    Notes: The ability to withstand complete submergence was studied in R2 seedlings raised by self-pollination from 158 R1 plants regenerated from callus of mature rice seeds (cultivars FR13A and Calrose). Compared to parental controls, significant improvement in submergence tolerance as assessed by percentage survival, number of new leaves and chlorophyll content of the third leaf, was found in 5 of the 115 cv. FR13A somaclones and 3 of the 43 cv. Calrose somaclones tested using an aquarium system in a controlled-temperature glasshouse. With some exceptions these improvements were also observed in the R3 generation when tested under field conditions in Thailand. Variation in other agronomically important characters was observed in the R2 plants grown in a temperature glasshouse under 2 contrasting environments.

    Notes: Abstract. Hermansky-Pudlak syndrome (HPS) is one of the few genetic disorders associated with severe pulmonary fibrosis. Fifty percent of affected patients die as a result of respiratory insufficiency. Fibrosis is thought to be caused by the accumulation of ceroid, an insoluble fluorescent lipoprotein, both extracellularly and in the lysosomes of alveolar macrophages. In addition to pulmonary fibrosis, HPS is characterized by oculocutaneous albinism and a reduction in the number of platelet dense bodies. CD63 is a protein that was described originally in platelet lysosomes. It localizes to the membranes of melanosomes and platelet dense bodies. CD63 is decreased dramatically in the lysosomes and dense bodies of patients with HPS. We theorized that CD63, a membrane protein common to lysosomes, melanosomes, and platelet dense bodies, may play a role in HPS. We sought to characterize the gene coding for this protein in HPS lymphoid cell lines. The coding region for CD63 was sequenced in control and HPS cell lines. Messenger RNA from HPS and normal cell lines was examined by Northern analysis. Genomic DNA from the same cell lines was examined by Southern analysis and polymerase chain reaction (PCR). CD63 protein in lymphoid cell lines and peripheral blood monocytes was compared by Western analysis. We found no mutations in the coding region of CD63 in an HPS cell line. We also found no diminution in the quantity of CD63 RNA by Northern analysis and no gross defects in the structural gene by PCR and Southern analysis, suggesting that the CD63 structural gene, promoter, and untranslated regions were normal. Western analysis showed that the 43-kDa protein was present in control and HPS lymphoid cell lines and peripheral blood monocytes in equivalent amounts. Although CD63 is an attractive candidate for the primary defect of HPS, the disease is probably not caused by a mutation in the CD63 gene.

    Notes: Summary The effects of creep at constant conditions of moisture content and the effects of deformations occurring during cycling of moisture content have been studied in loaded beams of particleboard and hardboard. The variations in stiffness in loaded and unloaded beams of particleboard during moisture cycling were also determined. Contrary to the findings of some workers but in agreement with the findings of another, the increases in the deflections of loaded beams of particleboard during cycling of moisture content were due mainly to desorption of moisture. Qualitatively similar behaviour was found for particleboard, hardboard and wood but quantitatively the effects were greatest in particleboard and hardboard. It appears that the basic mechanism of the transient effects in bending, arising with the simultaneous action of load and change in moisture content, may be similar in wood, particleboard and hardboard.

    Description / Table of Contents: Zusammenfassung Nach einer Begründung der Wichtigkeit der Rheologieforschung werden die hier verfolgten Einzelfragen kurz erläutert. Durch rheologische Untersuchungen kann zwar das voraussichtliche Verhalten des Holzes meist mit genügender Genauigkeit vorausgesagt werden, aber die Kenntnisse auf diesem Gebiet sind noch lückenhaft. Zu den wichtigsten Zusammenhängen gehört die Kenntnis der Wechselwirkungen zwischen Belastung einerseits und veränderlicher Holzfeuchte andererseits. Engere internationale Zusammenarbeit könnte hier, wie auch in anderen Fragen sehr nutzbringend sein.
    Notes: Summary The reasons for the interest in the rheology of wood and the favoured lines of research are stated. The response of wood can often be predicted adequately on the basis of previous studies but notable gaps remain in our knowledge. Future research should avoid going over well trodden ground and should concentrate on filling some of the gaps listed. The factor of greatest importance is the interaction of load with change in moisture content. International cooperation in rheological research on wood would be of great benefit.

