Heme oxygenase (HO)-1 catalyzes the rate-limiting step in heme degradation. The aim of the study was to determine HO-1 expression and distribution pattern in glial cells during differentiation and after exposure to glutamate (GLU) (100 μm). In untreated cells at 7 days in vitro (DIV) HO-1 expression was undetectable whereas increased at 14 and 21 DIV. Exposure to GLU for 24 h at 7, 14 and 21 DIV increased the expression of HO-1; this effect was prevented by preincubation with GYKI 52466. In untreated cells at 7 DIV, HO-1 fluorescence was weak; the protein appeared in the cytosol and the fluorescence intensity increased at 14 and 21 DIV. Compared with untreated cells, the addition of GLU increased the fluorescence and the enzyme translocated in the nucleus and nucleoli. These results demonstrate the presence of HO-1 in the glial cells and the translocation in the nuclear compartment of astrocytes during differentiation.
