    Notizen: Abstract The sex pheromone component of maleLutzomyia longipalpis tergal gland extract was isolated and its activity confirmed by bioassay. Whole tergal gland extract was analyzed by HPLC and fractions were collected as they eluted from the detector. Each fraction was tested in an attraction bioassay with virgin unfed femaleLutzomyia longipalpis. HPLC analysis showed that whole extract contained several peaks; one large peak, one small peak and several minor peaks. Purity of the HPLC fractions was determined by GC analysis. The bioassays revealed that the large peak was responsible for most of the observed female behavior. The addition of the small peak to the large peak improved the response although by itself the small peak failed to elicit any significant behavior. Minor peaks failed to elicit any response. Chemical analysis revealed the large peak to be a relatively nonpolar hydrocarbon.

    Notizen: Abstract The occurrence inHotea gambia adults of a sexual dimorphism in the divided dorsal abdominal scent gland (dg 1)is reported. Counts made of ducted secretory units indicate that female dg 1 regresses at the end of larval development, unlike male dg 1 which undergoes no regression. Other dorsal abdominal scent glands (dg 2, dg 3) which function in the larvae cease to function during the imaginai moult. From gas chromatographic, mass spectrometric and [1H]NMR data, the identity of the secretion from male adult dg 1 was established as virtually pure (E)-2-hexenol (a 100-mg mature male adult contains 0.5–1 μl of secretion). 2-Hexenol was also found in the reduced female adult dg 1. In the sexually monomorphic metathoracic scent gland, (E)-2-alkenals (C6, C8) and (E)-4-oxohex-2-enal, together with monoterpenes (β-pinene, limonene) but not 2-hexenol, were identified. The vapor of (E)-2-hexenol is repellent to both sexes ofHotea adults and toxic to blowfly (Calliphora) eggs.

    Notizen: Tumor-promoting phorbol esters, such as 12-0-tetradecanoyl-phorbol-13-acetate (TPA), stimulate mouse peritoneal exudate macrophages to undergo DNA synthesis without added macrophage growth factor (MGF). Resident peritoneal macrophages do not respond in this manner. Plasminogen activator (PA) levels of both exudate and resident peritoneal macrophages are elevated by TPA. Thus the phorbol esters appear to mimic the action(s) of MGF and may be useful in understanding the events involved in macrophage and precursor cell growth responsiveness.

    Notizen: The tumor-promoting phorbol diester, 12-O-tetradecanoylphorbol-13-acetate (TPA) was found to act both independently of and synergistically with the mononuclear phagocyte specific colony stimulating factor (CSF-1) to stimulate the formation of macrophage colonies in cultures of mouse bone marrow cells. In contrast, TPA did not synergize with other CSF subclasses that stimulate the formation of eosinophil, eosinophil-neutrophil, neutrophil, neutrophil-macrophage, and macrophage colonies, nor with either of the two factors required for megakaryocyte colony formation, megakaryocyte CSF, and megakaryocyte colony potentiator. In serum-free mouse bone marrow cell cultures TPA retained the ability to independently stimulate macrophage colony formation. However, TPA-stimulated colony formation was suboptimal and delayed in serum-free cultures that could support optimal colony formation in the presence of CSF-1. In addition, TPA did not directly compete with [125I]CSF-1 at 4°C for its specific, high-affinity receptor on mouse peritoneal exudate macrophages. However, a 2-hour preincubation of the cells with TPA at 37° caused almost complete loss of the receptor. Thus, TPA is able to mimic CSF-1 in its effects on CSF-1 responsive cells in some aspects (the spectrum of target cells, the morphology of resulting colonies, and the ability to down-regulate the CSF-1 receptor) but it is not able to mimic CSF-1 in other ways (TPA alone cannot stimulate the full CSF-1 response, TPA does not stimulate the most primitive CSF-1 responsive cells, and TPA does not bind to the CSF-1 receptor).

    Notizen: Colony-stimulating factors (CSFs) stimulate granulocyte-macrophage production from single hemopoietic progenitor cells. Various preparations of purified CSFs of two different subclasses have been shown here to stimulate a plasminogen-dependent fibrinolytic (plasminogen activator) activity from resident and starch-induced mouse peritoneal macrophages. Lymphocyte supernatants also stimulate macrophage plasminogen activator (PA) activitty. Since they contain colony stimulating activity, it is possible that one or more sublcasses of CSF in these supernatants is responsible for this effect. Since both colony-stimulating and macrophage growth activities have been detected at inflammatory sites, these findings could reflect a role for CSF in inflammatory processes.
