    Notes: Abstract A group of 23 patients with advanced head and neck cancer were treated with highly selective intra-arterial (IA) cisplatin 150 mg/m2 delivered rapidly through microcatheters. The systemic effects of cisplatin were neutralized by concurrent administration of sodium thiosulfate. Two-to-threefold higher tumor platinum contents were detected in tumor biopsies after selective IA cisplatin administration compared to historicol controls (treated with 100 mg/m2 IA). Cisplatin-induced DNA modification in human tumor biopsies was quantitated using the antiserum NKI-A59. High levels of cisplatin DNA adducts were detected which correlated linearly with the tumor platinum content (r 2=0.62). The addition of radiotherapy to this high dose intensity cisplatin treatment resulted in a 92% complete response (CR) rate (12 of 13 patients achieved a CR). Since no difference in tumor platinum content was detected between patients receiving or not receiving radiotherapy (13 and 10 patients, respectively), but the response rate was substantially different (12 CR and 1 partial response with radiotherapy versus 6 partial and 4 non-responders without radiotherapy), these data suggest that the high platinum levels achieved by selective IA infusion were sufficient to produce enough interaction with radiotherapy to cause a 92% CR rate. Whether this interaction is additive or synergistic is as yet unclear.

    Notes: Abstract  The formation and persistence of platinum-DNA adducts were studied with immuno(cyto)chemical methods in male and female Sprague-Dawley rats treated with a single i.p. dose of carboplatin. Linear dose-effect curves were observed for kidney and liver with an immunocytochemical assay using NKI-A59 antiserum that recognizes intrastrand cross-links. With this method, no staining of the nuclei due to platinum-DNA damage could be observed in the spleen, testis, uterus, or ovary after administration of up to 80 mg/kg carboplatin. A homogeneous staining of the nuclei in the liver was observed. The nuclear staining in the kidney was somewhat more intense but less homogeneous, with small groups of intensely stained nuclei occasionally being seen in the outer cortex. An approximately 15 to 20-times lower dose of cisplatin than of carboplatin was needed to reach equal staining levels in the liver and kidney. Plateau staining levels in both tissues were reached at between approximately 8 and 48 h after administration of the carboplatin. This was followed by a significant reduction in the kidney samples, whereas the staining levels in the liver section seemed to be more persistent. No major difference was observed between male and female rats in the formation and removal of DNA damage in these tissues. The levels of the various DNA adducts were measured with a competitive ELISA in liver, kidney, spleen, testis, and combined ovary/uterus samples collected at 8 and 48 h after carboplatin administration. At both 8 and 48 h, the highest platination levels were observed in the kidney, followed—in decreasing order—by the liver, combined uterus and ovary samples, spleen, and testis. At 8 h after administration of carboplatin, the relative occurrence of the bifunctional adducts Pt-GG (34%), Pt-AG (27%), and G-Pt-G (32%), was similar in all tissues. The same held for the monoadducts that amounted to about 7% of the total DNA platination. These data indicate that in the first few hours after carboplatin treatment, no preference for the formation of Pt-GG adducts was observed, which confirms our earlier observations obtained with cultured cells. When the total DNA-platination levels (calculated from the sum of the adducts) seen at 8 and 48 h after treatment were compared, a substantial decrease in DNA platination was observed in the kidney (37%), liver (30%) and ovary/uterus (39%), whereas the repair levels in the testis (9%) and, probably, the spleen (18%) were substantially lower. In all tissues studied, only the relative occurrence of the Pt-GG adducts increased between 8 and 48 h, and as a result, at 48 h, after carboplatin administration the Pt-GG adduct was the major adduct persisting in the DNA samples.

    Notes: The versatility of the quadrupole ion trap as a mass spectrometer in which the mass resolution may be varied over a wide range has been demonstrated. Mass resolution of 0.8 × 107 and 1.2 × 107 have been obtained for ions of m/z 414 and m/z 614, respectively, well in excess of the normal value of 3m, where m is the mass of ion. At the highest mass resolution, peak-widths at half-maximum are but 52 × 10-6 u. While it has been shown previously that enhanced mass reslution in the quadrupole ion trap can be achieved by reduction of the mass scanning rate, the range of mass resolutions reported here were achieved by reducing the mass scanning rate, in stages, by an overall factor of ca 5.5 × 104.

