    Notizen: The high resolution infrared spectrum of the CH stretching fundamental of the formyl radical (HCO) has been observed by means of infrared kinetic spectroscopy using 308 nm (XeCl) excimer laser flash photolysis of formaldehyde or acetaldehyde followed by diode or difference frequency laser probing of the transient absorption. The high resolution spectra obtained were assigned and fitted with rotational, spin–rotational, and centrifugal distortion constants. The ν1 band origin is 2434.48 cm−1. New ground-state constants are reported from a least-squares fit combining this ν1 data with previous microwave and FIR LMR measurements.

    Notizen: Optical properties of Eu3+ ions in oxyfluoride glass ceramics and in the precursor glasses (30 SiO2, 15 Al2O3, 29 CdF2, 22 PbF2, 1.5 YF3, and 2.5 EuF3, in mol %) have been analyzed and compared. The Eu3+ ions in the glass ceramics are incorporated into a crystalline environment with higher symmetry and lower energy coupled phonons. Emission measurements indicate that this crystalline phase is EuF3, instead of PbxCd1−xF2 as generally considered. © 2001 American Institute of Physics.

    Notizen: The kinetics of forward and backward energy transfer processes in fluoroindate glasses double doped with Yb3+ and Er3+ has been studied. It was possible to excite selectively Yb3+ ions and detect emission due only to these ions or combined with the emission coming from the Er3+ ions. At low concentration of Yb3+ (0.1 mol %) the emission decay of these ions is nonexponential when there is back transfer from Er3+ ions; the dynamics is well described using the fluorescence "transfer function" model. Moreover, the evolution of the emission from the 4I11/2 (Er3+) level is in good agreement with the behavior predicted by the model. The critical radii obtained from the fitting for forward and backward energy transfer are compared with those calculated with the Dexter formula. When the Yb3+ concentration is increased, the migration among these ions is very important and the emission decay of the Yb3+ ions is nearly exponential with the lifetime of these ions. This behavior is explained considering that, at high Yb3+ concentration and due to the migration, the transfer from Yb3+ to Er3+ ions is restricted to very closed couples of ions with high back transfer probability. The limitations of the fluorescence "transfer function" model associated with the correlation effect in the excitation of Er3+ ions by energy transfer are discussed. © 1999 American Institute of Physics.

    Notizen: Optical properties of Yb3+–Tb3+ codoped silica sol-gel samples have been studied after the gel to glass transition. Different upconversion emissions have been observed under near infrared excitation at about 1 μm. The Tb3+ ions are excited by means of energy transfer processes from Yb3+ ions. The temporal evolution of the blue-green upconversion emissions coming from Tb3+ ions and their dependence on the excitation intensity at about 1 μm has been studied. The experimental results are in good agreement with a cooperative resonant energy transfer mechanism from Yb3+ ions. An efficient backtransfer process is observed from Tb3+ ions towards Yb3+ ions. The upconversion efficiency, which is limited by this backtransfer process, has been obtained and compared with other upconversion results in similar matrix. © 2001 American Institute of Physics.

    Notizen: Abstract: In the transmitter-gated ion channel class of receptors, the members of which are all believed to be heterooligomers, the number and arrangement of the subunits are only known with any certainty for the nicotinic acetylcholine receptor from Torpedo electric fish. That receptor has been shown to possess a pentameric rosette structure, with five homologous subunits (α2βγδ) arranged to enclose the central ion channel. The data were obtained by electron image analysis of two-dimensional receptor arrays, which form as a consequence of that receptor's exceptionally high abundance in the Torpedo membranes and are therefore not attainable for other receptors. We have applied another direct approach to determine the quaternary structure of native ionotropic GABA receptors. We have purified those receptors from porcine brain cortex and analysed the rotational symmetry of isolated receptors visualized by electron microscopy. The results show the receptor to have a pentameric structure with a central water-filled pore, which can now be said to be characteristic of the entire superfamily.

    Notizen: Abstract: Polyclonal antibodies have been raised in rabbits against the glycine receptor antagonist strychnine, coupled through a 2-amino substituent to the antigenic protein keyhole limpet haemocyanin. Strychnine binding of the predominantly immunoglobulin G (IgG) class of antibodies was measured by incubation with [3H]strychnine, followed by adsorption of IgG onto Staphylococcus aureus cells and filtration through glass-fibre filters under vacuum. Only strychnine and structurally related alkaloids or derivatives were able to inhibit [3H]strychnine binding to the IgG. A significant rank correlation was found between the potencies of these compounds to inhibit [3H]strychnine binding to the antibodies and to the glycine receptor in mouse spinal cord membranes. In contrast, preincubation of strychnine antibodies with a variety of ligands at other neurotransmitter, drug, or hormone receptors in the CNS (at 10−4M) failed to inhibit binding significantly. The failure of glycine to inhibit strychnine antibody binding is consistent with previous suggestions that the recognition sites for this amino acid on the CNS receptor may be conformationally distinct from those for the antagonist alkaloid. Strychnine antibodies may now help in the identification and purification of possible endogenous ligands at this alkaloid binding site in the CNS.

    Notizen: Abstract: The GABA and benzodiazepine binding sites on GABAA receptors are allosterically coupled. The in vitro binding of 2 nM [3H]flunitrazepam to cortical and cerebellar membranes prepared from drug-naive rats was potentiated ∼1.6-fold by 100 γM GABA. Potentiation in both regions was significantly reduced 4 or 12 but not 24 h after a single dose of 15 mg/kg diazepam. At 24 h after the last of 14 daily doses of diazepam, no differences in GABA poten-tiation were observed. Diazepam-induced changes in GABAA receptor δ2-subunit gene transcription and α1-, β2-, and δ2-subunit steady-state mRNA levels did not appear to be temporally related to allosteric uncoupling.
