    Notes: Abstract We describe two asymptomatic newborns with nutritional vitamin B12 deficiency in whom increased urinary methylmalonic acid was detected by routine neonatal screening at 3 weeks of age. Both infants were exclusively breast-fed. One mother suffered from pernicious anaemia, and the other was a strict vegetarian. Both mothers had no clinical or haematological abnormality, aside from a borderline mean corpuscular volume for the vegetarian mother. This report illustrates the early appearance of functional vitamin B12 deficiency in breastfed infants of vitamin B12-depleted mothers. It also demonstrates that urinary methylmalonic acid measurement is a sensitive indicator of tissue vitamin B12 deficiency.

    Notes: Summary It is well documented that chloroplast DNA (cpDNA) recombination occurs at a relatively high frequency during sexual reproduction of unicellular green algae from the Chlamydomonas genus. Like the cpDNAs of most land plants, those of Chlamydomonas species are divided into two single-copy regions by a large inverted repeat sequence, part of which encodes the chloroplast rRNA genes. In the present study, we scored the inheritance of polymorphic loci spanning the entire chloroplast genome in hybrids recovered from reciprocal interspecific and F1 crosses between Chlamydomonas eugametes and C. moewusii, and from these data, estimated the density of recombination junctions within each region of recombinant cpDNAs. Our results indicate that recombination junctions occur at highly variable frequencies across the three main domains of the chloroplast genome. The large inverted repeat sequence was found to exhibit at least a five-fold higher density of recombination junctions compared to one of the singlecopy regions, whereas junctions in the latter region were five-fold more abundant relative to those in the other single-copy region. This marked difference in the densities of recombination junctions implies that the extent of genetic linkage between two given chloroplast loci will depend not only on their physical distance, but also on their locations within the genome.

    Notes: Abstract  A number of genes involved in the biosynthesis of the epicuticular wax (EW) of Arabidopsis thaliana have recently been isolated through genetic approaches. In view of the evidence in favor of the importance of EW compounds in the adaptation of higher plants to a number of physiological and ecological stresses, we have used clones of some of these genes to genetically engineer constructs with which to manipulate EW biosynthesis in transgenic A. thaliana plants. All our constructs were placed under the control of the near constitutive CaMV 35S promoter. We were able to complement mutant plants with the construction in the sense orientation as well as induce phenocopies of the eceriferum phenotype by transforming wild-type plants with both the sense and antisense constructs. We observed reduced fertility in the wild-type plants transformed with the 35SCER3sense or 35SCER3antisense constructs but not in those transformed with the 35SCER2sense or 35SCER2antisense constructs.
