    Notizen: Polymer solution dynamics may be inferred from light scattering spectra of dissolved optical probe particles. We compare a variety of probes in solutions of several polymers. In the "overlapping'' concentration/molecular weight regime, the Stokes–Einstein equation fails by up to a factor of 2, while the probe diffusion coefficient D follows a scaling law D/D0=exp(−aMγcνRδ) (c, M, and R are the polymer concentration, molecular weight, and the probe radius, respectively). Experimentally, γ=0.8±0.1, ν=0.6–1.0, and δ=−0.1 to 0, contrary to the theoretical predictions γ=0 and δ=1. With very high molecular-weight polymers, we observe a further "entangled'' regime, characterized by huge (104) failures of the Stokes–Einstein equation and the appearance of "fast'' modes in the scattering spectrum.

    Notizen: The retrovirus integrase (IN) protein is essentialfor integration of viral DNA into host DNA. The secondary structure of thepurified IN protein from avian myeloblastosis virus was investigated by bothcircular dichroism (CD) spectroscopy and five empirical prediction methods. The secondary structures determined from the resolving of CD spectra through a least-squares curve fitting procedure were compared with those predicted from four statistical methods, e.g., the Chou-Fasman, arnier-Osguthorpe-Robson, Nishikawa-Ooi, and a JOINT scheme which combined all three of these methods, plus a pure a priori one, the Ptitsyn-Finkelstein method. Among all of the methods used, the Nishikawa-Ooiprediction gave the closest match in the composition of secondary structureto the CD result, although the other methods each correctly predictedoneor more secondary structural group. Most of the α-helix and β-sheet states predicted by the Ptitsyn-Finkelstein methodwere in accord with the Nishikawa-Ooi method. Secondary structural predictions by the Nishikawa-Ooi method were extended further toinclude IN proteins from four phylogenetic distinct retroviruses. The structuralrelationships between the four most conserved amino acid blocks of these IN proteins were compared using sequence homology and secondary structure predictions.

    Notizen: Quasi-elastic light scattering (QELS) spectroscopy was for the first time used to measure the mutual diffusion coefficient Dm of titanium dioxide (TiO2) particles of 0,37 μm and 0,31 μm diameters in a poly(ethylene oxide) melt (weight-average mol. wt. 7 500) between 85 °C and 160 °C. Dm exhibits a strong dependence on the temperature. The effective shear imposed by diffusing particles on the polymer melt was estimated through a comparison of the viscosity obtained by a cone-and-plate viscometer with the microviscosity calculated from the diffusion coefficients and hydrodynamic radii of the particles through the Stokes-Einstein equation. The rate of shear found was ≥ 104 · s-1.
