    Notizen: Efficient methods for using triple and fourfold γ-ray coincidence data to give information on spins and parities of levels in secondary fission fragments are described. Triple-γ angular correlations and double-γ angular correlations culled from triple coincidences with one γ ray observed isotropically for selectivity have been used to give information on the multipole nature of transitions; directional linear polarization data culled from fourfold coincidences with one isotropically observed γ ray have been used to provide parity information. Results for known cascades agree with predicted correlation and polarization patterns. Examples of the techniques applied to transitions in 106Mo produced in the decay of 248Cm are given. © 1998 American Institute of Physics.

    Notizen: Previous Richtmyer–Meshkov instability experiments carried out in shock tubes have been hampered by the need to separate the two gases with a thin plastic membrane. As a result, many of these experiments have had poor agreement with the linear stability theory of Richtmyer [Commun. Pure Appl. Math. 23, 297 (1960)]. This limitation has been removed in the present investigation by the use of a novel technique in which the interface is formed by flowing light (N2) and heavy (SF6) gases from opposite ends of a vertical shock tube. Both gases exit the shock tube through slots in the test section walls leaving behind a flat motionless interface which is then given a sinusoidal initial shape by gently oscillating the shock tube at a prescribed frequency in the horizontal direction. A weak shock wave (Ms=1.10), generated in the shock tube, impacts the interface and produces the instability. Photographs of the interface, which is visualized by seeding the heavy gas with a water droplet fog and illuminating it with a strobe light source, provide particularly clear views of the developing instability far into the nonlinear regime. In addition, amplitude measurements obtained from these photographs are found to be in good agreement with Richtmyer's theory. © 1997 American Institute of Physics.

    Notizen: The ATP-binding cassette (ABC) transporter superfamily is one of the most widespread of all gene families and currently has in excess of 1100 members in organisms ranging from the Archaea to man. The movement of the diverse solutes of ABC transporters has been accepted as being strictly unidirectional, with recent models indicating that they are irreversible. However, contrary to this paradigm, we show that three solute-binding protein-dependent (SBP) ABC transporters of amino acids, i.e. the general amino acid permease (Aap) and the branched-chain amino acid permease (Bra) of Rhizobium leguminosarum and the histidine permease (His) of Salmonella typhimurium, are bidirectional, being responsible for efflux in addition to the uptake of solutes. The net solute movement measured for an ABC transporter depends on the rates of uptake and efflux, which are independent; a plateau is reached when both are saturated. SBP ABC transporters promote active uptake because, although the Vmax values for uptake and efflux are not significantly different, there is a 103−104 higher affinity for uptake of solute compared with efflux. Therefore, the SBP ABC transporters are able to support a substantial concentration gradient and provide a net uptake of solutes into bacterial cells.

    Notizen: Abstract We generate second-sound resonances in a cylindrical cavity in3He-4He mixtures using vibrating Nuclepore filters. The properties of the cavity resonances are well described by the theory proposed by d'Humieres et al. The theory predicts the strength of cavity resonances as well as the phase shift between in- and out-of-phase components of the signal. By fitting these quantities we are able to get information about the compliance of the membrane, the distance of the membrane to the backplate, and the resistance to flow through the membrane. This enables us to predict the reflection coefficient of second-sound waves off the transducer. We also study the effect of a strong magentic field on the resonances.

    Notizen: Day-6 rabbit blastocysts were able to accumulate [3H]quinuclidinyl benzilate (QNB) from their environment. This accumulation was reduced approximately 50% in the presence of 1.5 × 10-4 M atropine (an accepted antagonist for ligands which bind to muscarinic cholinergic receptors). The accumulation of QNB was sensitive to temperature and was apparently saturable. In the presence of 2 nM QNB, Day-6 blastocysts accumulated 30.3 ± 2.0 fmoles per blastocyst. When the cellular elements alone were examined, lesser amounts of specific binding were detected. Owing to the complexity of this multicompartmental system, Scatchard analysis did not provide meaningful results. This accumulation appears higher than that reported for other tissues such as rabbit heart homogenates or rabbit uterine endometrial cells. This muscarinic cholinergic accumulation may have some role in blastocyst-maternal recognition.

    Notizen: Platelet-activating factor (1-O-alkyl-2-acetyl-sn-glycero-3-phosphorylcholine; PAF) is a very potent phospholipid, which has been demonstrated to stimulate smooth muscle and change vascular permeability. PAF has been detected in the rabbit preimplantation uterine endometrium and has been demonstrated to bind specifically to rabbit uterine membranes. To evaluate the possible role of PAF in maternal-embryonic chemical communication, we report here that rabbit blastocysts can accumulate [3H]PAF from their environment. Blastocysts were able to accumulate [3H]PAF as time-, buffer-, age-, and concentration-dependent functions. The accumulation was inhibited by some PAF receptor antagonists, such as U66985, as well as by unlabeled PAF and lyso-PAF, indicating that the accumulation process may be receptor mediated. The data support the current model of PAF as a paracrine factor in preimplantation stages of reproduction.
