    Notizen: Foot-and-mouth disease virus was examined for its stability during cooking in tissues from infected cattle. The 01 (CANEFA 2) serotype of foot-and-mouth disease virus survived in lymph node tissues after heating for 2 hr at 69°C, for 1 hr but not for 2 hr at 82°C, and for 15 min but not for 0.5 hr at 90°C. Incorporation of 1% NaCl into suspensions of infected lymph nodes enhanced viral survival after heating for 0.5 hr but not for 1 hr at 90°C. The virus did not survive in either ground beef or meatballs contaminated with infected lymph node tissue, when processed to internal temperatures of 93.3, 96.1 or 98.8°C using commercial thermal processing procedures. Accurate temperature measurements were achieved with a temperature sensitive indicator disc developed in this study.

    Notizen: Summary An enduring level of antibody was produced in swine for 90 days by the inoculation of a trivalent vaccine combined with oil adjuvant. The antigenicity of the virus type determined, to considerable degree, the antibody level attained. This relatively simple method of protecting swine from foot-and-mouth disease appears to have a practical application in future field studies.

    Notizen: Summary Serums of swine inoculated with chemically-treated foot-and-mouth disease virus (FMDV) combined with an oil adjuvant had an enduring level of antibody and the swine were resistant to challenge by exposure to FMD-infected swine for at least 90 days. The antibody level was significantly higher in the animals inoculated with the treated virus and oil adjuvant than in the animals inoculated with the treated virus and aluminium hydroxide adjuvant as indicated by analysis of variance of mouse protective dose (PD50) values. Swine recovered from experimental infection with FMDV may become reinfected when exposed to homologous virus.

    Notizen: Summary Mice treated with divinyl ether-maleic anhydride (DVE/MA) 7 days before, 2 days before, or simultaneously with foot-and-mouth disease (FMD) viral antigens (aqueous or oil emulsified) were more resistant to virus infection and released antibody earlier than mice treated with antigen alone. Simultaneous treatment of swine with these antigens plus DVE/MA resulted in a slightly enhanced early antibody response, which, in a test with oil-emulsified antigen, was not associated with an early induced antiviral resistance to FMD. In general, the primary and secondary antibody responses to oil antigen plus DVE/MA were the same as in control swine receiving the antigen alone. However, swine sensitized with aqueous antigen plus DVE/MA responded faster and developed higher antibody titers after a secondary treatment with aqueous antigen than swine treated similarly but without DVE/MA.

    Notizen: Summary The duration of immunity was studied for a 365-day period in swine vaccinated with a foot-and-mouth disease trivalent vaccine, inactivated for 48 hours with acetylethyleneimine and combined with an oil adjuvant. A satisfactory degree of immunity was afforded at 90 and 180 days postinoculation (DPI). At 365 DPI, vaccinated swine still maintained a degree of resistance to infection despite marked lowering of neutralizing antibody. At 6 months, lesions at inoculation sites were of very limited extent and appeared as scattered islands of small yellowish streaks of fibrous tissue and some spherical nodules. Histologically, lesions were multiple, foreign-body-type granulomas characterized by oil vacuoles, reticuloendothelial cell proliferation and fibrosis. Tissues surrounding lesions were normal. At 12 months, these lesions were almost impossible to locate. Microscopically, they were comparable to those observed at 6 months. Whether these lesions are more serious than those obtained with current products and other adjuvants and whether these constitute a carcass blemish remain to be determined.

    Notizen: Summary The pattern of foot-and-mouth disease antibody formation in swine was similar to that produced in cattle and guinea pigs. In the early stages the antibody appeared to be a macroglobulin of the 19 S-class and the later-appearing antibody a γ-globulin of the 7 S-class. Evidence suggestive of another class of 7S antibody with an electrophoretic mobility faster than the γ2-globulin is presented. The differences in antibodies produced by the various inoculums appeared to be quantitative only. There appeared to be a correlation of the class and level of antibody produced and the degree of resistance to infection with a homologous virus.

Steers inoculated with the treated virus and aluminium hydroxide were not protected. Although minimal numbers of animals were used, the results parallel those obtained in swine in experiments where larger numbers of animals were used. Experiments with larger numbers of steers are needed to confirm these findings.

    Notizen: Summary Local reactions at inoculation sites of swine vaccinated with foot-and-mouth disease antigens emulsified with oil adjuvant were examined at 7, 14, 32, 45, 62 and 90 days postinoculation. The serologic data indicate that either the subcutaneous or intramuscular route, as well as the different inoculation sites selected induce a similar antibody response. Lesions at all inoculation sites were characterized by subacute and chronic granulomatous tissue reaction and the continuous presence of oil vacuoles. There is little doubt that some degree of reaction at the site of inoculation is to be expected and thus the site and route are of utmost importance in reducing the possibility of a carcass blemish.
