    Notizen: Abstract Investigations in animals have suggested that conjugation of paracetamol may be reduced in malaria. We have measured plasma concentrations and the urinary excretion of paracetamol and its phase II metabolites in eight Thai patients during uncomplicated falciparum malaria and in convalescence, following a 1000 mg single oral dose. The apparent oral clearance (Malaria, 3.6; Convalescence, 3.9; ml·min−1·kg−1), the elimination half-life (Malaria, 3.8; Convalescence, 3.7 h) and apparent volume of distribution (Malaria, 1.2; Convalescence, 1.2; l·kg−1) of paracetamol were similar during malaria and convalescence. In addition, the urinary excretion of paracetamol and its major phase II metabolites and their formation clearances from paracetamol were not significantly different between the two study phases. These data show that clinical malaria infection has no effect on the conjugation of paracetamol in man.

    Notizen: Abstract An analysis is made of the laminar natural convection of incompressible fluids over a slender, hollow circular cylinder with the inner surface at a constant temperature ofT b . The temperature of the outer surface must be solved from the coupled conduction of the cylinder and the natural convection of the fluid over the cylinder. The objective of this paper is to investigate the effect of conduction on the heat transfer characteristics of the natural convection boundary layer of the fluid. A wall conduction parameter,p, is introduced which is a measure of the heat conductivities of the solid and the fluid and the thickness of the cylindrical shell. The governing differential equations, being non-similar, are solved by a finite-difference method. Numerical results are generated for a series of values ofp's and Prandtl numbers. The present analysis shows that the overall effect of conduction of the cylinder is to reduce the heat transfer. Any calculation of the heat transfer rate based on the assumption of a constant wall temperature overestimates this quantity. The effect increases for large values ofp.

    Notizen: Abstract The problem of natural convection flow and heat transfer induced by a vertically oriented cone with constant surface temperature is treated in this paper. The cone is assumed to have transverse wavy configurations. The resulting boundary layer flow is described by two coupled parabolic partial differential equations. These equations are solved numerically using the Keller-box method for a sinusoidal wavy cone. The effect of sinusoidal waves on the local Nusselt number is determined and presented on graphs. The local Nusselt number is found to be lower than that of the corresponding flat cone.

    Notizen: Summary A human granulocyte colony-stimulating factor (hG-CSF) gene was synthesized and inserted into a trp expression vector for over-expression in E. coli. A strong expression vector was constructed, and a simple purification procedure including in vitro refolding was established. The final productivity of hG-CSF was 500~600mg per l culture, and the purified hG-CSF showed the proliferation of neutrophils in vivo assays.

    Notizen: Prolonged incubation of quiescent 3T3, 3T6, and A431 cells with the P2Y purinoceptor agonists ATP, ADP, or AMPPNP reduced the mitogenic responses of target cells to a further challenge by these agonists, as measured by [3H]thymidine incorporation. The mitogenic desensitization was agonist-specific, for no effect was seen on DNA synthesis stimulated by epidermal growth factor, insulin, bombesin, 12-0-tetradecanoyl-phorbol-12 acetate (TPA), or adenosine. The desensitization was completely reversible, since after a 24 hr incubation in the absence of ATP, the cells responded fully to the mitogenic action of ATP. The presence of a low level of cycloheximide blocked recovery, suggesting that down-regulation of the P2Y receptor may have occurred during desensitization. In Swiss 3T3 cells, stimulation of DNA synthesis occurs predominantly by activation of arachidonic acid release, followed by its oxidation to prostaglandin E2 and stimulation of adenylyl cyclase. Interestingly, prolonged preincubation with ATP produced a similar degree of desensitization of DNA synthesis and of ATP-dependent arachidonic acid release and cAMP accumulation. Furthermore, this was true for both wild type cells and mutants with a defective cAMP-dependent protein kinase (PKA). We conclude that homologous desensitization is likely due to uncoupling of the P2Y purinoceptor from phospholipase A2, and this process does not require activation of protein kinase A. © 1995 Wiley-Liss Inc.
