Anomeric Methyl 2-Deoxy-4-thio-D-erythro-pentofuranosidesSelective benzoylation of 3-deoxy-1,2-O-isopropylidene-D-ribo-hexofuranosee (1) at -15° gives 6-O-benzoyl-3-deoxy-1,2-O-benzoyl-3-deoxy-1,2-O-isopropylidene-5-O-(p-tolylsulfonyl)-D-ribo-hexofuranose (3). 3 on treatment with sodium methoxide furnishes 5,6-anhydro-3-deoxy-1,2-O-isopropylidene-L-lyxo-hexofuranose (4) which with sodium benzyl oxide yields 6-O-benzyl-3-deoxy-1,2-O-isopropylidene-L-lyxo-hexofuranose (5). Treatment of 5 with p-tolylsulfonyl chloride gives 6-O-benzyl-3-deoxy-1,2-O-isopropylidene-5-O-(p-tolylsulfonyl)-L-lyxo-hexofuranose (6). Nucleophilic displacement of the tolylsulfonyloxy group of 6 with thioacetate anion provides 5-S-acetyl-6-O-benzyl-3-deoxy-1,2-O-isopropylidene-5-thio-D-ribo-hexofuranose (7). Selective hydrolysis of 7 affords 5-S-acetyl-6-O-deoxy-5-thio-D-ribo-hexofuranose (8). Glycol cleavage of 8 with neutral sodium metaperiodate yields 4-S-acetyl-5-O-benzyl-2-deoxy-4-thio-aldehyde-D-erythro-pentose (9) which is converted to a mixture of the anomeric methyl 5-O-benzyl-2-deoxy-4-thio-D-erythro-pentofuranosides (10 and 11) by treatment with methanolic hydrogen chloride. These syrupy compounds are converted respectively to their crystalline 3-O-brosylates (12 and 13). Debenzylation of each of the isomers 10 and 11 with sodium in liquid ammonia affords methyl 2-deoxy-4-thio-α-D-erythro-pentofuranoside (14) and methyl 2-deoxy-4-thio-α-D-erythro-pentofuranoside (15).

Die selektive Benzoylierung von 3-Desoxy-1.2-O-isopropyliden-D-ribo-hexofuranose (1) bei -15° liefert 6-O-Benzoyl-desoxy-1.2-O-isopropyliden-D-ribo-hexofuranose (2), die mit Tosylchlorid in 6-O-Benzoyl-3-desoxy-1.2-O-isopropyliden-5-O-tosyl-D-ribo-hexofuranose (3) übergeht. Aus 3 entsteht mit Natriummethylat 5.6-Anhydro-3-desoxy-1.2-O-isopropyliden-L-lyxo-hexofuranose (4), die durch Natriumbenzylat in 6-O-Benzyl-3-desoxy-1.2-O-isopropyliden-L-lyxo-hexofuranose (5) umgewandelt wird. Mit Tosylchlorid erhält man daraus 6-O-benzyl-3-desoxy-1.2-Oisopropyliden-5-O-tosyl-L-lyxo-hexofuranose (6). Die nucleophile Verdrängung der in 6 enthaltenen Tosyloxy-Gruppe durch Thioacetat-Anion führt zu 5-S-Acetyl-6-O-benzyl-3-desoxy-1.2-O-isopropyliden-5-thio-D-ribo-hexofuranose (7), die zu 5-S-Acetyl-6-D-benzyl-3-desoxy-5-thio-D-ribo-hexofuranose (8) hydrolysiert werden kann. Aus 8 gewinnt man durch Glykolspaltung mit neutralem Natriummetaperjodat 4-S-Acetyl-5-O-benzyl-2-desoxy-4-thio-aldehydo-D-erythro-pentose (9), die mti methanolischer Salzsäure zu einem Gemisch der anomeren Methyl-5-O-benzyl-2-desoxy-4-thio-D-erythro-pentofuranoside (10 und 11) reagiert. Die sirupösen Verbindungen 10 und 11 können durch Umsetzung mit p-Brom-benzolsulfochlorid in kristallines Methyl-5-O-benzyl-3-O-(p-brom-benzolsulfonyl)-2-desoxy-4-thio-α (bzw). β)-D-erythro-pentofuranosid (12 bzw. 13) übergeführt werden. 10 und 11 ergeben mit Natrium in flüssigem Ammoniak Methyl-2-desoxy-4-thio-α-D-erythro-pento-furanosid (14) und Methyl-2-desoxy-4-thio-α-D-erythro-pentofuranosid (15).
