between previous descriptions of the innervation of the ciliary epithelium, demonstrating complex subepithelial plexuses of adrenergic nerve fibers, and the results of the present study may be due to the fact that the fluorescence technique used in previous studies could have labelled some other tissue component in addition to adrenergic nerve fibers. Results with orcein and aldehyde fuchsin stains indicate that the disposition of elastic fibers in the ciliary processes is similar to previous descriptions of adrenergic nerve fibers as determined by fluorescent techniques.

    Notes: The distribution of total polyadenylated RNA and mRNAs from the β-actin, fibronectin, and cytokeratin Endo A genes was examined in preimplantation mouse embryos using in situ hybridization of riboprobes to RNA in sections of embryos. Polyadenylated RNA was found in the cytoplasm of all cells of blastocyst-stage embryos, whereas the specific mRNAs displayed three distinct patterns of expression: uniform throughout the embryo (β-actin), enriched in the inner cell mass (fibronectin), and enriched in the trophectoderm (Endo A). In eight-cell embryos, the polyadenylated RNA was more concentrated in nuclei than in the cytoplasm (as noted previously), although this was not the case in blastocysts, nor was it true for the specific mRNAs that were examined. These experiments demonstrate that there is localized gene expression in the early mouse embryo, which correlates with the formation of the trophectoderm and the inner cell mass.

    Notes: Approximately 70% of all bladder cancers are superficial at the time of presentation. Superfecial bladder cancer includes tumors confined to the urothelium (clinical stage Ta) or lamina propria (stage T1) and flat carcinoma is situ (stage Tis). Because the biological behavior of bladder neoplasms is variable, several important prognostic factros must be addressed. Multivariate analyses have shown that factors predictive of tumor recurrence and tumor progression include multifocal tumors, high grade tumors. T1 tumors and positive urinary cytology after transurethral resection (TUR). The patient with superficial bladder cancer should be monitored via endoscopy supplemented by urinary cytology, using either voided or bladder irrigation specimens and urinalysis. Frequent intravenous urography is not required, even in high grade tumors, as long as the clinical and pathologic studies remain negative and the patient is asymptomatic.The “gold standard” of treatment for superficial is TUR of the entire tumor. Despite TUR, new tumors will occur in approximately 50% of all patients; those at highest risk for tumor recurrence and progression require adjuvant intravesical therapy after TUR. A variety of drugs are used as intravesical therapy, including thiotepa, mitomycin C, doxorubicin hydrochloride, Bacillus Calmette-Guerin (BCG), epirubicin, and interferon. Although associated with the most toxicity, BCG appears to be the most effecacious agent in increasing the time to recurrence and progression and in reducing the recurrence rate. © 1992 Wiley-Liss, Inc.

    Notes: To assess systematically the structural and functional aspects of histone gene transcription within a chromosomal context, we stably integrated an extensive set of human histone H4 gene constructs into mouse C127 cells. Levels of expression were determined by S1 nuclease protection assays for multiple mouse monoclonal cell lines containing these human H4 genes. For each cell line, we quantitated the number of integrated human H4 genes by Southern blot analysis. The results indicate that the expression of the human H4 gene is in part copy number dependent at low gene dosages. However, the level of expression varies among different cell lines containing similar numbers of copies of the same H4 gene construct. This result suggests that position-dependent chromosomal integration effects contribute to H4 gene transcription, consistent with the roles of long-range gene organization and nuclear architecture in gene regulation. At high copy number, the level of human H4 gene expression per copy decreased, and endogenous mouse H4 mRNA levels were also reduced. Furthermore, in vivo occupancy at the human H4 gene immediate 5′ regulatory elements, as defined by genomic fingerprinting, showed copy number-dependent protein/DNA interactions. Hence, human and mouse H4 genes compete for titratable transcription factors in a cellular environment. Taken together, these results indicate cross-species compatibility and suggest limited representation in vivo of the factors involved in regulating histone H4 gene transcription.

    Notes: Oogenesis and embryonic development in the marine sponge, Haliclona ecbasis, were studied using standard histological procedures.When the oocytes reach a diameter of about 30 μ, nurse cells begin to aggregate around them. Then when the oocytes are about 36 μ in diameter, they begin to engulf the associated nurse cells. Whole nurse cells are engulfed; and although the nucleus of the nurse cells disappears either as or soon after the cells are engulfed, the cytoplasm remains essentially unchanged. The accumulation of these cells within the oocytes most of the cytoplasm is nurse cell cytoplasm.During cleavage of the egg, the engulfed nurse cells are gradually fragmented, but otherwise appear unchanged. At the same time the cytoplasm of the nurse cells is progressively incorporated into that of the blastomeres by what appears to be fusion process. When the latter process is complete, the embryo develops into a typical parenchymula larva.

    Notes: Vernalized gemmules of the marine sponge Haliclona loosanoffi were cultured at 20°C, fixed at 24-hour intervals (0-11 days), and processed for light microscopy by using a variety of absorption and fluorescent staining methods. The cytochemistry and morphology of development were compared to the well-studied developmental patterns of freshwater sponges and to the patterns described in the marine sponge Suberites domuncula. The precocious development of H. loosanoffi gemmules involves early morphogenesis occurring within the unhatched gemmule, as opposed to the patterns in freshwater sponges, where most development occurs after the gemmule hatches. Definitive sponge tissue surrounding a single osculum is present 9 days after release from dormancy.
