Notes: Metalloporphyrin Complexes. VI. Synthesis of Polymethacrylates with Covalently Bound Iron (III)-protoporphyrin IXConvenient synthesis of polymeric iron (III)-protoporphyrin IX (hemin) compounds is described. The covalent linkage via the carboxylic groups of hemin is possible by the reaction of the activated ester of hemin with polymethacrylates containing aliphatic amino groups. Polymethacrylates with aromatic amino groups are suitable to immobilize the hemin via its methine bridges. The hemin-containing polymers are characterized by visible spectra and e.p.r.-spectroscopy.

Notes: New imidazole containing polymers were synthesized by nucleophilic substitution reactions of poly(ethylene methacrylate arylsulfonates) with imidazole, benzimidazole and some of their derivatives. The reaction time and the solvents used had varying effects on the binding rates. The conversion was best reproducible at high reaction speed in ethanol.

Notes: Imidazole containing polymers were synthesized by polymer-analogous reactions of poly(ethylene methacrylate arylsulfonates) with substituted imidazole derivatives. The imidazole derivatives with exocyclic functional groups were bound to the polymer matrix via spacers. By further polymer-analogous reactions the exocyclic functional groups were modified, so as to give rise to imidazole containing polymers with primary amino groups or carboxyl groups.

Notes: The parameters and the measuring principle of a urease electrode are described. The urease is physically entrapped in cellulose triacetate on the active surface of a pH glass electrode with an internal reference electrode. The Michaelis constant of immobilized urease was similar to that of soluble enzyme (KM = 2.40 mmol/L). The average apparent enzyme activity amounts to 2 IU per sensor preparation. An enzyme loading factor of 0.04 was determined. It indicates kinetically controlled reaction conditions of the sensor. A fixed-time measuring procedure provides a response time of 30 seconds and a linear response within a concentration range of 2 to 40 mmol/L urea in real biological samples (e.g., serum).

Notes: In crude membrane fractions of rat pancreatic islets and of RIN-A2-cells, forskolin and NaF stimulated adenylate cyclase activity. Basal and stimulated enzyme activity was approximately 3 to 6 fold higher in membranes of RIN-A2-cells than in membranes of islet cells. In RIN-A2-cells GppNHp and NEM inhibited forskolin-stimulated cyclase system of RIN-A2-cells contains inhibitory and stimulatory N-proteins and that there are critical thiols related to Ni, Ns and/or the catalytic unit. Thus, membrane fractions of RIN-A2-cells may be an appropriate model for studies on the adenylate cyclase system of insulin-producing cells.
