    Notizen: Abstract The antidepressant and other behavioral effects of clorgyline, a preferential inhibitor of monoamine oxidase (MAO) type A, were compared with those of pargyline, a preferential inhibitor of MAO type B, in 16 depressed patients. In a subgroup of more severely depressed patients, clorgyline treatment for 4 weeks resulted in significant improvement on both observer-rated and self-rated scales, while minimal changes occurred during pargyline treatment. Similarly, in a crossover study that included 8 patients examined with multiple scales, clorgyline had generally greater antidepressant and antianxiety effects than did pargyline, although pargyline had some activating effects and also tended to produce more side effects. MAO type A inhibition may be more important than MAO type B inhibition for antidepressant efficacy.

    Notizen: Abstract Seven of nine depressed patients experienced a 4.3-fold increase in rated euphoria and activation following 30 mg d-amphetamine in a replicated dose, double blind study. d-Amphetamine was 2 to 2.3-fold more effective in producing activation, euphoria, and antidepressant effects than the same dose of l-amphetamine. Co-treatment with lithium carbonate produced a 60% (P〈0.001) attenuation of the activation and euphoria responses to d-amphetamine. The responses to l-amphetamine were almost completely abolished by lithium. This study raises the possibility of lithium carbonate use as an adjunct in the treatment of amphetamine addiction.

    Notizen: Abstract Large oral doses of l-tryptophan yielded negligible antidepressant effects in unipolar depressed patients, but partial antidepressant and antimanic effects in some patients with bipolar affective disorder. Marked changes in cerebro-spinal fluid, platelet and urinary levels of serotonin and 5-hydroxyindoleacetic acid in these patients during l-tryptophan administration suggest that the low response rate of severely depressed patients is probably not attributable to impaired l-tryptophan metabolism during depression.
