Notes: Abstract Secretoneurin is a newly discovered peptide found in high concentrations in brain. We have studied the release of secretoneurin and noradrenaline from superfused hypothalamic slices from rat brain. Both electrical stimulation and potassium induced depolarisation released secretoneurin and noradrenaline from these slices in a calcium-dependent manner. Electrical stimulation caused a preferential release of noradrenaline when compared to the secretion elicited by high potassium. The time course of secretoneurin release was more protracted than that of noradrenaline. The calcium channel blocker ω-conotoxin inhibited only the electrically induced release of noradrenaline, whereas nifedipine inhibited only that of secretoneurin. These results establish that secretoneurin is secreted from neurons. Inhibition of this release by nifedipine is consistent with the concept that secretion from large dense core vesicles occurs at sites different from that of small vesicles and depends on calcium influx via L-type calcium channels.

Notes: Summary 1. 3H-leucine and3H-tyrosine were added to the perfusion medium of isolated bovine adrenal glands in order to study the synthesis and secretion of radioactively labelled catecholamines and proteins. 2. Various times after the injection of the labelled amino acids the soluble proteins of the microsomal fraction were isolated and subjected to polyacrylamide gel electrophoresis in three different buffer systems. The distribution of protein-bound radioactivity within the gels was determined. Most of the label was found to be confined to proteins which behaved like the specific soluble proteins of chromaffin granules, i.e. the chromogranins. 3. Stimulation of the adrenal gland with carbachol induced a release of catechol-amines, protein, and protein-bound radioactivity. In the absence of Ca2+ the secretion of all these components was abolished. The highest specific radioactivity of the proteins secreted upon stimulation was reached 4 h after the injection of the labelled precursors. The labelled proteins, secreted upon stimulation, could be identified as chromogranins. 4. Carbachol induced the release of highly labelled catecholamines after the injection of3H-tyrosine. The highest specific radioactivity of these catecholamines was already observed at the first stimulation with carbachol i.e. 30 min after3H-tyrosine. 5. These results demonstrate that in isolated bovine adrenal glands radioactively labelled chromogranins and catecholamines can be synthesised and can be secreted upon stimulation with carbachol.

Notes: In field desorption mass spectrometry it is in many cases more expedient to heat the field anode indirectly by i.r.-radiation instead of the usual method of electric heating to achieve higher relative molecular ion intensities.

Notes: The field desorption temperatures of some compounds are determined by indirect heating of the emitters. The temperatures thus obtained are lower, compared with those determined by other authors. The reasons for this deviation are discussed.

Notes: The mass spectra of some amino acids have been studied using the field desorption method. All amino acids yield molecular or quasi-molecular ions, even in the case of arginine and cystine, where these ions cannot be detected with the electron-impact (EI) or chemical ionization (CI) methods. The fragmentation is reduced as compared to EI, CI and FI.

Notes: A low resolution, high sensitivity study has been made of the field desorption mass spectrum of glucagon, a 29 amino acid residue underivatized peptide. The highest molecular weight fragments observed, when spectra were taken under conditions which minimized surface reactions of the peptide on the emitter, were found to contain no more than 18 amino acid residues. The most probable fragmentation processes for ions larger than m/e 600 were the production of fragments containing 10, 11 or 12 amino acid residues.

Notes: The acoustic emission technique is a non-destructive testing method used for inspection and surveillance of structures and pressurized components. In particular, the purpose of the measurement on the 5 MW research reactor SAPHIR was to investigate and evaluate the abnormally high noise levels during operation of the heat exchangers of the cooling system. The results showed that 5 heat exchangers out of the total of 9 (assembled in 3 groups) were responsible for the higher noise level. This is probably due to some abnormal vibration of the tube bundles therein, caused by the through-flow of the pool water. Indeed, attempts of localizing the source showed that there must be several, distributed sources.
