    Description / Table of Contents: Résumé Le système décrit ici permet d'étudier avec précision le vol des insectes. Il comporte principalement une série de manèges. La très faible inertie de leur partie mobile facilite l'envol et l'arrêt du vol de l'insecte permettant ainsi une très bonne expression du comportement de vol. Le rayon, rigide et léger, auquel est suspendu l'insecte est fixé à un axe vertical mobile entre deux fins roulements coniques à billes. La partie mobile comporte, en outre, un disque percé à sa périphérie de trous équidistants. Les trous, défilant entre un émetteur et un capteur d'infra rouge, envoient des signaux à un appareillage électronique. Ces signaux, convenablement traités, parviennent à un visualisateur optoélectronique consultable à tout instant sur commande manuelle. Il renseigne à la fois sur le temps écoulé depuis le début de l'expérience, le numéro de l'insecte concerné et le nombre de tours de manège effectués. Il explore successivement les données relatives à l'ensemble des manèges. Aux intervalles choisis, une imprimante imprime automatiquement ces données. En complément, un enregistreur graphique indique sous forme de courbes la répartition temporelle du vol de chaque insecte et l'évolution de sa vitesse au cours du test.
    Notes: Abstract The use of the system described here makes it possible to measure the flight activity of insects. The basic apparatus is a roundabout actuated by the insect in flight. Twelve roundabouts can be used simultaneously. Each roundabout consists of a vertical frame carrying a rotatable unit between two small conical ball races. The extremely low inertia of this unit part facilitates starting and stopping by the flier. This unit carries a horizontal disc pierced with holes on its circumference, which turns between an infrared emitter and a receptor. Each receptor receives a signal each time a hole passes between it and the emitter. The signals, appropriately treated, are fed into three recorder units. An opto-electronic visualizer provides information on the time elapsed since the beginning of the experiment, the code number of the insect concerned and the number of revolutions of the roundabout. At regular, chosen intervals, a printer automatically prints out this information. A graphical recorder shows the flight activity pattern of each insect during the test and the progress of its flight speed.

    Notes: Abstract  The human Lutheran (Lu) blood group antigens are carried by two glycoproteins (gps) that belong to the immunoglobulin (Ig) superfamily. These gps represent adhesion molecules that function as the unique erythroid receptors for laminin. We report here the cloning and functional expression of the orthologous mouse Lu mRNA as well as the genomic organization of the mouse Lu gene. The deduced human and mouse Lu gps share 72.5% identity and similar organization of the Ig-like domains. As in the human, the mouse Lu gene is organized in 15 exons. The proximal promoter showed consensus CACC-binding sites whereas the distal promoter exhibits a GATA-1-binding site and multiple E boxes. Like the human gene, the mouse Lu gene is also widely expressed among tissues but is transcribed as a unique 2.4-kb mRNA species. Expression of the mouse Lu mRNA is upregulated upon dimethyl sulfoxide-induced erythroid differentiation of murine erythroleukemia cells (MEL). During mouse embryonic development, the Lu transcript is detected as early as day 7 of gestation. Analysis of transfected human erythroleukemia K562 cells indicated that the adhesive properties of the Lu gps to laminin are conserved between human and mouse.

    Notes: Summary A complementary cDNA clone (900 bp) representing the 3′ untranslated region and almost the entire coding sequence of the human erythrocyte membrane glycophorin C has been used to determine the chromosomal location of the blood group Gerbich locus by in situ hybridization. The results indicate that this locus is assigned to the region q14–q21 of chromosome 2.

    Notes: Abstract Rh-related transcripts present in bone marrow samples from several species of nonhuman primates (chimpanzee, gorilla, gibbon, crab-eating macaque) have been amplified by RT-polymerase chain reaction using primers deduced from the sequence of human RH genes. Nucleotide sequence analysis of the nonhuman transcripts revealed a high degree of similarity to human blood group Rh sequences, suggesting a great conservation of the RH genes throughout evolution. Full-length transcripts, potentially encoding 417 amino acid long proteins homologous to Rh polypeptides, were characterized, as well as mRNA isoforms which harbored nucleotide deletions or insertions and potentially encode truncated proteins. Proteins of 30–40 000 M r, immunologically related to human Rh proteins, were detected by western blot analysis with antipeptide antibodies, indicating that Rh-like transcripts are translated into membrane proteins. Comparison of human and nonhuman protein sequences was pivotal in clarifying the molecular basis of the blood group C/c polymorphism, showing that only the Pro103Ser substitution was correlated with C/c polymorphism. In addition, it was shown that a proline residue at position 102 was critical in the expression of C and c epitopes, most likely by providing an appropriate conformation of Rh polypeptides. From these data a phylogenetic reconstruction of the RH locus evolution has been calculated from which an unrooted phylogenetic tree could be proposed, indicating that African ape Rh-like genes would be closer to the human RhD gene than to the human RhCE gene.

    Notes: Summary A cDNA clone, RhIXb (1384 bp), encoding the entire protein sequence of a human blood group Rh polypeptide has been used to map the Rh locus, by in situ hybridization, to the region p34.3–p36.1 of chromosome 1. Two other unrelated cDNA clones, pUCA2 (750bp) and pUCIII (1600 bp), isolated during the cloning procedure of the Rh cDNA were investigated simultaneously, and assigned to chromosome 3p21.1–3p22 (clone pUCA2) and to chromosome 22q12.1–22q13.1 (clone pUCIII).

    Notes: Abstract As the chimpanzee R-C-E-F blood group system appears to be the chimpanzee counterpart of the human Rhesus (RH) system, we have tried to determine whether chimpanzee Rh-like genes encode R-C-E-F-related proteins. Chimpanzee genomic DNA, digested by any of eight endonucleases and hybridized with three Rh exon-specific probes, exhibits a high degree of polymorphism. Analysis of DNA from unrelated individuals of different R-C-E-F types revealed that the presence of some restriction fragments is correlated with particular R-C-E-F types. The cosegregation of these fragments with R-C-E-F haplotypes was confirmed by family studies. Oligonucleotides complementary to regions flanking human exons were used as PCR primers on chimpanzee DNA; the resulting amplified fragments were identical in size to their human counterparts. Moreover, the nucleotide sequences of the fragments present a high degree of similarity to the corresponding human regions.
