    Notizen: Thermal trauma causes tissue damage by membrane destabilization and energy depletion at the cellular level, resulting in tissue necrosis and inflammation leading to delayed cell death. One therapeutic approach is to block the immediate triggering of the inflammatory cascade that results in prolonged hypermetabolic responses and immune dysfunction while promoting the expression of growth factors. In the present study, we determined hepatic gene expression responses to insulin-like growth factors-i (IGF-I) gene transfer to burned rats using high-density DNA microarray assays. The expression of 123 out of ~8,800 genes assyed (1.4% of total) were significantly altered. Of these, 42 genes were altered irrespective of treatment by burn trauma (p 〈 0.05). Changes in gene expression were confirmed by measuring mRNA levels using reverse transcription-polymerase chain reaction and protein levels by Western blot assays. DNA microarray analyses showed two major mechanisms that mediated beneficial outcomes after IGF-I gene transfer in the burned rat livers. These mechanisms were the stimulation of IGF binding protein potentiation of peripheral IGF-I and the inhibition of the burn-augmented pro-apoptotic and oxidative mitochondrial metabolites stimulated by thermal trauma.

    Notizen: Severe burns cause not only skin injury but several marked systemic derangements. During wound healing, matrix metalloproteinases (MMPs) and the tissue inhibitors of metalloproteinases play an important role in tissue regeneration and remodeling processes. Therefore, in the present study, we determined the serum levels of MMPs and tissue inhibitor of metalloproteinase-1 in burn patients over time. Serum samples from 12 severely burned children (mean age 7.9 ± 2.5 years) with 〉40% total body surface area burns were obtained within 0.5 hours, 3, 7, and 21 days after injury. Pro-MMP-1, MMP-3, MMP-9, and tissue inhibitor of metalloproteinase-1 serum levels were assayed by enzyme-linked immunoassay and compared to normal healthy volunteers. Two-way analysis of variance and Bonferroni's test were used for statistical analysis. Pro-MMP-1 levels in the serum were significantly elevated by the seventh day after burn. MMP-3 and MMP-9 levels showed significant increases by day 3 and 21 compared to normals, respectively. Tissue inhibitor of metalloproteinase-1 levels did not change with time after burn but were significantly higher by 3 days after burn compared to normal serum. In conclusion, changes in MMPs and tissue inhibitor of metalloproteinase-1 occur in burn patients and those changes may be a mechanism beneficial to wound healing. (WOUND REP REG 2003;11:177–180)

    Notizen: SYNOPSIS. Tetratrichomonas gallinarum was found in the blood of 3 chickens experimentally infected with Borrelia anserina. The blood infection was successfully reproduced in healthy chickens and was accompanied by evidence that the trichomonads multiplied in the blood, ingested red blood cells, and were themselves phagocytized by leucocytes. T. gallinarum was maintained in serial passage in chicken embryos as well as in the usual trichomonad culture media.

    Notizen: Thermal stresses that develop in aluminum interconnects upon cooling from an annealing temperature are investigated using crystal plasticity theory. The crystal plasticity model is implemented assuming either isotropic hardening or a physically based hardening model developed for single crystals. The dependence of the predicted thermal stresses on crystal orientation and on interconnect aspect ratio is examined. The results are compared with recent observations of electromigration-induced transgranular voids to ascertain the contribution of the thermal stresses in the failure of aluminum interconnects. The analysis clearly demonstrates the necessity of accounting for all slip systems when modeling the effects of texture in single-crystal thin films. In addition, the calculations suggest that proper consideration of the hardening description is necessary for accurate predictions of thermal stresses. © 1995 American Institute of Physics.

    Notizen: One of the major issues in the development of large, rule-based expert systems is related to improving their performance efficiency. One way to address this issue is by reducing the number of unsuccessful tries a system goes through before executing a rule to establish a goal or an intermediary fact. On the average, the number of unsuccessful tries can be reduced if the rules that are tried first are those that are expected to execute most frequently, and this can be established by extracting information on the probability distributions of the input parameters. In this paper, a rule base is modeled as a network and simulated to investigate potential performance improvements by changing the order used to test the rules. The model of the rule base is also used to investigate performance gains achieved by parameter factorization and premise clause reordering.

    Notizen: The INO2 gene of Saccharomyces cerevisiae is required for expression of most of the phospholipid biosynthetic genes. INO2 expression is regulated by a complex cascade that includes autoregulation, Opi1p-mediated repression and Ume6p-mediated activation. To screen for mutants with altered INO2 expression directly, we constructed an INO2-HIS3 reporter that provides a plate assay for INO2 promoter activity. This reporter was used to isolate mutants (dim1) that fail to repress expression of the INO2 gene in an otherwise wild-type strain. The dim1 mutants contain mutations in the OPI1 gene. To define further the mechanism for Ume6p regulation of INO2 expression, we isolated suppressors (rum1, 2, 3) of the ume6Δ mutation that overexpress the INO2-HIS3 gene. Two of the rum mutant groups contain mutations in the OPI1 and SIN3 genes showing that opi1 and sin3 mutations are epistatic to the ume6Δ mutation. These results are surprising given that Ume6p, Sin3p and Rpd3p are known to form a complex that represses the expression of a diverse set of yeast genes. This prompted us to examine the effect of sin3Δ and rpd3Δ mutants on INO2-cat expression. Surprisingly, the sin3Δ allele overexpressed INO2-cat, whereas the rpd3Δ mutant had no effect. We also show that the UME6 gene does not affect the expression of an OPI1-cat reporter. This suggests that Ume6p does not regulate INO2 expression indirectly by regulating OPI1 expression.

Unfortunately, past articulations of the theory have adopted a static orientation, with the result that organizational learning - a dynamic process - has been excluded from the discussion. Presents a model designed to incorporate organizational learning effects into the resource-based view. Then applies the model to a number of case examples to demonstrate the importance of including organizational learning as a component of resource-based theory. Concludes that organizational learning is itself a strategic capability or resource important to the process of building and maintaining competitive advantage.
