Notizen: Summary Glutathione functions to scavenge oxidants or xenobiotics by covalently binding them and transporting the resulting metabolites through an adenosine 5′-triphosphate-dependent transport system. It has been reported that the intracellular concentration of glutathione decreases in diabetes mellitus. In order to elucidate the physiological significance and the regulation of anti-oxidants in diabetic patients, changes in the activity of the glutathione-synthesizing enzyme, γ-glutamylcysteine synthetase, and transport of thiol [S-(2,4-dinitrophenyl)gluta- thione] were studied in erythrocytes from patients with non-insulin-dependent diabetes and K562 cells cultured with 27 mmol/l glucose for 7 days. The activity of γ-glutamylcysteine synthetase, the concentration of glutathione, and the thiol transport were 77 %, 77 % and 69 %, respectively in erythrocytes from diabetic patients compared to normal control subjects. Treatment of patients with an antidiabetic agent for 6 months resulted in the restoration of γ-glutamylcysteine synthetase activity, the concentration of glutathione, and the thiol transport. A similar impairment of glutathione metabolism was observed in K562 cells with high glucose levels. The cytotoxicity by a xenobiotic (1-chloro-2,4-dinitrobenzene) was higher in K562 cells with high glucose than in con- trol subjects (50 % of inhibitory concentration; 300 ± 24 μmol/l vs 840 ± 29 μmol/l, p 〈 0.01). Expression of γ-glutamylcysteine synthetase protein was augmented in K562 cells with high glucose, while enzymatic activity and expression of mRNA were lower than those in the control subjects. These results suggest that inactivation of glutathione synthesis and thiol transport in diabetic patients increases the sensitivity of the cells to oxidative stresses, and these changes may lead to the development of some complications in diabetes mellitus. Notizen: Abstract. Background: Because of its poor prognosis, the diagnosis of hepatocellular carcinoma with sarcomatous change (HCCSC) is clinically important. The purpose of this study is to elucidate the characteristic CT findings of HCCSC. Methods: Two-phased dynamic incremental CT images of six histologically proven HCCSC were retrospectively reviewed. Results: All tumors (100%) exhibited peripheral enhancement on delayed CT images. Lymphadenopathy was observed in 100% (six of six patients); intrahepatic metastases, in 83% (five of six). Both metastatic lesions showed findings similar to those of the primary hepatic tumors, such as peripheral enhancement. Histopathologically, delayed and/or prolonged peripherally enhanced areas consisted of viable cancer cells with sarcomatous changes. Conclusions: The appearance of HCCSC on CT is that of an irregularly demarcated intrahepatic mass with delayed or prolonged peripheral enhancement, frequently with intrahepatic metastases and lymphadenopathy.

    Thema: Medizin
Notizen: Summary The occurrence and topographic analysis of granulovacuolar degeneration (GVD) in the hippocampal cortex of mentally normal controls (75 cases) and patients with Alzheimer's dementia (AD; 17 cases which included Alzheimer's disease and senile dementia of Alzheimer type), multi-infarct dementia (MID; 16 cases), Pick's disease (PD; 5 cases) and atypical dementia [5 cases; non-Alzheimer, non-Pick dementia with Fahr's syndrome (NANPDF)] were investigated. GVD was rarely found in control cases below the age of 60 years. In elderly normal brains, the statistically most representative ranking order of predilection for GVD (in decreasing severity) was: in the 60 s, CA1〉prosubiculum 〉CA2 (no GVD was found in the CA3 and CA4); in the 70 s, CA1〉prosubiculum 〉CA2 〉CA3〉CA4; in the 80 s, CA1〉prosubiculum 〉CA2〉CA3〉CA4; in the 90s, CA1〉prosubiculum 〉CA2〉CA3〉CA4. In the brains of demented patients, the rank order for GVD was: for AD, CA1 〉CA2〉CA3〉 prosubiculum 〉CA4; for MID, CA1 〉 prosubiculum 〉CA2〉CA3〉CA4; for PD, CA1 〉CA2〉CA3〉 prosubiculum 〉CA4; and for atypical dementia (NANPDF), CA1〉CA2〉 prosubiculum 〉CA3〉CA4. The similarity of the predilection to ranking order was noted both in normal aged subjects and in MID as well as both in AD and in PD. The qualitative investigation disclosed that the affected neurons with GVD in the cases of AD were found in all the examined areas outside the hippocampus (gyrus praecentralis, temporal and occipital cortex, globus pallidus, amygdaloid nuclei, mammillary bodies, medial thalamic nuclei, red nuclei, nuclei basalis of Meynert, dentate nuclei and inferior olivary nuclei) and, contrary to the results in AD, there were no affected neurons with GVD in the cases of MID. The occurrence and distribution of GVD in demented patients were different in these respective disorders.

Notizen: Abstract Polyglucosan bodies (PGBs) in the brain of a 12-year-old Holstein cow exhibiting no signs of neurological abormality were examined by light and electron microscopy and immunohistochemistry. PGBs were disseminated throughout the brain, especially in the pallidum, thalamus and cerebellum. Cow PGBs were found in the neuronal perikaryon and in the neuropil. These were round, slightly to severely basophilic, and were strongly positive for the periodic acid-Schiff reaction. The cow PGBs were immunoreactive for monoclonal antibodies raised against human polyglucosan. Electron microscopic analysis revealed that they were composed of branching filaments, glycogen granules and electrondense meterial. These findings indicate that cow PGBs closely resemble human or canine PGBs in Lafora's disease and PGBs found in aged animals that have been reported previously.

Notizen: Contents A method of simulating permanent magnets in analytical approach for the transfer-matrix-method is given. The method is suited for deriving analytical expressions of the magnetic fields and electromagnetic forces in electrical machines containing permanent magnets. The method was applied to the analysis of a long-stator linear synchronous motor with permanent magnet excitation. Measurements of the forces in the 150-pole Samarium-Cobalt type linear synchronous motor for electric propulsion were used to verify this permanent magnet modeling analysis. Comparison with experimentally obtained results shows a very good agreement.

Notizen: Abstract Plasma was used from rat, mouse, guinea pig, rabbit, dog, cat, sheep, goat, monkey, horse, pig, cow, fox, mink, porpoise, deer, manatee, seal, elephant, raccoon, pigeon, macaw, and humans; clotting time and gel formation activities by the compact-colony forming active substance (CCFAS) extracted from a strain ofStaphyloccoccus aureus and relative staphylococcal clumping factor reaction were determined. Experimental results showed that every plasma was clotted and gel formation of plasma was observed by the CCFAS, however, although it was shown by the other plasmas, no clumping-factor reaction was observed with plasma from guinea pig, goat, and elephant.
