    Notizen: Abstract Background. We constructed the expression profile of proximal tubules, which is a database of 3′-directed partial cDNA sequences randomly collected from mouse proximal tubules. By comparing lists with those of various tissues, genes unique to each tissue can be identified. Methods. One of the sequences, GS4068, corresponding to a tissue-specific gene found only in mouse renal proximal tubules, was cloned and identified as mouse uroguanylin. Northern blot analyses were performed using mRNA isolated from the kidney and intestine of dehydrated and NaCl-loaded mice. In situ hybridization was done to localize its expression in the kidney. Results. In situ hybridization demonstrated that it was located around the corticomedullary junction of the kidney. Seventy-two h of dehydration induced the mRNA expression in the kidney but not in the intestine. Acute NaCl loading, however, did not induce mRNA in the kidney or in intestine. Conclusion. Mouse uroguanylin was localized presumably in the proximal tubules of the kidney. Its mRNA in the kidney was induced by 72-h dehydration, but not by acute NaCl loading.
