    Notizen: The present study was undertaken to examine the structural features that may be important to explain the immunogenicity of the (110-121) peptide sequence (FWRGDLVFDFQV) of VP3 capsid protein of hepatitis A virus. A conformational analysis of the preferred conformations by CD and molecular mechanics was carried out. Present results suggest that the interaction with liposomes as biomembrane model induces and stabilizes the amphipathic β-structure of the peptide.To study the contribution of amino acid replacements at the RGD tripeptide as well as the influence of the peptide chain length on peptide conformation, solid-phase peptide synthesis of several peptide analogs was carried out and the peptide conformation was studied using CD spectroscopy. The results show that the RGD sequence is necessary to induce the β-structure in the presence of liposomes. © 1998 John Wiley & Sons, Inc. Biopoly 45: 479-492, 1998

    Notizen: Trinuclear complexes containing pyrazolate bridging ligands of the type [NBu4]2[{(C6F5)2 M(μ-pz)(μ-X)}2M′] (M, M′= Pd or Pt; X = Cl, OH, or pz) (Hpz = pyrazole) have been prepared using [NBu4][M(C6F5)2(acac)] (acac = acetylacetonate) or [NBu4]2[M2(C6F5)4(μ-OH)2] and [M′Cl2(Hpz)2] or [M′(Hpz)4]2+ as starting materials. Asymmetric homo- and heterobimetallic complexes of the types [NBu4][R2M(μ-pz)2Pd(η3-allyl)] and [R2M(μ-pz)2M′L2] (allyl = C3H5 or C4H7; L2 = 2 PEt3 or bipy; R = C6F5 or C6Cl5; M, M′ = Pd or Pt) have been obtained starting from [NBu4][MR2(pzHpz)] and [Pd(η3-allyl)(acac)] or [M′L2Cl2], respectively. The identity of the new complexes has been established by NMR (1H, 19F and 31P) spectroscopy. The crystal structure of [{(C6F5)2Pd(μ-pz)(μ-Cl)}2Pd]2- has an inversion centre, with a bent appearance of the Pd3(μ-pz)2(μ-Cl)2 moiety. The crystal structure of [(C6F5)2Pd(μ-pz)2Pd(η3-C4H7)] has also been determined by single-crystal X-ray diffraction, where a boat conformation of the central “Pd2N4” six-membered rings is observed.

    Notizen: The compound cis-[Ni(C6F5)2(PhCN)2] has been used as the starting material for the preparation of the bimetallic complexes [(C6F5)2Ni(μ-X)2Ni(C6F5)2]2- (X = SCN, OCN, N3), [(C6F5)2Ni(μ-SCN)2Pd(C6F5)2]2-, [(C6F5)2Ni(μ-NCS)2Pd-(dppe)] [dppe = 1,2-bis(diphenylphosphanyl)ethane], and the trimetallic[(C6F5)2Ni(μ-NCS)2Pd(μ-SCN)2Ni(C6F5)2]2-. The mononuclear compounds [(C6F5)2NiLX] (L = PPh3, P(C6H4MeO-p)3; X = SCN, OCN) are obtained by reaction between [(C6F5)2Ni(&μ-X)2Ni-(C6F5)2]2- and L. The treatment of cis-[Ni(C6F5)2(PhCN)2] with [M(SAr)2(dppe)] leads to the formation of the arylthiolate complexes [(C6F5)2Ni(μ-SAr)2M(dppe)] (Ar = Ph, C6H4Me-p, C6H4NO2-p; M = Ni, Pd, Pt). The X-ray diffraction study of [(C6F5)2Ni(μ-SPh)2Pd(dppe)] shows that both metal atoms, Ni and Pd, are coordinated in a slightly distorted square-planar geometry and the μ-thiolato groups are in the syn conformation.

