    Notes: Summary This study represents an effort to tabulate the normal mean cross-sectional diameters of human striated muscle fibers in post-mortem material ranging in age from five months gestation through senescence. Age, sex, height and weight of the subjects were taken into account. Cases with neuromuscular illnesses or inanition were specifically excluded. All measurements represent mean narrow fiber diameter of celloidin embedded material sampled at the maximum diameter of the muscle belly. Noteworthy findings include a rapid increase in mean narrow diameter of all muscles except gastrocnemius from gestation to the immediate neonatal period. This was followed by a slower gradual increase in fiber diameter until the age of puberty when again a rapid increase was noted in all muscles except the superior rectus. Following puberty, the superior rectus diameter remained relatively constant throughout life. The sternomastoid, deltoid, biceps, sartorius, quadriceps and gastrocnemius continued a gradual steady increase in fiber size until the late third to early fourth decade, thereafter slowly diminishing in size by the ninth decade. Data are presented to show that the fusiform shape of the biceps muscle cannot be entirely attributed to the fusiform shape of the individual fibers. Particular care must be taken in selecting the level of measurement as fiber diameters appear to be significantly larger near the maximum breadth of the muscle bely. Factors are presented for conversion of measurements between various methods of histologic processing. A useful rule is that the ratio of the sizes of fresh-frozen, fixed-frozen, celloidin and paraffin embedded fibers is roughly 10:9:8:7.

    Notes: Abstract  Modulation of 5-fluorouracil (FUra) using leucovorin (LV) is a standard treatment approach in patients with metastatic colorectal cancer. Modulation of FUra with interferon alfa has also shown some promise. Laboratory data have demonstrated increased cytotoxicity when FUra is combined with both LV and interferon. The current study examined the effects of double modulation of FUra using LV and interferon.Patients with measurable advanced colorectal cancer received bolus FUra 375 mg/m2 plus LV 20 mg/m2 daily for 5 days, repeated every 28 days. Recombinant human interferon alfa-2a, 3 million IU/m2 subcutaneously, was given daily on the days of chemotherapy then three times weekly. There was one complete response and nine partial responses (10/41) seen for an overall response rate of 24% (95% CI 12.0–40.0%). Overall, 70% of patients experienced one or more episodes of nonhematologic toxicity of grade 3 or more. Weight loss was common, with a mean decrease of 2.9 kg over the first two months (P〈0.0001). Improvements in tumor-related symptoms were balanced by increased fatigue and a deterioration in body weight and performance status. There was no evidence of progressive changes in FUra metabolism from interferon usage.

    Notes: Summary In the 3-, 33- and 66-day-old chicken, two muscles, the oxidative slow tonic anterior latissimus dorsi and the glycolytic fast twitch posterior latissimus dorsi were compared by the measurement of muscle fibre diameter and the fraction of total muscle tissue nuclei which were either myonuclei or satellite cell nuclei. Between 3 and 33 days there was a period of rapid growth (more marked in the posterior latissimus dorsi) which coincided with a sharp fall in numerical density of myonuclei and satellite cell nuclei (number per cubic millimetre muscle tissue). The fraction of all nuclei which were satellite cell nuclei declined steadily. The higher levels of myonuclei and satellite cell nuclei in the anterior latissimus dorsi were thought to be a reflection of its oxidative metabolism and the presence of multiple endplates. The volume of sarcoplasm occupied by single myonuclei in anterior and posterior latissimus dorsi muscles was shown to be considerably greater than that occupied by nuclei in other cell systems.
