    Notizen: Synopsis The functional morphology of the jaws of six species of butterflyfishes was investigated and related to their feeding behaviors. Utilizing observations and measurements of fresh-killed specimens as well as scanning electron microscopy of their dentitions, interspecific differences in the size, shape, orientation of the mouth, dentition, degree and function of jaw protrusion were related to their different feeding behaviors. The jaws of the six species showed a variety of adaptations for feeding on or over the reef as well as a repertoire of modulated jaw movements hitherto unnoticed in many studies. This radiation in morphologies was believed to be due to relatively simple changes of a few structural elements, changes that could occur relatively rapidly in the evolutionary time scale. The benefit of naturalistic observations in such functional morphological studies was emphasized.

    Notizen: Synopsis Ontogenetic dietary shifts are well studied in fishes. However, these shifts are rarely correlated with habitat and morphology. This study investigates: (1) ontogenetic dietary changes in mullet; and if dietary shifts do occur (2) whether they are correlated with feeding location, feeding behavior and/or feeding morphology. Stomach contents from eight size classes, each with 25 individuals, were analyzed using a gravimetric sieve fractionation procedure, Czechanowski's Quantitative Index and cluster analysis. Core samples (5 mm deep) were also analyzed in the same manner. Morphological characters were measured to detect differences among the size classes. Cluster analysis showed three distinct trophic units for Mugil cephalus in Cross Bayou: (1) 20.0 mm standard length — 30.0 mm SL; (2) 30.0 mm SL — 40.0 mm SL; and (3) 40.0 mm SL — 〉 100 mm SL. Comparison of the gut contents to the sediment contents for each size class showed an increase in similarity as the fish grew. Mullet gut contents from fish 20–25 mm SL were only 45% similar based on the weights of the prey items to the sediment subsample prey items, while stomach contents from fish 〉 100 mm SL were 84% similar to the sediment subsample contents. Gill raker length and interraker spacing of four gill arches were allometric with respect to standard length. Prey particle size within size classes appeared to be more correlated with feeding behavior than gill interraker spacing because smaller particles than predicted by the interraker spacing were ingested. It is suggested that smaller fish may be browsing more selectively than larger fish which graze.

    Notizen: Synopsis The functional morphology of the feeding apparatus and the feeding ecology of an assemblage of ten species of butterflyfishes was investigated using a comparative ecomorphological approach. Behavioral observations in situ and in vivo, morphological measurements of fresh-killed specimens, scanning electron microscopy, and kinematic modeling were utilized. The fishes show varying degrees of morphological and behavioral specialization and generalization. The more specialized species group according to how they feed, rather than what they feed on. The feeding guild concept is therefore not very instructive in understanding the functional significance of the feeding apparatus. Many of the morphologically specialized butterflyfishes demonstrate evolutionary convergence in feeding morphology. Whereas the more morphologically specialized species do generally have more ecologically specialized diets, these data do not particularly support the ecomorphological hypothesis in that similar diets do not correspond to similar morphologies.

    Notizen: Synopsis Ecomorphological correlates were sought among ten species of distantly related subtropical seagrass fishes. Morphometric data associated with feeding and microhabitat utilization were compared by principal components analysis, cluster analysis, and canonical correspondence analysis to dietary data. Morphology was generally a poor predictor of diet except for a group of mid-water planktotrophic filter feeders. Separation of the species along morphological axes appears to be related more to microhabitat utilization resulting in three major groups: (1) a group of planktotrophic, mid-water fishes specialized for cruising and seeking out evasive prey characterized by a compressed fusiform body, forked caudal fin, long, closely spaced gill rakers, short to intermediate! length pectoral fin, pointed pectoral fin, large lateral eye, short head, and a terminal or subterminal mouth; (2) slow swimming, less maneuverable epibenthic fishes that pick or suck their prey off the substrate. They are united by more rounded caudal and pectoral fins, and short or no gill rakers; and (3) a group of more mobile and maneuverable epibenthic foragers characterized by a more compressed, sub-gibbose body, long, pointed pectoral fins, forked caudal fins, large lateral eyes, subterminal mouth, and greater jaw protrusibility. Cases of convergence in trophic and microhabitat utilization characters were apparent in some of the groups.

    Notizen: The anatomy of the feeding apparatus of the lemon shark, Negaprion brevirostris, is investigated by gross dissection, computer axial tomography, and histological staining. The muscles and ligaments of the head associated with feeding are described. The upper and lower jaws are suspended by the hyoid arch, which in turn is braced against the chondrocranium by a complex series of ligaments. In addition, various muscles and the integument contribute to the suspension and stability of the jaws. The dual jaw joint is comprised of lateral and medial quadratomandibular joints that resist lateral movement of the upper and lower jaws on one another. This is important during feeding involving vigorous head shaking. An elastic ethmoplatine ligament that unites the anterior portion of the upper jaw to the neurocranium is involved with upper jaw retraction. The quadratomandibularis muscle is divided into four divisions with a bipinnate fiber arrangement of the two large superficial divisions. This arrangement would permit a relatively greater force per unit volume and reduce muscle bulging of the jaw adductor muscle in the spatially confined cheek region. Regions of relatively diffuse integumental ligaments overlying the adductor mandibulae complex and the levator palatoquadrati muscle, interspersed with localized regions of longer tendonlike attachments between the skin and the underlying muscle, permit greater musculoskeletal movement relative to the skin. The nomenclature of the hypobranchial muscles is discussed. In this shark they are comprised of the unsegmented coracomandibularis and coracohyoideus, and the segmented coracoarcualis. © 1995 Wiley-Liss, Inc.

