    Notizen: Metallic ferrofluids based on magnetic particles in mercury have been produced by two new methods. Alloy particles of Fe-B, Fe-Co-B, Fe-Ni-B, and Co-B were prepared by reduction of the transition metal ions in aqueous solutions by NaBH4 and subsequently suspended in mercury. In one preparation, the metallic mercury was added to the solution with precipitates and in another method produced simultaneously with the precipitates by reduction of mercury ions to metallic mercury. The samples were characterized by magnetization and Mössbauer spectroscopy measurements.

Species of Tetrahymena, including T. vorax, T. thermophila, T. pyriformis, and T. pigmentosa, were tested for cloning efficiency in proteose peptone and in synthetic nutrient media to which were added hemin, protoporphyrin IX, chlorophyllin, or asolectin, an impure mixture of phospholipids. All species could be cloned with high efficiency in the crude media. In unsupplemented synthetic medium the cloning efficiencies were 0–10%, around 50%, around 50%, and 90–100% for T. thermophila, T. vorax, T. pyriformis, and T. pigmentosa, respectively. The first three were all stimulated to 90–100% by addition of the porphyrin or phospholipid compounds mentioned above. Uroporphyrin III and coproporphyrin I and III had no effect. We suggest that cells unable to form clones suffer from a lack of cellular energy. This situation may be alleviated by our additions: certain porphyrin rings may be built into cytochromes and phospholipids may be used as fuel. Thus, the synthetic media used so far for these ciliates have not been optimal.

    Notizen: This report contains the newest directions for preparation of the synthetic nutrient medium for some Tetrahymena species that do not require lipids. In the standard medium T. thermophila. strains SB 210 and 281, multiply at 37°C with doubling times of around 2 h and at 26°C around 5 h. We have established multiplication rates as functions of variations in the composition of the medium. In media in which all components are present at one-third of the normal concentrations and only the essential amino acids are included, growth and multiplication become sharply dependent on glucose in strain SB 281. Such media may be used for selection and enrichment of certain specified cell lines.

    Notizen: Using a hemolytic assay system, we have detected cytolytic activities in extracellular medium from Tetrahymena thermophila and T. pyriformis. In addition, we have identified two phospholipase activities (types A and C) from the same source by thin layer chromatography analysis of the breakdown products of a phospholipid preparation. The hemolytic activity peaks at low pH values. It is inhibited by egg lecithin, supporting the view that a phospholipase activity is involved in the cytolytic processes. Cytolytic activities may play important roles in Tetrahymena, both in nutrition, especially in parasitic and scavenger forms, and in defense against predators. Tetrahymena is probably partly protected from its own released cytolytic phospholipase by having a high proportion of phosphonolipids on its surface membrane.

    Notizen: Fluorescein-labelled plastic microbeads, with or without covalently attached β-l, 3-D-glucan, were injected into the peritoneal cavity of mice. Peritoneal cells were subsequently analysed by flow cytometry according to fluorescence and light scatter and separated into fluorescence positive and negative cells. We report that cells from animals treated with glucan-plastic beads produced large amounts of prostaglandin E2 (PGE2) whether the cells actually contained heads or not. On the other hand, cells from animals treated with glucan-plastic beads produced less thymocyte-stimulatory activity—presumably corresponding to interleukin 1 (IL-l)—than cells from control animals treated with commercial latex beads. However, when indomethacin was added, either in vivo or in vitro, cells from animals treated with glucan-plastic beads produced more thymocyte-stimulalory activity than controls. We interpret this to mean that glucan-plastic beads stimulate both IL-l and PGE2, but that under circumstances where the cellular cyclo-oxygenase is not inhibited, the PGE2 will block IL-1 production.

    Notizen: Macrophages obtained from animals treated withβ-1.3-D-glucan-derivatized plastic beads were greatly stimulated, as judged by morphology, esterase release, and cytostatic effect on L-929 tumour cells in vitro. The pretreatment of mice with such heads conferred an apparent absolute local resistance to an otherwise lethal pneumococcal infection but had no effect on the growth of intrapcritoneal AA ascites sarcoma. Moreover, peritoneal cells from animals pretreated with glucan beads did nut protect the animals in a Winn assay.

    Notizen: Decimation of cereal growth and yields by hemiparasitic Striga species cannot be accounted for entirely by the removal of host-plant resources. The production of toxic compounds by the parasite has been suggested. An investigation of three species of the economically important Striga and the related Alectra vogelii has now resulted in the isolation of eight iridoid glucosides (mussaenosidic acid, mussaenoside, gardoside methyl ester, bartsioside, isoaucubin, melittoside, aucubin and eurostoside), two caffeoyl phenylethyl glycosides (calceolarioside  A and verbascoside) as well as shikimic acid and trigonelline, all identified by NMR spectroscopy. The iridoids are potent cytotoxins and probably represent an anti-herbivore defence system common to Scrophulariaceae (sensu lato). This has the potential to explain differences in tolerance apparent for contrasting host taxa and cultivars. The nature of the iridoids present also provides additional validation of the recent transferral of parasitic Scrophulariaceae (s.l.) to Orobanchaceae.

    Notizen: A survey has been carried out in all Danish anaesthetic departments (n = 80) regarding the attitude towards the use of epidural/spinal lumbar analgesia in patients who were receiving prophylactic anticoagulant therapy for the prevention of thromboembolism. About 60% of the departments used the techniques in patients receiving low-dose heparin and no side effects had been experienced. Spinal and epidural anaesthesia were in general regarded as being contraindicated in patients fully anticoagulated with vitamin K antagonists. In the world literature, the attitude towards the combination is conflicting. No randomised trial has been performed and complications are almost entirely confined to patients fully anticoagulated with vitamin K antagonists. Only one case of an epidural haematoma has been recorded when subcutaneous low-dose heparin was used as thromboprophylaxis.

    Notizen: The aim of the study was to investigate a possible effect of omeprazole on the characteristics of the gastric emptying of liquid and solid in healthy subjects. The study was performed as a double-blind crossover study and the gastric emptying studies were performed after 10 days of treatment with placebo or omeprazole 40 mg o.m. Omeprazole was without effect on the characteristics of liquid emptying or the lag phase of solid. It does, however, decrease the emptying rate of solid as the omeprazole group had a median half-time duration of the linear emptying period which had twice the duration of the corresponding figure in the placebo group.

    Notizen: SYNOPSIS. Rates of removal of suspended India ink particles from the surrounding medium by 2 ciliates, Tetrahymena pyriformis strain GL and Tetrahymena vorax strain V2S, have been measured. It is evident from the results that the food vacuoles concentrate the suspended particles, clearing a volume of the surrounding suspension fluid 500 × greater than the total volume of food vacuoles made during the same period of time.
