    Notes: Abstract The kinetics of penetration, activation and detoxification of benzo(a)pyrene were determined by near U.V. microspectrofluorimetric measurements on single living cells. This technique allows one to monitor the different intracellular fluorescent species present in a subcellular microvolume by using spectral decomposition of the fluorescence data. The T47-D cell line was chosen for its high capability of metabolization. The penetration involves a simple diffusion transfer through the cytoplasmic membrane of the cell, with a half-time of ∼ 2 min. The metabolization process gives rise, with more than a one hour delay after intracellular incorporation of the hydrocarbon, to a rapid conversion of B(a)P into unconjugated metabolites, leading to a transient accumulation of the 3OH-B(a)P metabolite in the cell. This feature may be related to the enhancement of cytochrome P1450 activity, induced by the B(a)P itself. The ability of the cell to increase its Cyt-P1450 level, after exposure to B(a)P, gives indirect evidence for the presence of the Ah gene complex in the T47-D cell line.

    Notes: Abstract · Background: Photodynamic therapy (PDT) is a new treatment modality which relies on a nonthermal light exposure of choroidal neovascularization (CNV) which has accumulated a photosensitizer, to produce a photochemical thrombosis. The aim of the present study was to develop a more quantitative outcome parameter that can be used to optimize a number of treatment parameters which in PDT are larger than many other modalities. · Methods: Nine patients underwent up to four standardized fluorescein angiography (FA) examinations on a Zeiss 30° retinograph using Kodak Trimax film. The 35-mm pictures were digitized. The CNV size was evaluated using the NIH Image 1.60 software, comparing the CNV surface to the disk area. The fluorescence intensity was measured comparing the grayscale fluorescence levels of the CNV classic pattern with the mean grayscale level of control areas. A combination of the CNV size parameter with the fluorescence contrast parameter was called “integrated contrast” (IC). The IC amplitude was then used to describe and compare the fluorescence before and after PDT. · Results: FAs on three normal patients defined the normal range of the IC. IC measurements on two patients with a classic CNV who underwent FA examinations respectively 1 and 3 months apart demonstrated the robustness and the reproducibility of the method. IC measurements on three patients who underwent PDT with three different treatment parameters demonstrated the feasibility of using this IC measurement as a PDT outcome criteria. · Conclusion: We have developed a method to transform a qualitative clinical evaluation of CNV leakage into a more quantitative grading.

    Notes: Positive and negative plasma desorption (PD) mass spectra of the di-deoxyribonucleoside monophosphate d(TpT) and of its biologically relevant ultraviolet-induced intramolecular photodimers are examined and discussed. The photodimers which were analysed by PD mass spectrometry include the cis-syn and trans-syn cyclobutyl isomers d(T[p]T), the pyrimidine-pyrimidone photoadduct (6-4)d(TpT) and its Dewar valence isomer. Molecular ions, quasi-molecular ions and several fragment ions are observed in all cases. It is shown that, despite the absence of mass differences between these dinucleoside monophosphates, the fragmentation pattern differs significantly between the two main classes: cyclobutane dimers and (6-4) adducts. PD mass spectrometry can therefore be envisaged for characterizing their formation within short DNA fragments.
