    Notizen: Abstract A new extremely thermophilic methane-producing bacterium was isolated from a submarine hydrothermal vent sample collected by a research team from the Woods Hole Oceanographic Institution using the manned submersible ALVIN. The sample was obtained from the base of a “white smoker” chimney on the East Pacific Rise at 20° 50′ N latitude and 109° 06′ W longitude at a depth of 2600 m. The isolate was a motile irregular coccus with an osmotically fragile cell wall and a complex flagellar system. In defined medium with 80% H2 and 20% CO2, the isolate had a doubling time of 26 min at 85° C. The pH range for growth was 5.2 to 7.0 with an optimum near 6.0. NaCl was required for growth with an optimum of 2 to 3% (w/v). The mol % G+C was 31%. In cell-free extracts, methane formation from methylcoenzyme M was temperature-dependent, and H2 or formate served as electron donors. Methane formation from H2 and CO2 occurred at a much lower rate. Oligonucleotide cataloging of the 16S ribosomal RNA established the isolate as a new species of the genus Methanococcus and the name Methanococcus jannaschii is proposed. The isolation of M. jannaschii from a submarine hydrothermal vent provides additional evidence for biogenic production of CH4 from these deep-sea environments.

    Notizen: Abstract Following growth in a chemically defined medium (CDM), five strains of Streptococcus uberis were tested for their ability to survive killing by bovine neutrophils. Strains 0140J, ST10, EF20 and C221 were easily killed, whereas strain C197C was highly resistant. The ability of strain 0140J to resist phagocytosis and killing was increased by supplementation of the growth medium with milk whey, casaminoacids, casein, or, to a lesser extent, bovine serum. Supplementation of the growth medium with yeast extract or bovine serum albumin did not affect the resistance of this strain. Following growth in CDM supplemented with casein, strains ST10 and C221, like strain 0140J, were significantly more resistant to killing by neutrophils. The resistance of strains EF20 and C197C was unaffected by the addition of casein to the medium; strain EF20 remained susceptible and strain C197C highly resistant to killing. The effect of supplementing the growth media with components other than casein was only studied for strain 0140J. Decapsulation of strains C197C, ST10 and 0140J, grown in CDM + casein, with type-X hyaluronidase did not significantly affect their ability to survive in the presence of bovine neutrophils.

    Notizen: Abstract Explant cultures of bovine mammary tissue taken from virgin heifers were used to examine adherence, colonization and cytopathogenesis ofStreptococcus uberis, Streptococcus agalactiae, Streptococcus dysgalactiae, Staphylococcus aureus andEscherichia coli in the putative target tissue. None of the five bacteria was able to adhere to healthy ductular epithelium but all showed a marked tropism for exposed connective tissue.S. aureus andE. coli induced a marked cytopathic effect in ductular epithelium after 6 hours in culture but the bacteria were not in close association with the affected tissue. No evidence could be found to support the hypothesis that adherence to epithelium might be the first stage in the pathogenesis of mastitis caused by these organisms.
