    Notizen: Abstract In experiments by Yoneyama et al. (1985) a hypertonic perfusing solution was used to record the c-wave from the isolated chick retina. We investigated whether the chick retina was surrounded by such hypertonic liquids. The following results were found: (1) the osmolarity of the subchoroidal humor was markedly higher than that of fluids from other areas of the eye; (2) the relative difference in osmolarity between both sides of the retina was 110–170 mOsmol in the light-adapted condition, whereas it was about 270 mOsmol in the dark-adapted condition; and (3) in the dark-adapted state the volume of the subchoroidal humor decreased about 40% but increased in osmolarity about 30–40%, whereas osmolarity of the subviteous humor decreased about 15–20 mOsmol. Results suggest the presence of an inward-directed pump mechanism for watersoluble components in the retina in addition to the proposed outward-directed pump mechanism of the pigment epithelium. The inward-directed pump may be more active in the dark-adapted state.

    Notizen: Abstract Retinal potentials were recorded from the eyes of anesthetized and immobilized chicks by a standard direct current method. The amplitude of the electroretinogram (ERG) c-wave was measured 2 and 5 sec after the onset of the light stimulus, as indexes of the fast-rise c-wave (cF-wave) and the slow-rise c-wave (cs-wave), respectively. An intravitreal injection of gamma-aminobutyric acid (GABA) at an estimated intravitreal concentration of 10−9-10−7M resulted in an increase of the amplitude of the cs-wave, a less remarkable change in the a- and cF-waves, and a slight decrease in the b-wave. The light peak of the retinal standing potential increased in amplitude following GABA administration (10−7-10−4M). Following an intravitreal injection of picrotoxin (10−5-10−3M), the polarity of the cs-wave changed from positive to negative and a significant decrease and deformation in the light peak was observed. The amplitude of the a-wave, however, increased in the range of the higher dose, while that of the b- and cf-waves decreased markedly but no polarity reversal of the cF-wave was found. The results may suggest that the GABA-ergic synapse plays a significant role in production of the cs-wave and the light peak, along with that of the pigment epithelium.

    Notizen: Abstract The relation between dopaminergic cells (and centrifugal fibers), the electroretinogram (ERG) c-wave, and the light peak were electrophysiologically investigated by observing the effects of a retrobulbar conduction block and intravitreal injection of either dopamine or haloperidol on these retinal responses. The retrobulbar conduction block (1% lidocaine) caused a decrease in the amplitude of the c-wave and the light peak in newly hatched chicks. Injections (2–20 μl) containing dopamine (0.1–10 mM) or haloperidol (1.3–13 mM) were given intravitreously while the responses were recorded. Although intravitreous injection of saline for control resulted in no observable change in the responses, dopamine selectively augmented the c-wave of ERGs and the light peak, but not the a-, b-, and d-waves. Haloperidol decreased first the light peak and later the c-wave. The augmentation of the retinal responses by dopamine and their reduction by haloperidol was statistically significant. The estimated threshold concentration of dopamine in the vitreous cavity was 1–3.5 μM. Since in many species the interplexiform cells have been found to contain dopamine, we hypothesize that the modulatory effects on the c-wave and the light peak in this preparation may be due to a centrifugal feed-back loop which includes the interplexiform cells to the horizontal and bipolar cells and the horizontal cells to the cones.

    Notizen: Summary Two methods for the direct separation of the enantiomers of trimetoquinol hydrochloride, denopamine and timepidium bromide were investigated, namely capillary zone electrophoresis (CZE) using cyclodextrins (CDs) (CD-CZE) and high-performance liquid chromatography (HPLC) using chiral stationary phases (CSPs). By CZE, separation of the enantiomers of trimetoquinol was successfully achieved when using β-cyclodextrin (β-CD), heptakis(2,6-di-0-methyl)-β-cyclodextrin (DM-β-CD) and β-CD polymer, but direct HPLC separation of the enantiomers was not successful. The enantiomers of denopamine were successfully separated by both CZE, employing DM-β-CD under acidic conditions, and HPLC, with cyclodextrin immobilized CSPs (CD-CSPs). Timepidium bromide, which is a racemic drug, was successfully enantioseparated by both CD-CZE and HPLC with CD-CSPs. The CD-CZE mode was selected as the optical purity testing method for trimetoquinol and denopamine because of the superiority of capillary electrophoretic (CE) techniques. Method validation results obtained clearly show that the CE method would be useful as an quality control method for assessing the optical purity of the drugs.

    Notizen: Summary The simultaneous stereospecific assay of four stereoisomers of diltiazem hydrochloride in bulk drug and aqueous solution was developed using HPLC on a Chiralcel OF column. The four isomers were quantitated with good precision by the internal standard method. The chiral inversion of (+)-cis-diltiazem hydrochloride in vitro, stability of its (2S, 3S) configuration in the solid and aqueous states was examined by HPLC. Chiral inversion of (+)-cis-diltiazem hydrochloride was not observed in the solid state, and its (2S, 3S) configuration was stable to heat, humidity and light. Chiral inversion of (+)-cis-diltiazem hydrochloride (2S, 3S) was observed in aqueous solution under UV, but not in aqueous solution stored at 80°C for 5h nor under visible light for 10 h. The (+)-cis-diltiazem hydrochloride (2S, 3S) epimerized to (+)-trans-diltiazem hydrochloride (2R, 3S) with a half-life of 5h in aqueous solution under UV but the reverse chiral inversion of (+)-trans-diltiazem hydrochloride (2R, 3S) to (+)-cis-diltiazem hydrochloride (2S, 3S) was not observed.

    Notizen: Summary Direct resolution of four stereoisomers of the related compound of diltiazem hydrochloride, namely desacetyl diltiazem hydrochloride, was studied by both normal and reversed-phase chiral HPLC. The four stereoisomers were completely resolved on a Chiralcel OF column. The technique developed was applied to a chiral inversion study of desacetyl diltiazem hydrochloride. This inversion was observed neither in the solid state, in aqueous solution at 100°C for 3 h nor under visible light for 10h, but was observed in aqueous solution under UV irradiation. The (+)-(2S, 3S)-cis-desacetyl diltiazem hydrochloride degraded with a half-life of 1.9 h in aqueous solution under UV and epimerized to (+)-(2R, 3S)-trans-desacetyl diltiazem hydrochloride. Similarly, (+)-(2S, 3S)-cis-desacetyl diltiazem hydrochloride degraded about three times faster than diltiazem hydrochloride. Reverse epimerization of (+)-(2R, 3S)-trans-desacetyl diltiazem hydrochloride to (+)-(2S, 3S)-cis-desacetyl diltiazem hydrochloride was not observed. The overall degradation was the result of two competitive processes, the epimerization and the decomposition of the benzothiazepin ring. The degradation and epimerization rate of (+)-(2S, 3S)-cis-desacetyl diltiazem hydrochloride in solution under UV depended upon the solvent, the aqueous pH, and concentration.

    Notizen: Summary A simple and sensitive method is proposed for the measurement of aldehydes in plasma. The plasma samples are treated with 1,3-cyclohexanedione (CHD) and after addition of the internal standard (the CHD derivative of octaldehyde) are analysed by reversed-phase high-performance liquid chromatography with an acetonitrile-water gradient as the mobile phase. Detection is by fluorimetry with excitation at 395 nm and emission at 457 nm. In plasma samples spiked with aldehydes, the calibration curves showed good linearity in the range 0–10 μg mL−1. The recoveries of aldehydes were quantitative; coefficients of within-day and between-day variation were less than 13% and 11%, respectively. Applications to human and some animal plasma samples are reported.
