The System 2-Haloethyl Ester / Choline Ester as a Two-Step Protective Group of the Carboxylic Function in Peptide ChemistryThe system 2-chloroethyl- and 2-bromoethyl ester/choline ester was investigated as a carboxylic acid protective function. The 2-chloroethyl-(OEt(Cl))- and 2-bromoethyl esters (OEt(Br)) (4) of amino acids are easily prepared and coupled smoothly with Boc- or Z-protected amino acids to the corresponding dipeptide esters 5 or 6. These esters are stable in moderately acidic and basic media; they are converted into the 2-iodoethyl esters 8 via sodium iodide in DMF and are then cleavable by zinc in an one-pot procedure. The hydrophobic 2-haloethyl esters may be modified by trimethylamine to give the hydrophilic choline esters 10 and 11, which are very stable to acids but which may be cleaved without racemization in a medium of 10-4 M hydroxyl ion concentration.
Notizen:
