Notizen: Surfaces of implantable and blood contact-devices accumulate adsorbed and denatured proteins. This anomalous layer of proteins may help trigger unwanted events such as activation of coagulation systems and, perhaps, chronic inflammation. Because, in many experimental systems, the purposeful coating of surfaces with albumin will biologically “passivate” materials, we have attempted to develop polymers which, when exposed to blood or body fluids, will spontaneously, selectively, and reversibly adsorb host albumin. We report here a novel derivatization technique for increasing the albumin affinity of implantable polyetherurethane (PU). The technique is based on the incorporation of high-molecular-weight dextran to which the albumin-binding dye Cibacron Blue is covalently attached. Somewhat surprisingly, the amounts of human albumin adsorbed by Blue Dextran-modified and unmodified PU are quite similar. There are, however, important differences. First, the binding of albumin to derivatized PU is specific and not readily blocked by proteins in albumin-depleted human serum. Second, the majority of albumin associated with derivatized PU appears to be reversibly bound. Third, the binding of albumin to derivatized PU evidently is mediated primarily through ligandspecific binding of the protein to the albumin-binding dextran-dye conjugate. We conclude that it is possible to produce implantable polymers having surfaces which display albumin-binding dyes that selectively and reversibly bind albumin. Materials with this property, when implanted or exposed to blood, should form an infinitely renewable coating of albumin derived from physiologic fluids. This surface modification strategy may spawn a new generation of implantable materials with improved biologic compatibility.

Notizen: Synthesis and characterization of 4-acetoxybenzoic acid and 6-acetoxy-2-naphthoic acid, labelled with deuterium in the rings and with carbon-13 and oxygen-17 in the carboxyl-positions, and of the corresponding homo- and copolyesters, are described. Kinetic studies of hightemperature deuteration were performed on 6-hydroxy-2-naphthoic acid. Mass spectrometry provides information about the degree of labelling with 2H, 13C and 17O. Composition and molecular weight of isotopically labelled polymers were monitored by 1H NMR after digestion in basic methanol. The thermal behavior was investigated by differential scanning calorimetry and thermogravimetry.

Notizen: The anisotropy of the thermal expansion of polyimide films was investigated . Out-of-plane or thickness direction coefficients of linear thermal expansion (CTE) were calculated from the difference between the coefficient of volumetric expansion (CVE) and the sum of the in-plane or film direction coefficients of linear thermal expansion for commercial and spin-coated PMDA//ODA and BPDA//PPD films and spin coated BTDA//ODA/MPD films. The CVEs were obtained from a pressure-volume-temperature (PVT) technique based on Bridgeman bellows. The CVE was shown to be essentially constant, independent of molecular orientation and thickness. A decrease in the in-plane CTEs therefore occurs at the expense of an increase in the out-of-plane CTE. In all cases the calculated out-of-plane CTE was higher than the measured in-plane CTE. The ratio of the out-of-plane CTE to the in-plane CTE was 1.2, 3.8, and 49.3 for the spin-coated BTDA//ODA/MPD, PMDA//ODA, and BPDA//PPD films, respectively. © 1994 John Wiley & Sons, Inc.

Notizen: A new Intermediate Scale Heat Release Rate Test Method is being developed primarily for testing assemblies in the vertical mode. Using the oxygen consumption calorimetry methodology and exhaust collection systems that already exist in many laboratories in the US and around the world, the apparatus is very economical to install. The sample size lends itself to reptitive testing in a short period of time. Sample preparation is generally quick and easy. Further development of the method should progress now that the method has been introduced at ASTM and will be introduced formally into ISO in the near future. There has been a favorable response by several laboratories who are considering construction of the apparatus. Additional assemblies tests are already underway and additional data will be presented in the near future.