    Notes: Summary Little is known about the effects of exercise training on neuromuscular junction morphology in skeletal muscle. The objectives of this investigation were: 1) to determine if exercise training would elicit changes in neuromuscular junction morphology, 2) to determine if exercise training of different intensities would evoke specific changes in neuromuscular junction morphology, and 3) to determine whether changes in neuromuscular junction structure occur independently of changes in muscle fibre type and size. Twenty-four age and size matched male Sprague-Dawley rats were randomly assigned to three groups: high-intensity trained (HIT), low-intensity trained (LIT), or untrained. Neuromuscular junction morphology of the soleus muscle was determined via immunofluorescent staining. Presynaptic acetylcholine vesicles were visualized with SV-2 antibody in conjunction with fluorescein isothiocyanate labelled secondary antibody. Postsynaptic acetylcholine receptors were identified with rhodamine labelled α-bungarotoxin. Laser scanning microscopy was used to produce images of synapses, which were used to quantitate the following: total area of SV-2 and α-bungarotoxin staining, density of acetylcholine vesicles and receptors, structural complexity, and synaptic coupling. To visualize nerve terminal branching, a smaller number of neuromuscular junctions were stained with C-2 antibody, which reacts with a neurofilament epitope, in conjunction with fluorescein isothiocyanate labelled secondary antibody. Total length of branching, number of branches, average length of branches, and ratio of secondary to primary branches per neuromuscular junction were determined. Citrate synthase activity, fibre type composition and fibre cross-sectional areas of the soleus muscle were assessed to determine the presence of a training effect in that muscle. Results indicate that training did induce hypertrophy of the neuromuscular junction that was independent of muscle hypertorphy. Although the HIT and LIT groups exhibited similar hypertrophic responses of the neuromuscular junction, the HIT group displayed more dispersed synapses than the LIT group. Neither exercise training program, however, resulted in altered densities of acetylcholine vesicles or receptors, nor did training significantly change synaptic coupling. Nerve terminal branching was also affected by exercise training. Neuromuscular junctions from the HIT group demonstrated a greater total length of branching, average length per branch, and number of finer, or secondary, branches than those of the LIT group.

    Notes: Quinacrine, an acridine derivative which competitively binds to ATP binding sites, has previously been shown to cause the reorganization of metaphase spindle microtubules (MTs) due to changes in interactions of non-kinetochore microtubules (nkMTs) of opposite polarity (Armstrong and Snyder: Cell Motil. Cytoskeleton 7:10-19, 1987). In the study presented here, mitotic PtK1 cells were treated in early anaphase with concentrations of quinacrine ranging from 2 to 12 μM to determine energy requirements for chromosome motion. The rate and extent of chromosome-to-pole movements (anaphase A) were not affected by these quinacrine treatments. The extent of anaphase B (kinetochore-kinetochore separation) was reduced with increasing concentrations of quinacrine. Five micromolar quinacrine reduced the extent of kinetochore-kinetochore separation by 20%, and addition of 12 μM quinacrine reduced the kinetochore-kinetochore separation by 40%. To determine the role of nkMTs in anaphase spindle elongationquinacrine-treated metaphase cells were treated with hyperosmotic sucrose concentrations, and spindle elongation was measured (Snyder et al.: Eur J. Cell Biol. 39:373-379, 1985). Metaphase cells treated with 2-10 μM concentrations of quinacrine for 2-5 min reduced spindle lengths by 10-50% prior to 0.5 M sucrose treatment for 5 min. This treatment showed a significant reduction in the ability of sucrose to induce spindle elongation in cells pretreated with quinacrine. As spindle length and birefringence was reduced by quinacrine treatment, sucrose-induced elongation was concomitantly diminished. These data suggest that quinacrine-sensitive linkages are necessary for anaphase B motions. Reduction in these linkages and/or MT length in the nkMT continuum may reduce the ability of the nkMTs to hold compression at metaphase. This form of energy is thought to drive a significant proportion of normal anaphase B in PtK1 cells and sucrose-induced metaphase spindle elongation.
