    Notizen: Abstract A rare (arteriolo-)capillomedullary venous anomaly of the cerebellum was examined with CT, MRI, and angiography. Unlike the usual venous angioma, this case had such extraordinary features as: infratentorial location, involvement of the whole left cerebellum, an arteriolocapillary component and a history of progressive vomiting and chronic constipation in a 7-year-old boy. CT and MRI were consistent with an extensive vascular malformation, but the actual diagnosis was reached by angiography.

    Notizen: Abstract This review will focus on the recent advance in the study of effect of transmembrane Ca2+ gradient on the function of membrane proteins. It consits of two parts: 1. Transmembrane Ca2+ gradient and sarcoplasmic reticulum Ca2+-ATPase; 2. Effect of transmembrane Ca2+ gradient on the components and coupling of cAMP signal transduction pathway. The results obtained indicate that a proper transmembrane Ca2+ gradient may play an important role in modulating the conformation and activity of SR Ca2+-ATPase and the function of membrane proteins involved in the cAMP signal transduction by mediating the physical state change of the membrane phospholipids.

    Notizen: Abstract In order to investigate the effect of transmembrane Ca2+ gradient on Gs mediated coupling of β-AR and adenylyl cyclase, β-AR from duck erythrocytes and Gs and adenylyl cyclase from bovine brain cortices were co-reconstituted into asolectin liposomes with different transmembrane Ca2+ gradient. These proteoliposomes were proven to be impermeable to water-soluble substances. The results obtained indicate that a physiological transmembrane Ca2− gradient (1000-fold) is essential for higher stimulation of adenylyl cyclase by hormone-activated β-AR via coupling to Gs and can be further enhanced by the decrease of such Ca2+ gradient within certain range (100 fold) following Ca2+ influx into cells during signal transduction. Fluorescence polarization of DPH revealed that transmembrane Ca2+ gradient modulates adenylyl cyclase and its stimulation by hormones through mediating a change in lipid fluidity. Correspondent conformational changes of β-AR were also detected from the fluorescence spectra and quenching of Acrylodan-labelled β-AR in those proteoliposomes. It is suggested that a proper transmembrane Ca2+ gradient is essential for the optimal fluidity of the phospholipid bilayer in the proteoliposomes, which favors the formation of a suitable conformation of the reconstituted β-AR and thus promotes the stimulation of adenylyl cyclase activities by hormone-activated β-AR via Gs.

    Notizen: This paper presents a general direct integral formulation for potential flows. The singularities of Green's functions are desingularized theoretically, using a subtracting and adding back technique, so that Gaussian quadrature or any other numerical integration methods can be applied directly to evaluate all the integrals without any difficulty. When high-order quadrature formulas are applied globally, the number of unknowns can be reduced. Interpolation functions are not necessary for unknown variables in the present paper. Therefore, the present method is much simpler and more efficient than the conventional one. Several numerical examples are calculated and compared satisfactorily with analytical solutions or published results. © 1998 John Wiley & Sons, Ltd.

    Notizen: Local analysis schemes capable of detailed representations of the micro-features of a problem are integrated with a macro-scale BEM technique capable of handling complex finite geometries and realistic boundary conditions. The micro-scale effects are introduced into the macro-scale BEM analysis through an augmented fundamental solution obtained from an integral equation representation of the micro-scale features. The proposed hybrid micro-macro BEM formulation allows decomposition of the complete problem into two sub-problems, one residing entirely at the micro-level and the other at the macro-level. This allows for investigations of the effects of the micro-structural attributes while retaining the macro-scale geometric features and actual boundary conditions for the component or structure under consideration. As a first attempt, elastic fracture mechanics problems with interacting cracks at close spacings are considered. The numerical results obtained from the hybrid BEM analysis establish the accuracy and effectiveness of the proposed micro-macro computational scheme for this class of problems. The proposed micro-macro BEM formulation can easily be extended to investigate the effects of other micro-features (e.g. interfaces, short or continuous fibre reinforcements, voids and inclusions, in the context of linear elasticity) on macroscopic failure modes observed in structural components.
