    Notizen: Summary The membrane phenotype of AML clonogenic cells (L-CFU) was analyzed in 19 AML patients using an in vitro culture technique after a complement-mediated lysis assay employing a panel of six monoclonal antibodies (McAb) -HLA-DR, FMC56 (CD9), FMC27 (CD9), CD14, CD15, CD41a-. Our results show that L-CFU has a heterogeneous but immature phenotype lacking on the expression of differentiation antigens (CD14, CD15, CD41a). In addition, we observed that the L-CFU phenotype is different from that of the whole blast cell population. Interestingly, L-CFU showed a higher expression of HLA-DR antigens with respect to their progeny. Upon analyzing whether the L-CFU phenotype was related to both the morphological and immunological features of AML blast cells, it was observed that, while there is no correlation with the FAB classification, there was a partial relationship between the immunological phenotype of AML blast cells and that of L-CFU. Accordingly, the more immature AML cases showed a more differentiated L-CFU phenotype (HLA-DR+, CD9+, FMC27+) when compared with cases with a more mature blast cell phenotype. These results suggest that those AML cases with a relatively immature myeloblastic phenotype may arise from a progenitor cell that has undergone partial differentiation and that is unable to acquire myeloid differentiation antigens, while those AML cases with mature blast cells might emerge from a very early L-CFU that has the capacity to undergo a greater degree of differentiation.

    Notizen: The paper describes an unexpected type of convergence behaviour occurring for a single, variable degree, primitive-type equilibrium element. For this element the number of independent stress fields is less than the number of independent boundary displacement variables that do not correspond to rigid element modes of displacement. This leads to the conclusion that the element is hypostatic and that, in the absence of self-stressing modes, no convergence can occur. Such ‘conventional’ counting procedures do not, however, reveal the whole picture, and numerical determination of the rank of the coefficient matrix of the equilibrium equations shows that, in practice, self-stressing modes can and do exist in a model which would conventionally be described as hypostatic. The rank deficiency in the coefficient matrix is shown to be due to the fact that, upon transformation, independent stress fields do not necessarily lead to independent boundary tractions. Generalization to conventionally iso- and hyperstatic models demonstrates that, whenever the coefficient matrix is rank-deficient, spurious kinematic modes coexist with self-stressing modes. The problem which reveals the curious convergence characteristics for the primitive-type element is resolved using a macro-type element, and it is seen that, with the larger degree of hyperstaticity available to this element, strictly monotonic convergence characteristics are observed. © 1997 John Wiley & Sons, Ltd.

    Notizen: Abstract We report here the isolation and characterization of a 2.3 kb genomicEcoRl fragment that co-localizes in the DNA puff C4 ofBradysia hygida with a 4 kbEcoRl fragment previously characterized as containing part of a gene amplified and expressed in the salivary gland at the time when puff C4 expands. Verification of the relative amount of DNA complementary to these two genomic fragments shows that they are unequally amplified in the salivary gland. The fragment containing part of the gene expressed when puff C4 expands amplifies about eight times more than the 2.3 kb fragment. This 2.3 kb fragment also carries sequences complementary to RNA species present in the gland in a period when puff C4 has already receded. Based on these data we discuss the nature of the DNA puff and the possible way in which amplification is occurring at these sites.

    Notizen: This paper illustrates a method whereby a family of robust equilibrium elements can be formulated in a general manner. The effects of spurious kinematic modes, present to some extent in all primitive equilibrium elements, are eliminated by judicious assembly into macro-equilibrium elements. These macroelements are formulated with sufficient generality so as to retain the polynomial degree of the stress field as a variable. Such a family of macro-elements is a new development, and results for polynomials of degree greater than two have not been seen before. The quality of results for macro-equilibrium elements with varying degrees of polynomial is demonstrated by numerical examples.
