    Notizen: Abstract The Y chromosome (Yaa gene) from autoimmune BXSB mice has been shown to be responsible for the acceleration of autoimmune symptoms when transferred to MRL/lpr mice. We examined the pathological, serological and immunological characteristics of MRL/lpr.Yaa mice and the suppressive effect of cyclophosphamide (CP) on the mice. MRL/lpr.Yaa mice spontaneously developed a massive lymphadenopathy characterized by hypergammaglobulinemia, the presence of several autoantibodies, and autoimmune disease. In MRL/lpr.Yaa mice, IL-2, IL-4 and IL-5 production in concanavalin A (Con A)-stimulated splenocytes were about 10-fold lower than in BALB/c mice at 5 weeks of age.The concentrations of these lymphokines remained low until the mice were 16 weeks of age. The production of IFN-γ and IL-6 in 16 week old MRL/lpr.Yaa mice was about 4- and 2-fold lower, respectively, though these levels were similar in both strains at 8 weeks of age. It was found that this dysregulation of T cell function was almost identical to that in MRL/lpr mice. Administration of CP to MRL/lpr.Yaa mice ameliorated nephritis, and suppressed production of autoantibodies and the accumulation of abnormal T cells. CP also significantly elevated the production of lymphokines. These findings suggest that an abnormality of T cell function may contribute to the autoimmune pathogenesis of MRL/lpr.Yaa mice and that CP probably ameliorates autoimmune disease by improving the T cell functions.

    Notizen: Abstract A full-length cDNA of the gene for phytochrome A from Arabidopsis thaliana was fused with the 35S promoter of cauliflower mosaic virus (CaMV35S-PHYA) and introduced into horseradish (Armoracia rusticana Gaert., Mey. et Scherb.) hairy roots. The phytochrome level in hairy roots transformed with CaMV35SPHYA was about three times greater than that in normal hairy roots and the rate of light-induced formation of adventitious shoots was also higher in the hairy roots transformed with CaMV35SPHYA. These results indicate that the light-induced formation of adventitious shoots on horseradish hairy roots is closely related to the phytochrome level.

    Notizen: Abstract  Three cDNAs for phytochrome A (phyA), designated ArPHYA1, ArPHYA2 and ArPHYA3, respectively, were isolated from a cDNA library prepared from proximal parts of the hairy roots of horseradish (Armoracia rusticana). The level of phyA was higher in those parts than in any other parts of hairy roots. The amino acid sequences deduced from the cDNAs were quite similar, with stronger homology to the product of PHYA of Arabidopsis thaliana than to products of genes for phytochromes of many other plant species. Levels of transcripts in roots and tubers were higher than those in leaves and petioles. The levels in hairy roots were highest at the proximal ends and increased with increased duration of culture in darkness. Furthermore, the levels in proximal parts decreased by 25–50% after light irradiation. The elevated levels of transcripts of PHYA genes at the proximal ends of hairy roots are discussed with reference to the light-induced formation of adventitious shoots.