    Notizen: This work is concerned with the design of hip prostheses using advanced fiber reinforced composite materials. The major focus of the study is to evaluate how the stiffness and strength of composite hip prostheses can be affected by variations in ply orientation and stacking sequence for a selected manufacturing method. This investigation involved both analytical and experimental work. An analytical model was developed which consists of a stress analysis and a failure analysis. A finite element program was developed during the course of the investigation for analyzing stresses, strains, and deformations of composite stems with a simplified configuration. Failure and mode of failure were predicted by appropriately selected failure criteria. Experiments were also performed on T300/976 graphite/epoxy composites to verify the analysis and the computer calculations. Both testing and analysis accounted for the various combinations of in-plane and out-of-plane (torsion) loading that can act on the prosthetic hip. Simplified composite stems with a 120 layer thickness were fabricated and tested. An excellent agreement was found between the measured strain data and the numerical calculations. Using the program, parametric studies were performed. It was found that an optimal design of hip stems can be achieved by using advanced fiberreinforced composite materials, but great care must be taken when selecting the appropriate ply orientation and stacking sequence for a chosen fabrication method.

    Notizen: In this paper, we show the synthesis of palladium(II) 4,5-phenylimidazole cyclometalated complexes. They have been characterized by IR, 1H- and 13C-NMR spectroscopy. The cyclometalated dimer compound 2 [Pd(C15H11N2)(μ-OAc)]2 and the cyclometalated monomer compound 5 [PdBr(SEt2)(C15H11N2)], having OAc and Br as leaving groups, interact with DNA, modifying its secondary structure (as measured by Tm and CD), without modifying its tertiary structure (as determined by measurement of the electrophoretic mobility in agarose gels). The monomeric compound 5 seems to be the one that induces the highest alterations in DNA secondary structure since it strongly modifies the CD spectrum of the DNA. Melting data of drug-DNA complexes suggest that, at low drug concentration, the 4,5-Imd ligand intercalates between the base pairs in the DNA molecule, increasing the Tm, while at high drug concentrations the palladium(II) centers destabilize the double helix, producing a lowering in Tm values. These results indicate that complexes containing planar structures might selectively bind to DNA that is not supercoiled, and that therefore it only has a secondary structure. © 1997 John Wiley & Sons, Ltd.

    Notizen: In this paper we describe the synthesis and chemical characterization of three new Pd(II)-imidazoline complexes: [PdCl2 (C6H5-CH2-C3H5N2)2] (2), [PdCl(SEt2) (C6H4-C3H5N2)] (5) and [Pd(C6H4-C3H5N2) (μ-Br)]2 (6). We have also analyzed the DNA modifications and in vitro antileukaemic activity of these compounds and of their previously reported analogs [Pd Cl2 (C6H5-C3H5N2)2] (1), [Pd (C6H4-C3H5N2) (μ-OAc)]2 (3), [Pd (C6H4-C3H5N2) (μ-Cl)]2 (4) and [Pt(C6H4-C3H5N2)(μ-Cl] (7). All these compounds modify the DNA secondary structure since they alter the melting temperature (Tm) of the DNA. Circular dichroism spectra indicated, moreover, that compounds 3, 5 and 6 induced higher modification on the double helix than compounds 1, 2 and 4. While compounds 1, 2 and 5 seem to induce slight changes in the electrophoretic mobility of the open and covalently closed circular forms of pUC8 DNA at high ri (input molar ratio of Pd or Pt to nucleotides), compounds 3, 6 and 7 do not modify at any ri the tertiary structure of the plasmid DNA. Antileukaemic tests suggest that compounds 1, 4 and 7 exhibit important cytotoxic activity since their IC50 values against HL-60 human leukaemic cells were below 10 μg ml-1. © 1997 John Wiley & Sons, Ltd.

    Notizen: p-Isopropylbenzaldehyde thiosemicarbazone (p-is.TSCN) (1) reacts with [Pt(µ-Cl)(η3-C4H7)]2 to form a dinuclear [Pt(µ-Cl)(p-is.TSCN)]2 complex (2) and a cyclometallated cluster [Pt(p-is.TSCN)]4 (3). Biological testing of these complexes against HL-60 and U-937 human leukemic cells suggest that complexes 2 and 3 may be endowed with important cytotoxic activity properties since they exhibit IC50 values (50% inhibition of cell growth) in the micromolar range, as does the clinically used drug cisplatin (cis-DDP). Analysis of the interaction of compounds 2 and 3 with DNA indicates that the kinetics of DNA platination due to compounds 2 and 3 is faster than that of cisplatin and that after 24 h of incubation most of the platinum centers are bound to DNA. Thus, it is likely that the cytotoxic activity displayed by compounds 2 and 3 may be correlated with their high level of DNA platination. Copyright © 1998 John Wiley & Sons, Ltd.
