    Notes: Zusammenfassung 1. Bei 2 Patientinnen mit chronischer Polyarthritis und 5 gesunden Vergleichspersonen wurde das Verhalten des Mineralstoffwechsels und der Corticoide unter Pyrifer, Insulin, ACTH und Irgapyrin untersucht. 2. ACTH, Insulin und Irgapyrin führten im Verlauf der Therapie zu einer deutlich verminderten Kochsalzausscheidung, die sich während der Weiterbehandlung wieder normalisierte oder von einer Mehrausscheidung gefolgt war. Unter Pyrifer ergab sich vermehrte Salzausschwemmung am Injektionstag mit nachfolgender NaCl-Retention. 3. Die Maximalwerte der Corticoidausscheidung unter Pyrifer, Insulin und ACTH differierten nur relativ wenig voneinander. Unter ACTH war jedoch die Ausscheidung während der ganzen Dauer der Therapie erhöht, unter Pyrifer meist am Injektionstag selbst, unter Insulin erfolgte der Anstieg nur zögernd im Verlauf der Therapie. 4. Die Eosinophilen verschwanden unter ACTH fast völlig aus dem peripheren Blutbild, unter Insulin und Pyrifer kam es zu einem regelmäßigen starken, aber nur kurzdauerndem Abfall. Unter Irgapyrin war der Abfall nur gering. 5. Ein Vergleich zwischen der Höhe des Corticoidanstiegs und dem therapeutischen Erfolg bei Polyarthritikern gab keine eindeutigen Zusammenhänge. Bei Asthmatikern blieb bei einem Teil der untersuchten Fälle der Steroidanstieg nach Pyrifer aus oder war relativ niedrig. 6. Die Möglichkeit eines Zusammenhangs zwischen Nebennierenreaktion und klinischer Wirkung wird diskutiert.

Es wird die Acetylcholinsynthese aus Acetat + Cholin, Citrat + Cholin und Acetat + Citrat + Cholin, sowie Cholincitrat (in Form von Neurotropan) unter anaeroben Bedingungen untersucht. 2. Äquimolekulare Mengen von Citrat oder von Acetat fördern die Acetylierung von Cholin in gleichem Ausmaße. Ein Zusatz von äquimolekularen Mengen von Citrat und Acetat im Verhältnis 1:1 bewirkt eine doppelte Acetylcholinsynthese, wie dies der Summe gebildeten Acetylcholins durch Acetat bzw. Citrat entspricht. 3. Cholincitrat (Neurotropan) citronensaures Salz des Cholins in ionogener Bindung acetyliert in gleichem Umfange Cholin wie eine Mischung der Einzelkomponenten Na-Citrat und Cholinchlorid. 4. Unsere Versuche bestätigen die Befunde vonLipton undBarron sowie vonMartius über den Mechanismus der anaeroben Citronensäurespaltung und die anaerobe Acetylcholinbildung. 5. Die besondere Wirksamkeit von Citrat auf die Acetylcholinsynthese ist nicht allein durch die bevorzugte Bereitstellung von Acetyl zur Transacetylierung auf das Cholin begründet, sondern es scheint, daß ein Teil der primär ionogen an das Cholin gebundenen Citronensäure auf Calciumionen entionisierend wirkt. Damit scheint das erst durch Ca-aktivierte Ferment der Acetylcholinspaltung, die Cholinesterase, gehemmt zu werden.

    Description / Table of Contents: Zusammenfassung Eine Inkubation nativer oder fixierter Kryostatschnitte in einem TNBT-haltigen Inkubationsmedium und eine anschließende Reduktion des TNBT zum Formazan führt zu einer nichtenzymatischen Anfärbung der Gewebsstrukturen. Diese Beobachtung wird licht- und elektronenmikroskopisch im Hinblick auf eine intrazelluläre Lokalisation des TNBT-Formazans näher analysiert. Elektronendichtes TNBT-Formazan konnte in der Zelle in unmittelbarer Nachbarschaft von Mitochondrienaußenmembranen, Membranen des endoplasmatischen Retikulums und in organellenfreien Bezirken nachgewiesen werden. Eine vorherige Lipidextraktion bewirkt eine Abschwächung der Reaktion. Es wird daraus der Schluß gezogen, daß TNBT unspezifisch an Lipoproteinstrukturen der Zelle adsorbiert wird und daß die beobachtete Lage des TNBT-Formazans durch Diffusionsphänomene beeinflußt wird. Möglicherweise kann das TNBT durch frei diffusible reduzierende Gewebskomponenten reduziert werden. Bei Verwendung von TNBT als Reduktionsindikator zum licht- und submikroskopischen Nachweis oxydativer Enzyme ist mit einer unspezifischen TNBT-Formazan-Lokalisation in Gewebsstrukturen zu rechnen. Die endgültige Bestätigung dieser Auffassung erfordert weitere Modellversuche.
    Notes: Summary Incubating native or fixed cryostat sections in a TNBT-containing medium and reducing TNBT to formazan, produces a non-enzymatic staining of tissue structures. This phenomenon is investigated light and electron microscopically; the emphasis is put on the intracellular localization of TNBT-formazan. In the cell electron-dense TNBT-formazan is found in the immediate vicinity of the external membranes of mitochondria, membranes of the endoplasmic reticulum, and in regions, which are free from organelles. The reaction becomes weaker if it is preceded by an extraction of lipids. It is therefore concluded that TNBT is absorbed non-specifically onto the lipoproteins of the cell and that the localization of TNBT-formazan is influenced by diffusion phenomenons. It seems possible that TNBT is reduced by free diffusible reducing tissue components. If TNBT is used as an indicator for reduction in light or submicroscopical investigations of oxidative enzymes a non-specific TNBT-formazan localization in tissue structures might be encountered. However, more work along these lines is needed to finally confirm this theory.

    Notes: Abstract More than 80 years ago, Alzheimer described changes in the brains of patients who had suffered hepatic failure. Astrocytes are primarily affected; their nuclei become swollen, their intermediate filament protein composition is altered and their cytoplasm becomes vacuolated. Cells with these features are called Alzheimer type II astrocytes and these changes have been attributed to the toxic effects of elevated ammonia levels. The present study investigates whether the dominant glia of another part of the central nervous system, the Müller cells of the retina, undergo similar changes. Retinae of patients who had died with symptoms of hepatic failure were processed for histology, histochemistry, and immunocytochemistry. Cell nuclei were measured from brain astrocytes (insula cortex), Müller cells, and retinal bipolar neurons. Hepatic failure resulted in the enlargement of nuclei in astrocytes and Müller cells, and the enhanced expression in Müller cells of glial fibrillary acidic protein, cathepsin D, and the β-subunit of prolyl 4-hydroxylase (glial-p55). In some retinae, signs of gliosis were also observed. We conclude that increased levels of serum ammonia resulting from hepatic insufficiency cause changes in Müller cells that are similar to those seen in brain astrocytes. We term this condition hepatic retinopathy.
