Notizen: Abstract 2,4-Dihydroxy-7-methoxy-1,4-benzoxazin-3-one (DIMBOA), a hydroxamic acid (Hx) occurring in wheat, was shown to deter feeding by the aphid Rhopalosiphum padi (L.), and to reduce BYDV transmission to the plant. Dual choice tests with wheat leaves showed the preferential settlement of aphids on leaves with lower levels of DIMBOA. Electric monitoring of aphid feeding behaviour showed that in seedlings with higher DIMBOA levels fewer aphids reached the phloem and they needed longer times to contact a phloem vessel than in those with lower levels. When aphids carrying BYDV were allowed to feed on wheat cultivars with different DIMBOA levels, fewer plants were infected with BYDV in the higher DIMBOA cultivars than in the lower ones. Preliminary field experiments showed a tendency for wheat cultivars with higher Hx levels to be more tolerant to infection by BYDV than lower Hx level ones.

Notizen: Abstract Volatiles from wheat and oat seedlings elicited attraction in apterae and alatae Rhopalosiphum padi. Cereal volatiles were identified by GC-MS and olfactometric tests were performed with each compound. Attraction of aphids was elicited by (E)-2-hexenyl acetate, (Z)-3-hexenol, (E)-2-hexenyl acetate, (E)-2-hexenol, (Z)-2-hexenol, n-heptanal, n-octanal, n-nonanal, n-decanal, benzaldehyde, and linalool. The difference between the sensory capacity of alatae and apterae is discussed in relation to migrations between hosts during their life cycle.

Notizen: Abstract DIMBOA glucoside (2-O-/gb-D-glucopyranosyl-4-hydroxy-7-meth-oxy-1,4-benzoxazin-3-one), the main hydroxamic acid (Hx) in intact wheat plants, was detected in the honey dew ofRhopalosiphum padi feeding on seedlings of six wheat cultivars that differed in their concentration of Hx, suggesting that the chemical circulates in the phloem. Neither the aglucone (DIMBOA) nor its main breakdown product were found in any of the honeydew samples. Honey dew production by aphids caged on seedlings of the wheat cultivars and DIMBOA glucoside concentrations in the honeydew followed biphasic curves when plotted against Hx concentration, suggesting passive ingestion of the chemical from the phloem at low Hx concentrations and limited ingestion due to feeding deterrency by Hx in mesophyll cells at high Hx concentrations. The presence of plant toxins such as Hx glucosides in the phloem sap, the main ingesta of aphids, and in the mesophyll cells, has major implications for plant defense, through a feeding deterrent effect during stylet penetration, and deterrency (antixenosis) along with antibiosis during feeding.

Notizen: Abstract GC-MS analysis of volatiles released from wheat infested with a high density of aphids showed the presence of 6-methyl-5-hepten-2-ol (sulcatol). The proportion of enantiomers present in the volatiles was determined by esterifying the mixture with (1S)-( − )-camphanic chloride and quantifying the esters. The mixture consisted of 75% (R)-( − ) and 25% (S)-( + ). The mixture of enantiomers as well as the racemate showed significant repellency towards apterous Rhopalosiphum padi in an olfactometer (15.7% and 14.4%, respectively, with 10 ng of stimulus). Single enantiomers or a mixture containing 25% (R)-( − )- and 75% (S)-( + )-enantiomers were inactive. The results are discussed in relation to the achievement of specificity by aphids in different pheromone-mediated behaviors.

Notizen: Abstract The influence of hydroxamic acids (Hx), plant secondary metabolites associated with aphid resistance in wheat, on the host acceptance and suitability of the aphid Sitobion avenae to the cereal aphid parasitoid Aphidius rhopalosiphi was evaluated. Aphids showed a reduction in mean relative growth rate and in body size in the wheat cultivar with higher Hx level. Reduction in aphid size was related to a decreased success in avoiding parasitoid oviposition. A minor increase in A. rhopalosiphi developmental time was observed in aphids feeding on the higher Hx cultivar. Experiments with different concentrations of DIMBOA glucoside, the main Hx in wheat, in artificial diets showed an increase in parasitoid developmental time at the highest concentration, with no change in other performance variables. The evidence is discussed in relation to the compatible utilization of host-plant resistance and biological control in integrated pest management.

Notizen: Abstract 2,4-Dihydroxy-7-methoxy-1,4-benzoxazin-3-one (DIMBOA), a hydroxamic acid involved in the resistance of cereals to aphids, was administered to adult individuals of the aphid Sitobion avenae in artificial diets. Effects on the cellular metabolism were inferred from the evaluation of several organelle marker enzymes. Catalase from peroxisomes and cytochrome c oxidase from mitochondria increased their activities about twofold when aphids were fed with 2 mM DIMBOA. The role of these enzymes in the metabolizing of xenobiotics by aphids is discussed. Biochemical and cytochemical evidences for the presence of peroxisomes in aphids are reported here for the first time.

Notizen: Abstract Phloem sap of wheat seedlings differing in whole leaf hydroxamic acid (Hx) concentrations was collected by cutting stylets of feeding aphids. DIMBOA-glucoside was the only Hx-related product found. Concentration of DIMBOA-glucoside in phloem sap showed a tendency to be negatively correlated with aphid performance.
