Summary Over the last 10 years, much has been learned about the human protein C anticoagulation system, a key pathway in maintaining blood in a fluid state in vivo. Three major glycoproteins, namely protein C, protein S and thrombomodulin, are involved in this pathway. The aim of this article is to discuss therapeutic potentials and issues and challenges for developing recombinant protein C, protein S and soluble thrombomodulin as novel antithrombotics. We also briefly discuss other drug design strategies that may be employed in modulating the activity of the protein C anticoagulant pathway under pathological conditions.