    Notizen: Nerve terminal-smooth muscle relationships were studied in pulmonary arteries of the cat using 5-hydroxydopamine to help differentiate adrenergic and nonadrenergic terminals. There was a periarterial plexus of nerves in the walls of pulmonary arteries that extended into the lung to innervate even small arteries having a single layer of smooth muscle cells. Adrenergic nerves surrounded all arteries and extended into the tunica media of the large arteries. There were also apparent cholinergic nerves around the pulmonary arteries, although this was confirmed by electron microscopy for medium- and small-sized arteries only. The relationships of nerve terminals to smooth muscle cells in pulmonary arteries suggest that release of norepinephrine by adrenergic terminals can produce both decreased compliance and increased resistance in the pulmonary vascular bed, and that acetylcholine released by cholinergic terminals may act directly on vascular smooth muscle or on adrenergic terminals to modulate release of norepinephrine. These results suggest that both sympathetic and parasympathetic nerves may have a regulatory role in the pulmonary circulation.

    Notizen: A 66-kDa sialoglycoprotein has been identified as the parental membrane molecule of an earlier described sialoglycopeptide (SGP), an 18-kDa molecule released by protease treatment of intact bovine cerebral cortex cells that was shown to be a potent inhibitor of cellular proliferation. The 66-kDa parental sialoglycoprotein (p-SGP) was purified approximately 2,400-fold, to apparent homogeneity, from bovine cerebral cortex cell membranes by its release during incubation with 3 M NaCl, preparative isolelectric focusing and lectin affinity chromatography. Although a membrane-associated molecule, the p-SGP appeared to be tightly bound to the cell membrane, since it was not released during incubations in the absence of 3 M NaCl. Incubation of the membrane preparations with 3 M urea proved to be too harsh, and the antigenicity required to follow the purification of the p-SGP was abolished. Analyses by SDS-PAGE, under reducing and nonreducing conditions, suggested that the p-SGP membrane component was a single polypeptide without subunit structure. The p-SGP was shown to be structurally related to the SGP fragment by immunoblots with IgG raised to the SGP inhibitor, and functionally related to the SGP by its ability to inhibit Swiss 3T3 proliferation at concentrations strikingly similar to that previous measured with the SGP fragment.

    Notizen: A novel cell regulatory sialoglycopeptide (CeReS-18), purified from the cell surface of bovine cerebral cortex cells has been shown to be a potent and reversible inhibitor of proliferation of a wide array of fibroblasts as well as epithelial-like cells and nontransformed and transformed cells. To investigate the possible mechanisms by which CeReS-18 exerts its inhibitory action, the effect of the inhibitor on the posttranslational regulation of the retinoblastoma susceptibility gene product (RB), a tumor suppressor gene, has been examined. It is shown that CeReS-18 mediated cell cycle arrest of both human diploid fibroblasts (HSBP) and mouse fibroblasts (Swiss 3T3) results in the maintenance of the RB protein in the hypophosphorylated state, consistent with a late G1 arrest site. Although their normal nontransformed counterparts are sensitive to cell cycle arrest mediated by CeReS-18, cell lines lacking a functional RB protein, through either genetic mutation or DNA tumor virus oncoprotein interaction, are less sensitive. The refractory nature of these cells is shown to be independent of specific surface receptors for the inhibitor, and another tumor suppressor gene (p53) does not appear to be involved in the CeReS-18 inhibition of cell proliferation. The requirement for a functional RB protein product, in order for CeReS-18 to mediate cell cycle arrest, is discussed in light of regulatory events associated with density-dependent growth inhibition. © 1994 Wiley-Liss, Inc.

    Notizen: Glucose is essential for the energy metabolism of some cells and conservation of glucose is obligatory for survival during starvation. The principal site of this glucose conservation is the mitochondrial pyruvate dehydrogenase (PDH) complex, which is regulated by reversible phosphorylation (phosphorylation is inactivating). In cells in which glucose oxidation is switched off during starvation, fatty acids are used as fuel, and acetyl CoA and NADH formed by β-oxidation promote phosphorylation of PDH complex by activation of PDH kinase. A longer-term mechanism further increases PDH kinase activity in response to cAMP and products of β-oxidation of fatty acids. Coordinated inhibition of glycolytic flux mediated by effects of citrate on PFK1 and PFK2 in muscles and liver results in an associated inhibition of glucose uptake. Similar mechanisms lead to impaired glucose oxidation in diabetes. © 1994 Wiley-Liss, Inc.

    Notizen: Previous attempts to physically separate the cell cycle inhibitory and protease in activities in preparations of a purified cell regulatory sialoglycopeptide (CeReS) inhibitor were largely unsuccessful. Gradient elution of the inhibitor preparation from a DEAE HPLC column separated the cell growth inhibitor from the protease, and the two activities have been shown to be distinct and non-overlapping. The additional purification increased the specific biological activity of the CeReS preparation by approximately two-fold. The major inhibitory fraction that eluted from the DEAE column was further analyzed by tricine-SDS-PAGE and microbore reverse phase HPLC and shown to be homogeneous in nature. Two other fractions separated by DEAE HPLC, also devoid of protease activity, were shown to be inhibitory to cell proliferation and most likely represented modified relatives of the CeReS inhibitor. The highly purified CeReS was chemically characterized for amino acid and carbohydrate composition and the role of the carbohydrate in cell proliferation inhibition, stability, and protease resistance was assessed. © 1995 Wiley-Liss, Inc.
