    Notizen: Abstract Representative strains of Gardnerella vaginalis were degraded using both an alkaline and an acid methanolysis and the fatty acid methyl esters released examined by thin-layer and gas chromatography. The profiles obtained were both qualitatively and quantitatively similar and were comprised of straight chain saturated and unsaturated non-hydroxylated fatty acids with hexadecanoic acid (16:0) and octadecenoic acid (18:1) the major components. All of the strains contained very characteristic polar lipid patterns consisting of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, five partially identified glycolipids and an uncharacterised phospholipid. Analyses of wall amino acid preparations using gas chromatography showed that Gardnerella vaginalis strains contain major amounts of alanine, glycine, glutamic acid and lysine. The chemical data support the integrity of the genus Gardnerella.

    Notizen: Fast atom bombardment mass spectra were successfully recorded for intact glycosylphenolphthiocerol dimycocerosates (phenolic glycolipids, PGLs) from Mycobacterium kansasii, M. leprae, M. tuberculosis, M. marinum, M. bovis and M. haemophilum. Characteristic fragment ions from the loss of the oligosaccharide moiety and one of the long-chain multimethyl-branched mycocerosic acids were observed in most cases. A tandem mass spectrometric experiment was carried out on the PGL from M. tuberculosis, revealing the type of mycocerosic acids esterified to individual homologues. Mass spectra of homologues separated by reversed-phase high-performance liquid chromatography gave information on the substitution pattern in certain cases. The potential of matrix-assisted laser desorption ionization spectroscopy was demonstrated by a successful analysis of the PGL from M. tuberculosis.

    Notizen: Mycobacteria synthesize a variety of unusual long-chain fatty acid esters and the systematic analysis of these is of great potential in the classification and identification of these bacteria. This paper describes the extraction, reductive fission and derivatization to tert-butyldimethylsilyl ethers of β-diesters, the phthiocerol dimycocerosates. The mass spectra of these ethers are characteristic of the parent alcohols and selected ion monitoring techniques have been applied to mixtures extracted from strains of Mycobacterium tuberculosis.
