    Notes: Abstract. Objective and Design: Recently, Ogawa et al. [17] reported that the peritoneal mast cells (PMCs) of rats can release histamine by substance P (SP) in a receptor-dependent manner. In the present study, we confirmed and extended their findings.¶Material: PMCs were isolated from six strains of rats. In some experiments, peritoneal cells in the non-MC fraction were used.¶Methods: PMCs were incubated with SP, neurokinin (NK) receptor agonists or antagonists, and histamine content in the supernatant was measured. In the binding assay, PMCs were incubated with [125I]BH-SP together with SP or NK receptor antagonists. NK-1 receptor mRNA was detected using a reverse transcription-polymerase chain reaction (RT-PCR) assay.¶Results: PMCs from Slc:Wistar and F344/NSlc were highly sensitive to SP, leading to histamine release, whereas those from Slc:SD and three other strains were not. PMCs from Slc:Wistar and F344/NSlc also released histamine in the presence of an NK-1 agonist. The histamine release induced by SP and the NK-1 agonist was inhibited by the NK-1 receptor antagonists, FK888 and CP-99,994. [125I]BH-SP binding experiments revealed that PMCs from Slc:Wistar rats possessed a single high affinity binding site for SP and that the binding was blocked by NK-1 receptor antagonists. Peritoneal cells in the non-MC fraction exhibited no appreciable binding. In the RT-PCR assay, expression of NK-1 receptor mRNA was evident in Slc:Wistar PMCs, but not in the non-MC fraction from Slc:Wistar or Slc:SD PMCs.¶Conclusion: These data demonstrate the existence of functional NK-1 receptors on freshly isolated PMCs in at least some strains of rats.

    Notes: Summary Twins with high-risk globoid cell leukodystrophy (GLD) at the 6th gestational month were studied morphologically and enzymatically. One of them was affected and the other was probably the carrier of this disease. Central nervous system and other viscera developed normally for the gestational age in both fetuses. The lesions of GLD were found in the fetus with marked deficiency of galactocerebrosidase, but not in the probable carrier. The lesions were located only in the considerably myelinated areas viz., the brain stem and the spinal cord. Ultrastructurally, the globoid cells in the fetal GLD seemed to originate from glial cells, probably oligodendroglia.

    Notes: Abstract Employing isoeletric focusing combined with enzyme-linked immunoelectrotransfer blot analysis, the fourth component of complement (C4) was analyzed in the two highly histocompatible, major histocompatibility complex homozygous groups (J and K) of Xenopus laevis. Each group had a characteristic C4 isoelectric focusing pattern, i. e., an isoelectric point range of 8.0–8.5 for J (C4 j C4 j ) and 7.6–8.1 for K (C4 k C4 k ). In (J x K)F1 frogs, C4 proteins were expressed in a codominant fashion (C4 j C4 k ). In the backcrossed progeny B1 [J × (J × K)F1], those with C4 j C4 j rejected (J × K)F1 skins hyperacutely (〈 17 days), were high responders against (J × K)F1 cells, and nonstimulators to J cells in mixed lymphocyte reaction (MLR), but they did not suffer from the graft-versus-host reaction (GVHR), even after the injection of (J x K)F1 cell-stimulated J splenocytes. On the other hand, the B1 frogs with C4 j C4 k acutely or chronically (〉 17 days) rejected (J × K)F1 skins, were low or nonresponders against (J × K)F1 cells and high stimulators to J cells in MLR, and they suffered from GVHR after the injection of prestimulated J splenocytes. These results argue for the notion that the genes equivalent to mammalian class III map to the MHC at the phylogenetic level of the anuran amphibian.

    Notes: Abstract. The objective of our study was to clarify the radiologic spectrum of disease entities belonging to the SAPHO syndrome (SAPHO being an acronym for synovitis, acne, pustulosis, hyperostosis, and osteitis). A retrospective analysis of radiologic data was undertaken to determine the relationship of the osteoarthritic changes seen in palmoplantar pustulosis (PPP, n = 179), acne (n = 3), psoriasis vulgaris (PsV, n = 355), generalized pustular psoriasis (GPP, n = 25), and chronic recurrent multifocal osteomyelitis (CRMO, n = 4). Osseous changes of PPP, acne, and CRMO overlap each other; 7 PPP, 2 acne, and 3 CRMO patients manifested stenocostoclavicular hyperostosis as well as hyperostosis of the spine, pelvis, and/or femur. These findings were not seen in either PsV or GPP patients. Thirteen PsV and 4 GPP patients had peripheral arthritis and/or symmetrical sacroiliitis, which were not observed in the PPP, acne, and CRMO patients. The PPP, acne, and CRMO patients may be grouped as belonging to the single disease entity, namely SAPHO syndrome. Our findings do not support the inclusion of PsV and GPP in the spectrum of this syndrome.

    Notes: Abstract A 10-month-old girl with mild developmental delay became hemiplegic after seizures. Cranial CT scan and magnetic resonance imaging (MRI) revealed no lesions related to vascular diseases, but brain atrophy on the right side was remarkable. Digital subtraction angiography showed slightly decreased visualization of peripheral branches of the right medial cerebral artery. Propionic acidaemia was diagnosed on the basis of high plasma levels of propionic acid and its metabolites and the elevated urinary excretion of these acids. With therapy, the levels of these acids fell, and her left hemiplegia disappeared 3 months later.

    Notes: Single-cell analysis is a very important field of research and is currently at the frontier ofphysical and biological sciences. Understanding how the phenotype of a single-cell arises from itsgenotype is a complex topic. Currently, the prevailing paradigm to analyze cellular functions is thestudy of biochemical interactions using fluorescence based imaging systems. However, theelimination of the labelling process is highly desirable to improve the accuracy of the analysis.Living cells are electromagnetic units; in as much they use electric mechanisms to control andregulate dynamic processes involved in inter alia signal transduction, metabolism, proliferation anddifferentiation. Recent developments in micro- and nanofabrication technologies are offering greatopportunities for the analysis of single cells; the combination of micro fluidic environments, nanoelectrodes/wires and ultra wide band electromagnetic engineering will soon make possible theinvestigation of local (submicrometer scale) dynamic processes integrating several events atdifferent time scales. In the paper, we present recent approaches which aim at investigating singlecellswith the help of MEMS and NEMS (Micro and Nano Electro Mechanical Systems) and ultrawide band (DC-THz) electromagnetic characterization techniques