    Notes: When the working point of a precursor ion confined within a quadrupole ion trap is moved to the vicinity of a boundary of the stability diagram, fragmentation of the precursor ion can be induced by virtue of energy gained from the radio-frequency trapping field. This behaviour is known as the ‘border effect’. When the isolated precursor ion is subjected to a cooling period of variable duration, in the presence of helium buffer gas and prior to experiencing the ‘border effect’, a high degree of control of the fragmentation pattern is achieved. The cooling period causes decreases in both ion kinetic energy and ion axial excursions. From this preliminary study, it is suggested that the deposition of internal energy during the ‘border effect’ can be varied by changing the duration of the cooling period. In addition to a degree of control of internal energy deposition, the amount of energy which can be deposited in this manner, though unknown, appears to exceed those energy levels achievable with resonance excitation and the ‘border effect’ without collisional cooling, in that an additional fragmentation channel is accessed.

    Notes: Commercial gas chromatography quadrupole ion-trap mass spectrometer (GC/ITMS) instruments have been used in a novel way for the study of negative ions formed from perfluorotributyl-amine and polychlorinated biphenyl compounds. Negative ions, formed during a normal electron impact ionization period in which 70 eV electrons were used, were isolated in an ion trap by the application of a negative DC voltage to the ring electrode that caused all positive ions to be ejected from the ion trap; in the subsequent analytical scan, negatives ions were detected with an electron multiplier biased normally so as to detect positive ions. The major negative-ion species detected from FC-43 were m/z 252 to m/z 633; 34 other negative-ion species were detected also, though in low abundance. The observed negative-ion mass range extended from m/z 252 to m/z 633. The signal-to-noise ratio of negative-ion mass spectra was enhanced by operation of the GC/MS instrument in GC mode and averaging the mass spectra acquired. In an examination of 3 hexachlorobiphenyl compounds, the molecular anion cluster around m/z 360 for the non-ortho-substituted congener 169 was observed with a signal-to-noise ratio of ca. 20 for 90 pg injected on column: the response for congener 169 was greater than that for congener 156 (mono-ortho-substituted) which, in turn, was greater than that for congener 153 (di-ortho-substituted).

    Notes: Beim System Stickstoff/Sauerstoff wurden bei Atmosphärendruck Wärme- und Stoffaustausch zwischen Dampf und Flüssigkeit verfolgt. Das Gas strömte dabei in Blasen, die an einer einzelnen Öffnung mit Kreisquerschnitt gebildet wurden, durch eine ruhende Flüssigkeit. Gaszusammensetzung, Flüssigkeits-zusammensetzung und Flüssigkeitsstand über der Düse konnten dabei beliebig, die Temperatur der Flüssigkeit, die Eintrittstemperatur des Gases, Düsendurchmesser und eingeblasener Gasstrom beschränkt variiert werden. Entstehung und Aufstieg der Blasen in der Flüssigkeit wurden in Zeitdehner-filmen festgehalten. Dabei wurde festgestellt, daß das Verstärkungsverhältnis in den untersuchten Intervallen der Variablen von folgenden Größen abhängt: Flüssigkeitszusammensetzung, Durchsatz durch die Düse, Austauschstrecke des Dampfes durch die Flüssigkeit und Richtungssinn des Stoffaustausches.

    Notes: A simulation study is described of the behaviour of ions confined in a quadrupole ion trap during each of two separate operations of a tandem mass spectrometric experiment. The two operations are those of mass-selective ion isolation and mass-selective resonance excitation to the point of ion ejection from the ion trap. The method of mass-selective ion isolation simulated is that of consecutive ion isolation. Simulation data indicate that the collisional history of the ions prior to the isolation process can greatly influence the degree to which ions survive this process. Simulation data for mass-selective resonance ejection are compared with experimental data obtained with a Finnigan-MAT ion trap mass spectrometer. In each operation, the facility with which ions absorb energy from the field within the ion trap, whether this field is derived from the R.F. drive potential or a supplementary potential, can determine the extent to which ions are retained within the ion trap during the two mass-selective operations described.
