    Notes: It is shown that the formation of the three-strand poly (A + 2U) complex is accompanied by a decrease in optical absorbance at 280 mμ, while the format ion of poly (A + U) is not. This difference makes it possible to demonstrate that in dilute solutions (∼10-4 monomolar), a 1 : 1 mixture of poly A and poly U always forms poly (A + U) at room temperature. At higher temperatures in solutions containing more than about 0.2M sodium ion, poly (A + U) is converted to poly (A + 2U) and free poly A. These results are consistent with the observations reported by Miles and Frazier, using infrared absorption spectra, at much higher polymer concentrations. It is concluded that most of the physicochemical studies of 1 : 1 mixtures of poly A and poly U in dilute aqueous solution refer to the two-stranded species.A simple thermodynamic analysis is developed to explain the two- to three-strand conversion, and with the use of a “phase diagram” showing conditions of stability of the various poly A-poly U structures as a function of salt and temperature, it is possible to estimate the value of the enthalpy of formation of poly (A + 2U).

    Notes: The double-stranded helical complexes of poly(dG-dC) and of poly(dG-m5dC) are shown to convert from B- to Z-DNA-type conformations at moderate or low ionic strengths, lower for the 5-methyl than for the non-methyl species, in a highly cooperative temperature-dependent equilibrium. In the presence of low concentrations of divalent ion, e.g., Mg2+, the temperature at which the B → Z transition occurs is virtually independent of the salt concentration and the B-conformation is favored at lower temperature, while the Z-conformation is favored at higher temperature. Since the Debye-Hückel screening parameter changes rapidly with ionic strength in this region, electrostatic interaction with the free ions appears to be only a small factor in the forces that promote the transition; the temperature dependence must derive principally from effects on the solvent. The temperature dependence at high salt concentrations is also reported.

    Notes: The effect of spermine upon the denaturation temperature (Tm) of DNA's of various base compositions has been found to depend upon both the base composition of the DNA and the pH of the solution. Measurement of the hydrogen ion titration curve of spermine as a function of temperature reveals that the net charge of the spermine molecule is undergoing a rapid change with temperature in the range of temperatures at which DNA denatures. Since the value of Tm depends upon base composition, the correlation of the effect of spermine upon Tm with the base composition of the DNA used may be explainable in terms of the changing degree of ionization of spermine. The binding of spermine to native DNA has also been studied by dialysis equilibrium. There is no significant variation either in the number of strongly binding sites or strength of binding with base composition. It is concluded that there is no evidence of correlation between the binding of spermine and the base composition of DNA.

    Notes: An exchange method is described for producing tritium-labeled native DNA in vitro with minimal physical damage to the DNA. Tritium-labeled calf thymus DNA prepared in this way has a specific activity of about 100 μCi/mmole of nucleotide (i.e., about 2 × 108 dpm/mmole). Sedimentation velocity at neutral and alkaline pH indicate that the product has an average of two single strand breaks per duplex molecule of molecular weight 6 × 106 daltons. The optical and thermal denaturation properties of the product are those of native DNA. The method should be particularly useful for labeling DNA from organisms that cannot be labeled conveniently in vivo.

    Notes: The conformation of single-stranded polyrA in aqueous solution has been measured at temperatures down to -12°C. The radius of gyration of low-molecular-weight polyrA varies very little with temperature in this range. By studying the dependence of radius of gyration on temperature for several polyrA fractions, we show that the dependence of the radius upon chain length is consistent with formation of a single-stranded rodlike structure at low temperature. The structure has an approximate length of 3.2 Å/nucleotide.
